Epigenetic regulation: methylation of histone and non-histone proteins

Abstract: Histone methylation is believed to play important roles in epigenetic memory in various biological processes. However, questions like whether the methylation marks themselves are faithfully transmitted into daughter cells and through what mechanisms are currently under active investigation. Previously, methylation was considered to be irreversible, but the recent discovery of histone lysine demethylases revealed a dynamic nature of histone methylation regulation on four of the main sites of methylation on histone H3 and H4 tails (H3K4, H3K9, H3K27 and H3K36). Even so, it is still unclear whether demethylases specific for the remaining two sites, H3K79 and H4K20, exist. Furthermore, besides histone proteins, the lysine methylation and demethylation also occur on non-histone proteins, which are probably subjected to similar regulation as histones. This review discusses recent progresses in protein lysine methylation regulation focusing on the above topics, while referring readers to a number of recent reviews for the biochemistry and biology of these enzymes

Abstract: Getting embryogenic callus is the first step for banana transformation. Enhancing embryonic-callus-rate is important for banana molecular breeding. In this paper, WIND1, an AP2/ERF transcription factor was added into the embryonic-callus-inducing medium with poly-Arg and nuclear localization signal (NLS). The embryogenic-callus-induced rate of the immature male florescence cultured on the media containing the recombinant protein Arg9-NLS-WIND1 was significantly higher than that of the immature male florescence cultured on the control media. Medium containing 0.01% of the protein Arg9-NLS-WIND1 had the best effect for inducing the embryogenesis of the immature male inflorescence. On the medium containing 0.01% of the protein Arg9-NLS-WIND1, 11.5% of the immature male flowers can develop embryonic callus. On the medium containing Arg9-NLS-WIND1, the protein might be transferred into cytoplasm of the explants by endocytosis due to the interaction between the poly-Arg transduction domain and the plasma membrane. After it entered the nucleus guided by NLS and bind with the target DNA domain, the somatic cell embryogenesis reactions were initiated and the embryonic callus formed.
Mercury Distribution in Tobacco (Nicotiana tabacum) Cell  [PDF]

Abstract: Problem: Removing mercury from polluted soil using transgenic plants is an ideal method. However, where mercury was stored in plant cell is not clear until now. Methods: Differential centrifugation and laser scanning confocal microscopy were used in this study. Results and findings: Results showed that after mercury was absorbed by tobacco plants, most of the mercury accumulated in roots. Mercury content in root was significantly higher than that in shoot. In the seedlings cultured in MS liquid medium containing 5 μmol/L mercuric chloride, the mercury content in cell wall was 6.6 μg/g. The mercury content in cell membrane fraction was 2.1 μg/g. The mercury content in supernatant fraction was 35.1 μg/g. Most of the mercury accumulated in roots located in liquid fraction, about 15% of the mercury was attached by cell wall. Only a small part assembled in cell membrane. Most of the mercury in liquid fraction located in vacuole. This suggested that after the mercury was accumulated in plant root, most of the mercury was transferred into vacuole. There was no important cellular organ in vacuole. The toxicity of mercury in vacuole will be much lower than that in cell membrane or cellular organs. Recommendation

Abstract: This paper presents a nonmonotonic logic-based approach to the problem of model management in uncertain environments. The proposed algorithm for uncertainty analysis provides a mechanism for uncertainty representation, propagation, evidence combination and problem solving under uncertainties. Furthermore, uncertainties about model structures, i.e., the probabilities of possible model structures, are represented by probability propositions supported by sets of ATMS's assumptions about uncertain situations. Hence, the management of uncertain causal link between models is realized in the processes of truth maintenance, belief assignment and adjustment about assumptions' environment, wherein, conflict situations presented in problem solving processes and related ATMS justifications about the results of problem solving are taken as new evidences for truth maintenance and the adjustment of degrees of belief.
An Improved Wavelet Filtering Algorithm and Its FPGA Implementation

Abstract: The de-noising of sensor data has become an important to research. Since the traditional de-noising method can’t achieve successful de-noising effect and the software-only method never meets a high real time capability. In this paper, we illustrate a novel threshold function based on the wavelet hard and soft threshold function. It is unlike ordinary function, which has overcome the defect such as the discontinuity of hard threshold function and an invariable dispersion between the estimated wavelet coefficients and the decomposed coefficients of soft threshold function. Moreover, we consider the hardware implementation of wavelet threshold filter on FPGA which adopt the pleated sheet structure of multiplier and fit to frame data. A detailed description of the simulation and implementation is given. Finally, the experiment result on-board is shown that our hardware implementation can meet the requirement of real-time signal processing.
A transient thermal cloak experimentally realized through a rescaled diffusion equation with anisotropic thermal diffusivity

Abstract: Transformation optics originating from the invariance of Maxwell's equations under the coordinate mapping has enabled the design and demonstration of many fascinating electromagnetic devices that were unconceivable or deemed impossible before [1-11], and has greatly contributed to the advancement of modern electromagnetism and related researches assisted with the development of metamaterials [12-15]. This technique has been extended to apply to other partial differential equations governing different waves [16-23] or flux [24-28], and has produced various novel functional devices such as acoustic cloaks [20-23] and Schrodinger's 'hat' [19]. In the present work we applied the coordinate transformation to the time-dependent heat diffusion equation [24-28] and achieved the manipulation of the heat flux by predefined diffusion paths. In the experiment we demonstrated a transient thermal cloaking device engineered with thermal metamaterials and successfully hid a centimeter sized strong 'scatter' (thermal disturber), i.e., a vacuum cavity. To facilitate reliable fabrication we adopted the rescaled thermal diffusion equation for various ingredient materials with nearly constant product of the density and heat capacity, and took the anisotropic thermal diffusivities as the key parameters for the design. Our results unambiguously show the practical possibility to implement the complex transformed thermal media with high accuracy and acquire some unprecedented thermodynamic functions, which we believe will help to broaden the current research and pave a new way to manipulate heat for novel device applications.
New Polyhydroxylated Furostanol Saponins with Inhibitory Action against NO Production from Tupistra chinensis Rhizomes

Abstract: Two furostanol saponins were obtained from the rhizomes of Tupistra chinensis Bak. Their structures were determined as 5β-furost-δ25(27)-en-1β,2β,3β,4β,5β,7α,22ξ,26- octaol-6-one-26-O-β-D-glucopyranoside (1) and 5β-furost-δ25(27)-en-1β,2β,3β,4β,5β,6β, 7α,22ξ,26-nonaol-26-O-β-D-glucopyranoside (2), on the basis of chemical and spectroscopic evidence. Both compounds displayed marked inhibitory action against NO production in rat abdomen macrophages induced by lipopolysaccharide (LPS) at 40 μg/mL.

Abstract: In this paper, the authors use ultrasonic data over a homogeneous surface to calculate the Kolmogorov constants of velocity, temperature, CO2, and water vapor. The authors find that the constants are all within the range determined by former experiments and that they are universally independent of the stability.
