Two Coregulated Efflux Transporters Modulate Intracellular Heme and Protoporphyrin IX Availability in Streptococcus agalactiae

Abstract: Streptococcus agalactiae is a major neonatal pathogen whose infectious route involves septicemia. This pathogen does not synthesize heme, but scavenges it from blood to activate a respiration metabolism, which increases bacterial cell density and is required for full virulence. Factors that regulate heme pools in S. agalactiae are unknown. Here we report that one main strategy of heme and protoporphyrin IX (PPIX) homeostasis in S. agalactiae is based on a regulated system of efflux using two newly characterized operons, gbs1753 gbs1752 (called pefA pefB), and gbs1402 gbs1401 gbs1400 (called pefR pefC pefD), where pef stands for ‘porphyrin-regulated efflux’. In vitro and in vivo data show that PefR, a MarR-superfamily protein, is a repressor of both operons. Heme or PPIX both alleviate PefR-mediated repression. We show that bacteria inactivated for both Pef efflux systems display accrued sensitivity to these porphyrins, and give evidence that they accumulate intracellularly. The ΔpefR mutant, in which both pef operons are up-regulated, is defective for heme-dependent respiration, and attenuated for virulence. We conclude that this new efflux regulon controls intracellular heme and PPIX availability in S. agalactiae, and is needed for its capacity to undergo respiration metabolism, and to infect the host.
Vertical Heterogeneity of Genotypic Structure and Toxic Potential within Populations of the Harmful Cyanobacterium Microcystis aeruginosa  [PDF]

Abstract: We investigated the vertical variability of toxic potential (i.e. proportions of cells containing microcystin genes) and genotypic structure within different populations of Microcystis aeruginosa that developed in deep artificial reservoirs on the Loire River (France). We demonstrated that a great qualitative vertical heterogeneity could exist within a single bloom of this cyanobacterium in deep lakes. Indeed, we observed important vertical shifts of both toxic potential and genotypic structure, whatever the bloom magnitude. These variations occurred mainly within the euphotic zone and proved to occur independently from abundance vertical shifts. One of the most striking results of this study is that the genotypic structure of a population of M. aeruginosa was more variable between different depths sampled at a single site than between different sites of the same reservoir sampled on top of the water column. In the same way the proportion of potentially toxic cells was sometimes more variable vertically than horizontally. The occurrence of such vertical heterogeneity in three different blooms suggests that this could be a frequent pattern within populations of M. aeruginosa.
Palynological and Physicochemical Characterization of Honey in the Sudano-Guinean Zone of Cameroon  [PDF]

Abstract: The palynological and physico-chemical characterization of honey was investigated in the Sudano- Guinean zone of Cameroon through the melissopalynology analysis. Results showed that honey with the dark amber color was the most represented. Sweet honey with less odour considered as pleasant highly predominate. The pH of honey is low and varies from 3.95 to 5.00; this ranking confirms the hypothesis that honey is from plant through visit of honeybees. The sugar content is very high ranging from 77.93% to 83.13%; the mean value is 78.90% and this value confirms that the honey is floral honey. The density value ranges from 1.39 to 1.43 and does not exceed the average standard (1.39 to 1.44 at 20). Water contents correspond to the standards norms. For free acidity, the highest value is 48.68 ± 0.28. There is a low positive correlation (r = 0.2) between free acidity and the pH of honey. Pollen analysis of honeys led to the identification of 41 pollen species belonging to 25 families. The highly represented families are Asteraceae and Myrtaceae. Two thirds of honey is monofloral and characteristics of three dominant species: Nymphaea maculata, Terminalia avicennioides and Syzygium guineense. The pollens of Syzygium guineense are present in all honey samples analysed.

Abstract: Sweet Sweetback’s Baadasssss Song represented a turning point in filmmaking when it was released in 1971. This article focuses on the interplay of the political and the aesthetic in a film that was conceived as a means of furthering change in the representation of African Americans. Not only did it support the emergence of black cinema by opening up filmmaking to black crews, but it also broached taboo subjects while depicting black sexuality (miscegenation) and police brutality. Melvin Van Peebles proposed a new heroic figure through the character of Sweetback, whose hypersexuality was to become a mercantile asset in Hollywood blaxploitation films. Nevertheless, the narrative’s reliance on a stereotypical Sweetback undermined its political message.

Abstract: Michel Houellebecq is a writer who cultivates an incongruous literary garden, where the necessity of describing contemporary objects as equipment for the manufacture of individuals can be said to precede his concern for literary conventions. This has been at the centre of much criticism formulated around Houellebecq’s novels. One book in particular by Eric Naulleau called Au Secour, Houellebecq Revient! attacks his novels on the grounds that their success relies on his charming the reader by using recognisable elements of everyday life and incorporating them in the novels, in such a way that they could no longer be distinguished from the world of everyday life.

Abstract: This article describes MorphoClust and MorphoNet, two methods for the unsupervised acquisition of morphological families. MorphoClust builds families by iterative conflations, similarly to hierchical clustering methods. The MorphoNet method relies on community detection in lexical networks. The nodes of these networks stand for words while edges represent morphological transformation rules which are automatically acquired based on graphical similarities between words. The two methods are applied to a German-English bilingual lexicon, both in isolation and in combination. We evaluate the results using the CELEX lexical database.

Abstract: In recent short stories written by diasporic Indian women writers, changes in terms of location or national identity are generally depicted as providing significant opportunities for Indian women to challenge and revise culturally-inscribed gender roles. In two collections of short stories written respectively by Ginu Kamani and Shauna Singh Baldwin, Junglee Girl and English Lessons and Other Stories, not only are ghosts employed to rupture boundaries between the living and the dead, but they also represent enabling presences which authorize female voices by resurrecting stories of women that have been silenced or forgotten. In these two books, the emergence of the archaic is recurrently tinged with liberating undertones since it opens up new spaces for identity by countering gendered expectations of ‘acceptable’ behaviour and by constructing alternative realities. My essay will therefore focus on the central metaphors of female ghosts and doubles as transitional figures through which women in transit empower themselves. My aim is to show how culturallydisplaced women appropriate the uncanny so as to engender new identities and assert the value of individual female experience. In these haunted narratives, I will contend, women move from a ghostly time of repetition to a ‘hauntology’ of new becomings. Abstract: Les recueils amoureux de Neruda sont l’étalon d’une poétique en évolution. Vingt poèmes d’amour proposent un lyrisme élégiaque : l’adresse est une vaine tentative de sortie de soi, elle ne parvient pas à toucher des figures de l’autre modelées par l’imaginaire. Au contraire, dans Les Vers du capitaine et La Centaine d’amour, l’adresse se précise sur un être unique. Mais paradoxalement, cette fixation du désir est en même temps ouverture à l’altérité et dépassement de la poésie intime. Neruda’s love poetry showcases the evolutions of his poetics. Twenty love poems offer an elegiac and solipsist version of lyricism : the poet creates multiple figures in an attempt to communicate with them, but fails to reach this aim. On the contrary, in the Captain’s Verses and the 100 Loves Sonnets, the address focuses on one single being. But paradoxically, this fixation of the poet’s desire gives access to otherness and to the collective dimension of poetry. Los libros de poemas de amor de Pablo Neruda son el marco de una poética en evolución. Veinte poemas de amor proponen un lirismo elegíaco : el apóstrofe es una vana tentativa para salir de sí pus no alcanza a tocar las figuras del otro modeladas por lo imaginario. Por el contrario, en Los versos del capitán y Cien sonetos de amor, el apóstrofe se objetiva en un ser único. Paradójicamente, esta fijación del deseo es al mismo tiempo una apertura a la alteridad y un ir más allá de la poesía íntima.
