CD134 as target for specific drug delivery to auto-aggressive CD4+ T cells in adjuvant arthritis

Abstract: In several autoimmune diseases, for example rheumatoid arthritis, the involvement of CD4+ T cells in disease induction has been suggested [1]. As a treatment strategy, the manipulation of CD4+ T cells by CD4-directed antibodies has therefore been studied extensively [2]. However, because anti-CD4 therapy targets the whole CD4+ population, CD4+ T cells not related to the disease or involved in disease regulation will also be affected. Ideally, only the auto-aggressive CD4+ T cells that are involved in the disease process should be targeted. Because for many human autoimmune diseases the exact antigens recognized by these cells are not known, a therapy would be favorable that specifically targets the auto-aggressive CD4+ T cells and does not depend on the definition of the crucial auto-antigen.Because auto-reactive CD4+ T cells become activated upon recognition of their cognate antigen in the periphery, they will be transiently marked by the expression of T cell activation markers. In this respect, CD134 (OX40 antigen) is an interesting candidate target molecule, because CD134 is expressed in vivo exclusively on activated CD4+ T cells (reviewed in [3]). In experimental autoimmune encephalomyelitis, a disease model for multiple sclerosis, it has been shown that CD134 is preferentially expressed on pathogenic CD4+ T cells that home to the target organ (namely the central nervous system) [4], and transiently marks the auto-aggressive T cells specific for myelin basic protein [5]. Moreover, in this T cell transfer model, depletion of CD134+ T cells with an anti-CD134 immunotoxin results in the amelioration of paralytic symptoms [6]. Interestingly, in patients with rheumatoid arthritis a high percentage of CD4+ T cells in synovial fluid express CD134 in comparison with peripheral blood T cells [6,7], suggesting that auto-aggressive CD4+ T cells may be transiently marked by surface expression of CD134 in arthritis too.Here, we investigated whether CD134 can be used as a tar

Abstract: Objectives. The main objective of this work was to establish the incidence of classic galactosaemia and primary congenital hypothyroidism in newborns in the Nkangala district of Mpumalanga. In the process a cost-effective protocol for neonatal screening of both diseases was developed. Study design and setting. Blood spot specimens were collected from a third (1 012 out of 3 297) of newborn infants in the Nkangala district of Mpumalanga province from June to November 2005. The specimens were subsequently screened for classic galactosaemia and hypothyroidism using metabolite quantification assays. Galactose-1-phosphate uridyltransferase (GALT) enzyme activity assays were also performed to confirm the reliability of the total galactose assays. The real-time polymerase chain reaction (PCR) was used to detect commonly occurring mutations in the GALT gene that cause galactosaemia. Thyroidstimulating hormone (TSH) levels were evaluated as a diagnostic metabolite for primary congenital hypothyroidism. Subjects and outcome measures. Informed consent was obtained from the babies' parents before commencement of screening. Total galactose levels above 0.9 mg/l and TSH concentrations above 25.1 mU/l were considered to indicate galactosaemia and hypothyroidism, respectively. A decrease in the total financial input on the screening protocol was evaluated for significance in cost reduction. Results. The prevalence of hypothyroidism was found to be 0.1%, while none of the newborns presented with classic galactosaemia. There was an up to 20% reduction in direct input costs of screening when our protocol was applied. Conclusion. Cost-effective newborn screening is possible when classic galactosaemia and congenital hypothyroidism are screened for simultaneously. Cumulative disease frequency plots confirm the already established fact that hypothyroidism tends to occur at higher frequencies than classic galactosaemia. Abstract: Sera were obtained from 100 patients with SLE and 100 patients with Sj?gren's syndrome. A total of 78 sera from patients with rheumatoid arthritis (RA) and 100 from normal healthy blood donors were also tested.Antibodies to Scl-70, Jo-1, and rRNP were not detected in any of the sera from the disease groups. In the normal control group, the only positive result obtained was in one sera that gave a weak positive result for anti-CENP-B.The FIDIS connective assay system simultaneously measures IgG antibodies directed against nine different specificities. The data presented indicate that this system provides an alternative strategy for the measurement of specific ANA to the use of multiple single analyte assays currently employed in many immunology laboratories.

Abstract: Reformed churches in the tradition of John Calvin and the well-known Synod of Dordt (1618-1619), have a long history of involvement of some kind in school education. This article looks into the famous – at least in some circles – church order of Dordt, and the present orders of three reformed churches: the Reformed Churches in South Africa, the Dutch Reformed Church and the Christian Reformed Church in North America. All three tend to be in the tradition of Dordt. In the Dutch society of the 17th century close ties between the Reformed Church and the government gave this church the opportunity to influence Dutch school education. Firstly the church used the angle of the all embracing kingdom of God in which church and school are different institutes in society but serve the same Lord. Secondly the Reformed Church also influenced school education from the viewpoint of the church as church by introducing its confessions as a point of departure and also it’s discipline in schools. The other three churches opted for a say in the spiritual direction in schools, but not to tie them to a specific church. They strive for Christian and not church schools. Article text in Afrikaans

Abstract: The article deals critically with current discourses on morality in African thought. These discourses reflect the ambivalence between those scholars seeking to define African morality within the parameters of a conventionalised, Western, religious episteme, and those pursuing an “Africanist” (Afrocentric) explanation which embraces an authentic mode of African knowledge construction within indigenous communities. The assumption that faith or religion is the foundation of African morality can only be partially endorsed when one grants space for hybrid moral constructions between Christianity and indigenous religion. However, African morality is not necessarily based on religion or faith, but on the beneficiary values of collective family and community well-being, without dissolving the individual’s character. In African thought, the “best” rational justification of the moral imperative is less of an issue than in current moral discourse.

Abstract: The Church Order of the Dutch Reformed Church puts the position of the Scriptures, its confessions of faith and its order for the church in a truly reformed perspective. In some instances this can be traced to the famous Synod of Dordt of 1618-1619. Supported by resolutions from the General Synod, these points of departure indicate the official stance of this church on these issues — a stance which can only be changed, be it officially, by changing the formulations in the Church Order. The Scriptures are regarded as the objective Word of God. With regard to the confessions, quia does not necessarily mean that one supports every reference or remark, but it indicates that the doctrine it confesses and formulates is according to the Scriptures. With regard to the offices, the assemblies and the relationships of the church, this Church Order differs from an attitude which has been accepted over many years by other reformed churches. Two dangers lie ahead for the Dutch Reformed Church. On the one hand, conservatism with a confessionalism that regards the confessions as canonical for all practical purposes. On the other hand, the church may endeavour to keep up with the times to such an extent that it exchanges Scriptures as a point of departure for humankind itself: It chooses to be liberal in the sense of an unrestricted freedom for humankind.
