The Evaluation of the Effects of Temozolomide on MGMT Gene Expression in MCF-7 and SKBR3 Human Breast Cancer Cell Lines  [PDF]

Abstract: Background and Aim: In this study, it was aimed to examine the cytotoxic effect of temozolomide (TMZ) treatment, on MCF-7 and SKBR3 cell lines, to study the methylation levels of MGMT gene expression and gene promoter region. Methods: The MTT test was performed to determine the effective dose of TMZ. The time-dependent cell survival test was performed after the IC50 value was found. Western blotting was performed to determine MGMT gene expression levels. High Resolution Melting (HRM) technique was used to determine the methylation levels of MGMT gene promoter region. Results: TMZ has been shown to have a high cytotoxic effect on SKBR3 cell line and low cytotoxicity on MCF-7. When MGMT expression levels before and after TMZ treatment were observed by western blotting, the gene expression levels of TMZ treatment were shown to decrease in both cell lines. It was observed that MGMT gene promoter region was hypermethylated in two cell lines, and that the application of TMZ further increased the methylation levels in the promoter region. Abstract: in this study, the cytogenetic effects of black tea and green tea were determined in cultured peripheral blood lymphocytes. results showed that black tea and green tea induced the mitotic and replication indexes and decreased micronuclei. but these data were not statistically significant for green tea. the effects of black tea on the micronucleus formation and mitotic index were statistically significant. the decrease in micronucleus counts indicated that black tea and green tea had considerable anticlastogenic and antigenotoxic effects as observed in vitro in human lymphocytes. thus, it could be concluded that tea polyphenols protected the normal cells from genotoxic or carcinogenic agents, which indicated the therapeutic and antioxidative role of catechins, flavonoids or other tea compounds.

Abstract: saf is a poet statesman who lived in the sixteenth century. He started his career in the state as a scribe during the reign of Sultan Murad the third, and later continued as a combatant soldier, and eventually he advanced in the rank of grand seignior. Beside his successful service in the state, he was especially known with his ecaat-name masnavi. The theme of ecaat-name, which is one of the important examples of gazavatname, is the Ottoman’s travel to the Orient between the years 1578-1585. ecaat-name, which has a considerable place among the works containing Islamic features in the Turkish war literature, is a remarkable masterpiece since it has many historical documentary and literary significance. Abstract: Nowadays one of the most debated subjects by the social scientists is anger and its further dimension aggression; because theintensity of the aggression emotion changes from person to person. There might be lots of reasons that determine individuals’aggressive behaviors and aggression behaviors can be projected in different ways depending on age. When developmental periodsare considered the behaviors that cause aggression in childhood, adolescence, adulthood or old age periods differentiate. Theenvironment in which individuals survive affects their emotional and behavioral development especially socio economicenvironment might cause aggression tendencies. This research in which aggression behaviors of high school students anduniversity students were observed according to several variables was carried on by survey method. The scope of the research wasconstituted of 2242 students who were students in Konya high schools and students in Sel uk University in 2008-2009educational semester (1294 high school (57,7%), 948 university (42,3%)). In order to gain research data “AggressionQuestionnaire” and “Personal Information Form” were used. Research’s data determined that the aggression behaviors of highschool students and university students differentiated according to aggression behavior sub dimensions. On the other handaggression behavior differentiated according to their gender. Recommendations were discussed according to findings of theresearch. Aggression concept is a multi dimensional concept like concepts used in behavioral sciences. For that reason thisconcept has different meanings in psychiatry, neurology, social psychology and psychology’s different sub branches. Aggressionand violence can be used for each other. There are several approaches which try to explain the reasons of aggression and itssources. These approaches observe the reasons of aggression as personal factors and factors independent from persons.Theoretical analyses depending on personal reasons try to explain the reasons with biological, instinctive and psycho-social causes.Individual is aggressive because s/he was genetically coded like that or structural abnormalities (hormonal disorders or braindamage) cause aggression. Theories focused on factors independent from persons developed a discourse which depends on social and cultural variables. Communication is vital for coordination andcollaboration within a firm, and for establishing relationships with clients. It is an essential skill for a manager. Today's businessworld places greater expectations on those who seek to lead. When recruiting new employees, increasingly companies say theylook for people with razor-sharp communication skills more than analytical ability. Boundaries have fallen at the national level,and this has been mirrored by an equivalent effect at the personal level. It is not enough to say what you want people to do - theywant to know why. "Knowing why" is as important as "knowing how".Beyond the extensive research done in job performance the past few decades, researchers have done some work on theorganization communication--job satisfaction construct. The prevailing evidence indicates these factors relate positively. Onlythree studies, though, have documented correlations specifically between. Jain's (1973) early study used a questionnaire interviewformat to examine the relationship between communication effectiveness of hospital supervisors and their job performance asperceived by their subordinates. Effectiveness criteria were supervisor communication behavior, supervisor-subordinatecommunication frequency and amount, employee knowledge of policies and procedures, employee communication satisfaction,and non- supervisory formal communication channels use. Results showed positive correlations between communicationeffectiveness and job performance, communication frequency and amount and job performance, and employee communicationsatisfaction and supervisor job performance. the positive relationship between organizational communication and job satisfactionappears to be well documented, while there is less reason to say this about the relationship between organizational communicationand job performance. Abstract: In general terms, the mortgage market has two faces, the primary and the secondary markets. The primary market covers housing mortgage loans. On the other side of the mechanism, these loans are securitized and issued in financial markets with different form of securities in secondary markets. The common name of these instruments is “mortgage backed securities - MBS”. A mortgage backed security is a financial instrument issued in capital markets for investors, derived from either backed by the cash flow of the housing loan repayments (which is called pay-through) or backed by directly selling the mortgage pools to the MBS issuers (which is called passthrough). The most trading secondary mortgage market instruments and world practices are explained in this article.

Abstract: Dirofilariasis is one of the zoonotic fiilarial infections inadvertently affecting the humans. It is caused by filarial nematodes of genus Dirofilaria, which naturally infects several domestic and wild animals. Dirofilaria is transmitted to human by mosquitoes (Diptera, Culicidae) of various species, such as Culex, Anopheles, Aedes which are probably the most important vectors in Turkey. The Dirofilaria parasite which cause of Dirofilariasis disease can be diagnosed using membrane filtrationacide phosphates histochemical staining, serological methods and molecular methods. Dirofilaria is a parasite that can cause serious pathological disorders and even deaths in carnivorous animals especially dogs and humans. In recent years, people are increasingly at risk of being infected with Dirofilariasis diseases due to increased dog or cat feding in their homes. Therefore, control programmes should be implemented to prevent the spread of the disease in our country and world. This review includes human Dirofilariasis agents, clinical examination and laboratory diagnostic methods of Dirofilariasis.

Abstract: Some factors may affect prognosis and may be used to determine long term life duration after myocardial infarction. Hence, risk classification after myocardial infarction is of great importance. Coronary reperfusion following fibrinolytic therapy may be detected invasively and non-invasively in myocardial infarction with ST-segment elevation. ST-segment resolution, which is one of non-invasive reperfusion criteria, might be used to determine prognosis, since it reflects myocardial microcirculatory circulation better, and it is an easy, simple, and inexpensive parameter used in clinical practice. In the present study, we evaluated the prognostic importance of ST-segment resolution degree.

Abstract: k Deryam is one of the representatives of minstrel literature of the 20th century. Like his predescessor and contemporary minstrels, he lived as a member of ordinary people, and naturally, grew up with folk culture. The main purpose of this study is to examine the folk culture elements that take place in Minstrel Deryami’s poems. To achieve this goal books about Deryam hasbeen scanned with great care; proverbs, idioms, place names, plant names, local sayings, beliefs, occupations, folk poets and minstrels, prayers, curses, religious motives, relatiosships, musical instruments, fairy-tale motives etc. in books have been struggled to examine. The datas have been shown in tables. It has been brought out that two indispensible words “rose” and “nightingale” of both divan literature and folk literature are mostly used in Deryam ’s poems.
