Colombia Forestal , 2008,
Abstract: this research includes the study of aerial parts (fruits) of lulo de monte (solanum quitoense var. septentrionale).the main objective of this research was to determine the secondary metabolites and especially the presence and quantity of alkaloidal saponins, which are common in the solanaceae family. these substances are used as precursors for the manufacture of several steroid-type drugs, hormones and anti-inflammatories. the study was conducted with species of three different origins and three different maturing stages gathered in the department of cundinamarca, colombia between 2.400 and 2.600 meters above sea level during the period from march to june. sapogenins were extracted, isolated and purified through various techniques used for this purpose. the presence of saponins was proved through physical and chemical analysis, and the structural elucidation through nmr and ir spectroscopic techniques. the presence of hecogenin in green fruits from the three sources was determined. this molecular structure corresponds to a non-steroidal sapogenin reported inthis species.

BMC Infectious Diseases , 2009,
Abstract: Background Antigen specificity and IgG subclass could be significant in the natural history of Chagas' disease. The relationship between the different stages of human Chagas' disease and the profiles of total IgG and its subclasses were thus analysed here; they were directed against a crude T. cruzi extract and three recombinant antigens: the T. cruzi kinetoplastid membrane protein-11 (rKMP-11), an internal fragment of the T. cruzi HSP-70 protein192-433, and the entire Trypanosoma rangeli HSP-70 protein. Methods Seventeen Brazilian acute chagasic patients, 50 Colombian chronic chagasic patients (21 indeterminate and 29 cardiopathic patients) and 30 healthy individuals were included. Total IgG and its subtypes directed against the above-mentioned recombinant antigens were determined by ELISA tests. Results The T. cruzi KMP-11 and T. rangeli HSP-70 recombinant proteins were able to distinguish both acute from chronic chagasic patients and infected people from healthy individuals. Specific antibodies to T. cruzi crude antigen in acute patients came from IgG3 and IgG4 subclasses whereas IgG1 and IgG3 were the prevalent isotypes in indeterminate and chronic chagasic patients. By contrast, the specific prominent antibodies in all disease stages against T. cruzi KMP-11 and T. rangeli HSP-70 recombinant antigens were the IgG1 subclass. Conclusion T. cruzi KMP-11 and the T. rangeli HSP-70 recombinant proteins may be explored together in the immunodiagnosis of Chagas' disease. Polarising the IgG1 subclass of the IgG response to T. cruzi KMP-11 and T. rangeli HSP-70 recombinant proteins could have important biological effects, taking into account that this is a complement fixing antibody.

Abstract: blood from the umbilical cord is an alternative for the isolation and culture of cd34+ hematopoietic stem cells useful for bone marrow replacement therapy in the treatment of neoplastic and nonneoplastic pathologies. the expression of proteins related to the differentiation of cd34+ hematopoietic stem cells under culture was evaluated in this paper. the blood samples from the umbilical cord were collected in bags with cpda (baxter?) and transported to the hematology laboratory of the faculty of sciences of the pontificia universidad javeriana. the isolation of the cd34+ hematopoietic stem cells was carried out by using the miltenyi biotec microbeads? system starting from mononuclear cells obtained by density gradient with ficoll-hypaque amershan biosciences?. the cd34+ cells were cultured in stem pro? medium supplemented with il3, scf and gm-csf between 5 and 11 days at 37o c and co2 5 %. cellular viability was defined. the morphological changes were determined by inverted microscopy, whereas the expression of the cd34, cd33 and cd38 proteins, and of bci-2 and cd95 was identified by flow citometry. 6 cultures (n=6) enriched with cd34+ hematopoietic cells were established, with a viability average of 84.25 % and a culture time from 5 to 11 days. in the morphological evaluation, it was observed the appearance of cytoplasmatic prolongations or magnupodia-like pseudopodes until the seventh day of culture. the coexpression of the cd34 y cd33 antigens with an increase of cd95 protein from the seventh day on was observed. the cd34+ hematopoietic stem cells taken from the blood of the umbilical cord presented cytoplasmatic correlations that were possibly associated with cell to cell interactions. in our culture system, the obtained hematopoietic stem cells initiated processes of myeloid differentiation that were indirectly related to the expression of bci-2 and cd95.

Abstract: phenylalanine ammonia lyase (pal) activity induction was evaluated in lulo fruits to determine the role of this enzyme in biochemical responses towards the pathogen colletotrichum acutatum. we studied the experimental conditions to obtain the enzyme, using lulo peel, and found that the best conditions for extraction were buffer of boric acid-sodium borate 0.1m ph 8.8, 1% sds, 3% pvpp and, for measuring the enzymatic activity: l- phenylalanine 5mm, ph 8.0, 20°c , 30 μl of extract and incubation during 45 min. then, we performed an in vivo assay using lulo fruits in three maturity stages, which were inoculated either with the fungus or sterile water. enzymatic induction was studied at five post-inoculation times, and it was found that there is a differential response as a consequence of the presence of the pathogen and the maturity stage. in semi-mature lulos, a bigger increase in pal was obtained among 12-48 hpi which is related to a higher content of phenolic compounds and to the latest development in the characteristic symptoms of anthracnose. these results allow postulating, in a preliminary way, a possible positive relationship between inductions of pal, phenolics and responses of tolerance to c. acutatum. for green and mature lulos we observed as well, an increase of pal activity but only at 12 and 24 hpi which showed relation with an increase in the total phenolic content. however, for these stages the phenolics were significantly lower at higher times.
