Isolation and identification of bioactive compounds responsible for the anti-bacterial efficacy of Lotus corniculatus var. S o Gabriel

Abstract: Lotus corniculatus (Fabaceae) is considered a forage plant utilized as food for ruminants in the south of Brazil. This herb is also actually used to treat intestinal infection in these animals. In our experiments, we evaluated the anti-bacterial activity of crude extract from L. corniculatus var. Sγo Gabriel were assayed against Gram-positive and Gram-negative bacterium. The crude extracted did not show any anti-bacterial activity, but the hexane fraction did on Bacillus cereus (MIC=100 μg/mL) and on Enterococcus faecalis, Listeria monocytogenes, Staphylococcus aureus, Staphylococcus epidermidis, Acinetobacter calcoaceticus, and Providencia alcalifaciens (MIC=600, 800 or 1000 μg/mL). The Ethyl acetate fraction (AcOEt) also showed important anti-bacterial activity on Bacillus cereus, E. faecalis, and Acinetobacter calcoaceticus (MIC=800 μg/mL). The oleanolic acid isolated from hexane fraction showed the same effect on Staphylococcus aureus methycillin-resistant (MIC=100 μg/mL), L. monocytogenes (MIC=25 μg/mL), and Bacillus cereus (MIC=25 μg/mL). Further, Kaempferitrin isolated from ethyl acetate fraction has also shown anti-bacterial activity on Shighella flexinerii (MIC=100 μg/mL), Salmonella typhimurium (MIC=100 μg/mL), A. calcoaceticus (MIC=100 μg/mL), E. faecalis (MIC=3.9 μg/mL), and Bacillus cereus (MIC=8.5 μg/mL). This study suggests that L. corniculatus var. Sγo Gabriel have potential pharmacological property for a new anti-bacterial drug development.
Methotrexate inhibits integrin adhesion molecules in the mouse model of pleurisy induced by carrageenan

Abstract: the aim of this work was to analyze the effect of methotrexate (mtx) upon leukocyte migration and expression of adhesion molecules cd11a/cd18 in the lung, 4 and 48 h after inflammation induction by carrageenan in mice. the results showed that mtx significantly decreased leukocyte influx and cd11a expression in the lung at 4 and 48 h of pleurisy (p < 0.01). mtx also inhibited cd18 expression at 4 h but not 48 h of pleurisy (p < 0.01). these results proved that mtx at the studied doses had important anti-inflammatory properties, acting primarily on leukocyte migration from the pleural cavity to the lung via inhibition of cd11a/cd18 expression in the mouse model of inflammation.

Drug resistance of Mycobacterium tuberculosis strains in southern Brazil

Abstract: introduction: the aim of this work was to evaluate the prevalence of mycobacterium tuberculosis (mt) strains with mutations that could result in resistance to the main drugs used in treatment in a region with one of the highest numbers of tuberculosis (tb) cases in southern brazil. methods: deoxyribonucleic acid (dna) from 120 sputum samples from different patients suspicious of pulmonary tuberculosis who attended the municipal public laboratory for mycobacterium sp. diagnosis was directly amplified and analyzed by pcr-sscp. the dna was amplified in known hotspot mutation regions of the genes rpob, ahpc, embb, katg, inha, and pnca. results: the percentage of samples positive by culture was 9.2% (11/120); 5% (6/120) were positive by bacilloscopy and mt-pcr, and dna fragments of the aforementioned resistance genes could be amplified from seven (7) of the eleven (11) samples with positive results, either by culture or pcr/bacilloscopy. all presented a sscp pattern similar to a native, nonresistant genotype, with the atcc strain 25177 as control, except for one sample (0.01%), which presented a sscp profile demonstrating mutation at the embb gene. conclusions: these results are consistent with the empirical observations by physicians treating tb patients in our region of a low occurrence of cases that are refractory to conventional treatment schemes, in contrast to other parts of the country. continued surveillance, especially molecular, is essential to detect and monitor the outbreak of mt-resistant strains.
Carvedilol atenua o estresse oxidativo na cardiopatia chagásica cr?nica

Abstract: background: there is increasing evidence suggesting that chagas disease involves oxidative damage and contributes to heart disease progression. objective: to evaluate the effect of carvedilol on oxidative stress markers in chronic chagas disease. methods: the study population included 42 patients with chagas cardiomyopathy and oxidative stress biomarkers were measured before and after a period of six months of treatment with carvedilol (37.5 mg/day). patients were considered according to the los andes classification and the activity of superoxide dismutase, catalase, glutathione peroxidase, s-transferase and reductase, myeloperoxidase and adenosine deaminase; levels of reduced glutathione, thiobarbituric acid reactive species, carbonyl protein, vitamin e and nitric oxide were measured in blood. results: after treatment with carvedilol, all groups showed significant reductions in levels of carbonyl protein and reduced glutathione, whereas the levels of nitric oxide and adenosine activity increased significantly only in group ia. moreover, most of the antioxidant enzymes showed decrease in activity in groups ia and ib. conclusion: the data suggest that treatment with carvedilol was effective in attenuating oxidative damage, an effect that may be particularly important in patients with chronic chagas' disease cardiomyopathy.
Chemical characterization of essential oils from Drimys angustifolia miers (Winteraceae) and antibacterial activity of their major compounds

Abstract: essential oils have been extensively studied in recent years as a natural source of new antimicrobial agents. in this work, essential oils of leaf and branch from drimys angustifolia growing in southern brazil were obtained by hydrodistillation and analyzed by gas chromatographies with flame ionization detector (gc-fid) and with mass spectrometer (gc-ms). drimenol and bicyclogermacrene were isolated by column chromatography from branch and leaf essential oils, respectively. oils, isolated compounds and combinations of them were assayed against gram-(+) and gram-(-) bacteria. the oils showed to be more active against bacillus cereus, with minimum inhibitory concentration (mic) 125 and 250 μg ml-1 for branch and leaf oils, respectively, strongly inhibiting bacterial growth. bicyclogermacrene was more active then drimenol, providing a mic value of 167 μg ml-1 against b. cereus. synergism was not observed in any of the combinations tested.

Abstract: this work reports the chemical composition as well as the antibacterial, antifungal and antiparasitic activities of the leaf essential oil from piper malacophyllum. the oil was extracted by hydrodistillation and analyzed by gc-fid, gc-ms and polarimetry. among the 28 compounds identified, (+)-camphor was the major constituent. the essential oil showed activity against most of the microorganisms tested, especially antifungal action, with a mic of 500 μg ml-1 against trichophyton mentagrophytes and cryptococcus neoformans. this is the first study reporting the composition and biological properties of leaf essential oil from p. malacophyllum.
The Silent Run: New Issues and Outcomes for University-Industry Relations in Brazil

Abstract: in order to develop innovations, companies are establishing university-industry (u-i) relations which are explained in theories like the triple helix. maybe this traditional view it is not enough to describe what happens in brazil. followed by a growth of articles published internationally, companies in technologically stable industries seem to be incorporating high-tech outcomes from universities, boosting sectors that once were apparently inert. this research will propose a different approach on u-i relations, focusing on the knowledge flow. results show that agriculture companies are increasing their relations as much as engineering companies. such relations are mainly based on raw technologies or applied solutions. start-up firms' relations are mainly based on informal contacts and patents are not used to protect technologies. the silent run shows that companies which appear to be technologically stable are now upgrading trough u-i, and patents does not seem to be the first option when protecting new technologies.
Evaluation of the antiinflammatory efficacy of Lotus pedunculatus

Abstract: The aim of this study was to investigate the antiinflammatory effect of the crude extract of Lotus pedunculatus. Aerial parts of Lotus pedunculatus were extracted by maceration with ethanol 96%. The solvent was evaporated under low pressure at 55°C to dryness, yielding crude extract. The antiinflammatory effects of Lotus pedunculatus were analyzed using carrageenan-induced mouse pleurisy. Significance was determined by analysis of variance, Dunnett′s or Student′s t -tests. Values of P < 0.05 were considered significant. The crude extract of Lotus pedunculatus inhibited leukocyte, neutrophil, myeloperoxidase, and adenosine-deaminase activities, as well as exudation, nitric oxide, interleukin-1 beta (IL-1β) and tumor necrosis factor-alpha (TNF-α) levels ( P < 0.05). These results indicate that Lotus pedunculatus has an important antiinflammatory action and its inhibitory activity may be due to not only to the inhibition of proinflammatory mediators, but also to the inhibition of the activated leukocytes.

Abstract: This paper addresses the topic of sustainable consumption and ecolabelling, and the impact that these issues have for organizations, especially as regards the production of consumer goods and its marketing/sales. It starts with a literature review focusing on the movement of conscious consumption and the development of the theme of sustainable certifications, ending with an assessment of Brazil in this context, where it was found that few academic studies have been conducted on this topic in general. Then we report the results of an exploratory qualitative research, conducted through in-depth interviews. The results of the research indicates a low level of knowledge and use of sustainable labels. There is a lack of information to consumers, aiming to facilitate the choice of more sustainable products. We noticed that people have not realized the influence they have with their power of choice, showing that consumption can be a space for political debate, both by the problems associated with it, and by understanding the consumers have power to change the business as usual.
