Genetic diversity between and within populations of Handroanthus heptaphyllus (Vell.) Mattos using microsatellite markers

Abstract: Handroanthus heptaphyllus (Vell.) Mattos, popularly known as ipê-roxo, is a species of the family Bignoneaceae much appreciated for its beauty, excellent quality wood which is used for making medicinal products and also in reforestation programs of degraded areas, as well as landscaping and restoration. The aim of this study was to investigate the genetic diversity between and within populations of H. heptaphyllus using microsatellite markers. The 192 seedlings were produced from seeds collected on 30 trees into the two populations of natural forest fragments in Botucatu region, S o Paulo, Brazil. Eight microsatellite loci were analyzed, with allelic polymorphism varying from six alleles for locus TAU22 to 14 alleles for loci TAU12, TAU30, and TAU31, with an expected mean number of alleles per locus (Ae) of 4.9. The mean expected heterozygosity (He) for the two populations was 0.785, the mean observed heterozygosity (Ho) was 0.609, and the fixation index (F) was low between populations, with a mean of 0.222. The gene differentiation between the two populations (Gst) was 0.100. We concluded that the higher genetic diversity is within populations; therefore, as far as germplasm collection programs in Botucatu region are concerned, it is recommended that a larger sampling of individuals should be considered within populations, thereby providing good genetic representativeness. The populations have enough genetic diversity to support genetic improvement and germplasm preservation programs.

Abstract: seven baby corn families were crossed in a complete diallel scheme. the single-cross hybrids obtained were assessed at s?o manuel experimental station of crop sciences school (faculdade de ciências agron?micas) of s?o paulo state university -botucatu (unesp). the experiment was set up in a randomized complete block design with three replications. the experimental plot consisted of one row, 5 m long, with 50 plants. rows were spaced 0.90 m apart. the traits assessed included: number of ears per plot (ne), weight of ears husked (mecp) and dehusked (mesp), length of ears husked (cecp) and dehusked (cesp), diameter of ears husked (decp) and dehusked (desp), plant height (ph) and ear height (eh). general and specific combining ability effects (gca and sca) were estimated by griffing's method 4, model 1. there was predominance of additive effects for weigth of ears husked and dehusked. however, for other parameters, such as length of ears dehusked, diameter of ears husked and dehusked, plant height and ear height, there was equivalence of additive and non-additive gene effects. the family 27 is indicated for obtantion of superior genotypes from the estimates of combining abilities for number of ears per plot, weight of ears husked and dehusked.

Abstract: the genetic characterization for peach varieties tropical and dourad?o was realized by the rapd method, also comparing with dourado-1, tutu, maravilha, rubro-sol, flordaprince, aurora-1 and talism? varieties, from iac "frutas de caro?o" germplasm active bank, maintained in núcleo de agronomia de cap?o bonito, using dna extracted of young leaves, with the purpose of establishing efficient markers in the variety identification. were obtained 31 markers from highly polimorphic primers opad10, opae04, opae07, opae09, opae11, opaj04 and opaj06, selected from 96 primers evaluated. the rapd markers used, indicated the relationship degree among varieties in an efficient way; except in varieties of free polinization origin. even among the varieties siblings as tutu and talism? had certain genetic distance (0,29), showing that the rapd is capable to characterize kindred materials. the tropical and dourad?o peaches varieties were well characterized in relation to the parents and others varieties, with genetic distances varying fron 0,26 to 0,89.

Abstract: A estrutura genética de Aspidosperma polyneuron Muell Arg. (Apocynaceae)foi estudada utilizando a análise de marcadores microssatélites, nas localidades de Bauru,Gália, Avaré, Piracicaba, Valinhos, Teodoro Sampaio, Botucatu, Porto Ferreira, municípiosdo Estado de S o Paulo, e Selvíria (Mato Grosso do Sul) onde foram coletadas folhas paraextra o de DNA da popula o de cada local. Foram obtidos sete locos polimórficos e quatromonomórficos. Analisando os locos polimórficos, observou-se que o número de alelosvariou de dois a seis, que demonstraram uma alta heterozigosidade (ho = 0,5209). Em oitopopula es estudadas, os coeficientes de fixa o f foram negativos com média de -0,0173 dasnove popula es e a fixa o total F foi 0,1034, enquanto o valor de coancestralidade θ p entrepopula es foi 0,1187. Foram também obtidos valores de fixa o dentro de popula es FIS(-0,0669), fixa o total FIT (0,0747) e fixa o genética entre popula es com FST (0,1343) eGST (Hedrick) (0,4875). O fluxo gênico médio obtido Nm foi 1,6121. Exibe estrutura genéticatípica de espécie alógama, com baixo coeficiente de endogamia e alta heterozigosidade, comisolamento entre popula es. A popula o Piracicaba indicou a maior eros o genética eisolamento; as outras popula es ainda mostram a variabilidade original para coletas visandoà conserva o e melhoramento genético.The genetic structure of Aspidosperma polyneuron Muell Arg. (Apocynaceae)was studied with SSR marker analysis, in Bauru, Gália, Avaré, Piracicaba, Valinhos, TeodoroSampaio, Botucatu, Porto Ferreira (S o Paulo State cities), and Selvíria city (Mato Grosso doSul State), where leaves were collected for extracting DNA from the population of each site.Seven polymorphic and four monomorphic loci were obtained. Analyzing polymorphic loci,allele number ranged from two to six alleles, and showed high heterozigosity (ho = 0.5209).In eight populations the endogamy inbreeding coefficient f was negative, the nine populationsaverage was -0.0173, and total inbreeding coefficient F was 0.1034, meanwhile the coancestryvalue θ p among populations was 0.1187. Values of fixation within population FIS (-0.0669),total inbreeding FIT (0.0747), genetic fixation among populations with FST (0.1343) andGST (Hedrick) (0.4875) index were also obtained. Abstract: A espécie Eucalyptus grandis é a mais comumente cultivada em estandes comerciais no Brasil e no mundo. A espécie é plantada como cultivar e também na forma de plantios clonais de seus híbridos interespecíficos. Este trabalho é um estudo da variabilidade genética em progênies de poliniza o livre de Eucalyptus grandis (Hill) Maiden, oriundas de pomar de sementes por muda, cuja procedência é originária de Coff’s Harbour – Austrália, de propriedade da empresa Duratex S.A., localizado na Fazenda Morro D’Ouro, no município de Botucatu. Teve como objetivo analisar as correla es entre diferentes caracteres nas diferentes idades, visando subsidiar a sele o precoce em programas de melhoramento para essa espécie. Os ensaios foram instalados em dois locais, Angatuba–SP e Len óis Paulista–SP, em fevereiro de 1988. O delineamento experimental utilizado foi o de blocos casualizados, com dez repeti es, seis plantas por parcela, ao espa amento de 3 x 2 m e totalizando 76 progênies. Os caracteres avaliados foram: (1) diametro à altura do peito – (DAP); altura total e volume de madeira. As avalia es foram feitas em quatro anos iniciando a partir do segundo ano em Len óis Paulista (local 2) até o quinto ano, sendo que em Angatuba (local 1) foram realizadas avalia es anuais do segundo ao sexto ano. Os resultados mostraram que as correla es entre os caracteres de crescimento apresentaram valores elevados (0,69 a 0,99), tanto genéticos como fenotípicos, facilitando os procedimentos de sele o no avan o das gera es de melhoramento, podendo ser realizada desde o terceiro ano de idade com maior peso para DAP e volume em um índice de sele o.The species Eucalyptus grandis (Hill) Maiden is the most commercial tree cultivated in Brazil and throughout the world. It is planted as varieties and also as interspecific hybrid clonal plantings. This work is a study of genetic variation in open pollinated progenies, a seedling seed orchard of E. grandis from a Coff’s Harbour provenance, located at Morro D’Ouro Duratex S.A., in the Experimental Station in Botucatu County, S o Paulo, Brazil. Abstract: Foi realizada a caracteriza o genética das cultivares de pêssego Tropical e Dourad o pelo método RAPD, em compara o com 'Dourado-1', 'Tutu', 'Maravilha', 'Rubro-sol', 'Flordaprince', 'Aurora-1' e 'Talism ' do Banco Ativo de Germoplasma de Frutas de Caro o do IAC, mantido no Núcleo de Agronomia de Cap o Bonito, utilizando DNA extraído de folhas jovens. Foram obtidos 31 marcadores genéticos a partir dos primers altamente polimórficos OPAD10, OPAE04, OPAE07, OPAE09, OPAE11, OPAJ04 e OPAJ06, selecionados de 96 primers avaliados. Os marcadores RAPD utilizados indicaram eficientemente o grau de parentesco entre cultivares, exceto em cultivares originadas de poliniza o livre. Verificou-se que, mesmo entre as cultivares irm s como 'Tutu' e 'Talism ', encontrou-se certa distancia genética (0,29), mostrando que por RAPD é possível caracterizar-se germoplasma aparentado. 'Tropical' e 'Dourad o' foram bem caracterizados em rela o aos parentais e demais cultivares, com as distancias genéticas variando de 0,26 a 0,89.
ATIVIDADE ENZIMáTICA EM PL NTULAS DE Eucalyptus grandis PROVENIENTES DE SEMENTES ENVELHECIDAS ARTIFICIALMENTE E NATURALMENTE

Abstract: O presente estudo empregou o procedimento de envelhecimento artificial, submetendo sementes por 96 horas à 42° C e 100% de UR para compará-las com sementes estocadas por 5, 10 e 15 anos e utilizou eletroforese de isoenzimas e atividade de peroxidase por método colorimétrico. Houve um decréscimo significante na germina o das sementes envelhecidas artificialmente, quando comparadas com as sementes estocadas. A atividade da peroxidase foi baixa, porém houve uma curva ascendente que acompanhou a idade das sementes. Observou-se um aumento nas atividades da malato desidrogenase e a-esterase 2 e 3 e uma diminui o acentuada para a-esterase 1 e baixa para a fosfatase ácida. Os resultados acompanharam o aumento do tempo de estocagem das sementes e do estresse que ocorre por causa das condi es de temperatura e umidade empregadas no processo de envelhecimento artificial.

Abstract: O objetivo desse trabalho foi quantificar a dissimilaridade genética entre progênies de P. caribaea var. caribaea para identificar genitores divergentes, visando gerar genótipos mais produtivos e avaliar a variabilidade total por meio de alguns caracteres fenotípicos de importancia comercial. Assim, um teste de progênies de P. caribaea var. caribaea foi implantado em delineamento experimental látice 10 x 10 triplo, com 99 progênies e uma testemunha comercial em Selvíria, MS. Foram avaliados os caracteres quantitativos antes e após desbaste aos 14 anos, tais como: altura total de plantas (m), diametro à altura do peito - DAP (cm); volume (m3 árvore-1); forma do fuste das árvores, densidade básica da madeira, na altura do DAP - DBM-1, e na metade da altura da árvore - DBM-2 (g cm-3); e sobrevivência das progênies (%). Houve baixa divergência genética entre as progênies de P. caribaea var. caribaea. O DAP foi o caráter que mais contribuiu para a divergência genética entre as progênies, tanto antes quanto depois do desbaste. As informa es geradas com base na distancia generalizada de Mahalanobis e o agrupamento das progênies pelo método de Tocher ser o usadas para definir as estratégias a serem adotadas em futuros programas de melhoramento e conserva o genética desta espécie. doi: 10.4336/2012.pfb.32.69.69 The objective of this research was to quantify the genetic dissimilarity among P.caribaea var. caribaea progenies to identify divergent parents aiming to generate highly productive genotypes and to evaluate the total variability by phenotypic characters of commercial importance. The progeny trial of P. caribaea var. caribaea was set up through 10 x 10 triple lattice design, with 99 progenies and a commercial control in Selvíria, MS, Brazil. We have evaluated quantitative characters before and after thinning procedures such as: total plant height (m), diameter at breast height (dbh) (cm); wood volume (m3 tree-1), stem form, wood density at dbh and wood density at the middle of the total height of tree stem (g cm-3), and survival of progenies (%). There was low genetic divergence among P. caribaea var. caribaea progenies. The diameter at breast height was the character that most contributes to genetic diversity among progenies, for both before and after thinning. The information of Mahalanobis distance and the clustering of progenies by Tocher's method will be important to define the future strategies for breeding programs and genetic conservation of the species. doi: 10.4336/2012.pfb.32.69.69

Abstract: eucalypt plantation has high economical importance in brazil; however, it has been attacked by various pathogens under different environmental stress conditions. disease resistance and survival under unfavorable environmental conditions have revealed that the eucalypt has developed highly efficient defense systems. here we show the results of the eucalyptus ests genome project (forests). using the expressed sequence tags (ests) obtained by the project, contigs of similar sequences from each cdna library induced and not induced by stress agents were formed, and cdna sequences similar to other already known molecules, such as plant-signaling molecules, phytoalexins, lignin biosynthesis pathways, pr-proteins and putative genes corresponding to enzymes involved in the detoxification of reactive oxygen species, were identified. we also present general considerations about the mechanisms of eucalyptus defense against biotic and abiotic stresses. these data are of extreme importance for future eucalypt breeding programs aimed at developing plants with enhanced resistance against pathogens and environmental stresses.
