Plasminogen Activator Inhibitor-1 4G/5G Gene Polymorphism and Coronary Artery Disease in the Chinese Han Population: A Meta-Analysis

Abstract: Background The polymorphism of plasminogen activator inhibitor-1 (PAI-1) 4G/5G gene has been indicated to be correlated with coronary artery disease (CAD) susceptibility, but study results are still debatable. Objective and Methods The present meta-analysis was performed to investigate the association between PAI-1 4G/5G gene polymorphism and CAD in the Chinese Han population. A total of 879 CAD patients and 628 controls from eight separate studies were involved. The pooled odds ratio (OR) for the distribution of the 4G allele frequency of PAI-1 4G/5G gene and its corresponding 95% confidence interval (CI) was assessed by the random effect model. Results The distribution of the 4 G allele frequency was 0.61 for the CAD group and 0.51 for the control group. The association between PAI-1 4G/5G gene polymorphism and CAD in the Chinese Han population was significant under an allelic genetic model (OR = 1.70, 95% CI = 1.18 to 2.44, P = 0.004). The heterogeneity test was also significant (P<0.0001). Meta-regression was performed to explore the heterogeneity source. Among the confounding factors, the heterogeneity could be explained by the publication year (P = 0.017), study region (P = 0.014), control group sample size (P = 0.011), total sample size (P = 0.011), and ratio of the case to the control group sample size (RR) (P = 0.019). In a stratified analysis by the total sample size, significantly increased risk was only detected in subgroup 2 under an allelic genetic model (OR = 1.93, 95% CI = 1.09 to 3.35, P = 0.02). Conclusions In the Chinese Han population, PAI-1 4G/5G gene polymorphism was implied to be associated with increased CAD risk. Carriers of the 4G allele of the PAI-1 4G/5G gene might predispose to CAD.
α-Adducin Gly460Trp Gene Mutation and Essential Hypertension in a Chinese Population: A Meta-Analysis including 10960 Subjects

Abstract: Background The α-adducin Gly460Trp (G460W) gene polymorphism may be associated with susceptibility to essential hypertension (EH), but this relationship remains controversial. In an attempt to resolve this issue, we conducted a meta-analysis. Methods Twenty-three separated studies involving 5939 EH patients and 5021 controls were retrieved and analyzed. Four ethnicities were included: Han, Kazakh, Mongolian, and She. Eighteen studies with 5087 EH patients and 4183 controls were included in the Han subgroup. Three studies with 636 EH patients and 462 controls were included in the Kazakh subgroup. The Mongolian subgroup was represented by only one study with 100 EH patients and 50 controls; similarly, only one study with 116 EH patients and 326 controls was available for the She subgroup. The pooled and ethnic group odds ratios (ORs) along with the corresponding 95% confidence intervals (95% CI) were assessed using a random effects model. Results There was a significant association between the α-adducin G460W gene polymorphism and EH in the pooled Chinese population under both an allelic genetic model (OR: 1.12, 95% CI: 1.04–1.20, P = 0.002) and a recessive genetic model (OR: 1.40, 95% CI: 1.16–1.70, P = 0.0005). In contrast, no significant association between the α-adducin G460W gene polymorphism and EH was observed in the dominant genetic model (OR: 0.88, 95% CI: 0.72–1.09, P = 0.24). In stratified analysis by ethnicity, significantly increased risk was detected in the Han subgroup under an allelic genetic model (OR: 1.13, 95% CI: 1.04–1.23, P = 0.003) and a recessive genetic model (OR: 1.43, 95% CI: 1.17–1.75, P = 0.0006). Conclusions In a Chinese population of mixed ethnicity, the α-adducin G460W gene polymorphism was linked to EH susceptibility, most strongly in Han Chinese.
Tumor Necrosis Factor-Alpha G308α Gene Polymorphism and Essential Hypertension: A Meta-Analysis Involving 2244 Participants

Abstract: Background The tumor necrosis factor-alpha (TNFα) G308A gene polymorphism has been implicated in susceptibility to essential hypertension (EH), but study results are still controversial. Objective and Methods The present meta-analysis is performed to investigate the relationship between the TNFα G308A gene polymorphism and EH. Electronic databases were searched and seven separate studies on the association of the TNF α G308A gene polymorphism with EH were analyzed. The meta-analysis involved 1092 EH patients and 1152 controls. The pooled odds ratios (ORs) and their corresponding 95% confidence interval (CI) were calculated by a fixed or random effect model. Results A significant relationship between the TNFα G308A gene polymorphism and EH was found in an allelic genetic model (OR: 1.45, 95% CI: 1.17 to 1.80, P = 0.0008), a recessive genetic model (OR: 3.181, 95% CI: 1.204 to 8.408, P = 0.02), and a homozygote model (OR: 3.454, 95% CI: 1.286 to 9.278, P = 0.014). No significant association between them was detected in both a dominant genetic model (OR: 1.55, 95% CI: 0.99 to 2.42, P = 0.06) or a heterozygote genetic model (OR: 1.45, 95% CI: 0.90 to 2.33, P = 0.13). Conclusion The TNFα G308A gene polymorphism is associated with EH susceptibility.
Tea polyphenol inhibits colorectal cancer with microsatellite instability by regulating the expressions of HES1, JAG1, MT2A and MAFA

Abstract: Objective: To investigate the mechanism of tea polyphenol in inhibiting microsatellite instability (MSI) of colorectal cancer.Methods: Using LoVo cells and SW480 cells treated with aqueous solution of tea polyphenol, cell proliferation was detected by methyl thiazolyl tetrazolium (MTT) method, changes in microsatellite sequences were detected by genescan method and changes in gene expression of LoVo cells were detected by illumina expression arrays and quantitative real-time polymerase chain reaction (PCR).Results: The proliferation inhibition rates of LoVo and SW480 cells treated with tea polyphenol increased with the increasing of drug concentration and showed an increasing tendency with time. The proliferation inhibition rate of LoVo cells with tea polyphenol was higher than that of SW480 cells, and there was a significant difference in the proliferation inhibition rates at 24 h, 72 h and one week. The microsatellite sequence of LoVo cells treated with tea polyphenol remained stable. The gene expression arrays and quantitative real-time PCR suggested that tea polyphenol inhibited the gene expressions of MT2A, MAFA, HES1 and JAG1 nearly two-fold over controls. It was also found that tea polyphenol inhibited the BAX and p38 genes with a more than two-fold difference but did not significantly inhibit the nuclear factor-κB pathway.Conclusion: Tea polyphenol significantly inhibited the proliferation of MSI colorectal cancer cells and stably maintained the microsatellite state in MSI colorectal cancer. Tea polyphenol inhibited the gene expressions of HES1, JAG1, MT2A and MAFA, up-regulated the gene expression of BAX and down-regulated that of P38. Further research is required to investigate how these pathways are interrelated.
Effects of Huangqi Guizhi Wuwu Decoction given by different administration methods on rats with frostbite and the mechanism

Abstract: Objective: To establish a rat model of frostbite and to evaluate the effects of different administration methods of Huangqi Guizhi Wuwu Decoction (HGWD), a compound traditional Chinese herbal medicine for warming meridians to disperse cold, on rats with frostbite.Methods: Frostbite in rats was induced by the method of soaking feet in hypothermia ethanol-water mixture. Levels of interleukin-6 (IL-6), tumor necrosis factor-β (TNF-β), thromboxane B2 (TXB2) and 6-keto-prostaglandin F1α (6-keto-PGF1α) in serum of rats treated with different administration methods of HGWD, such as oral administration (Oral HGWD), soak (Soak HGWD), and oral administration plus soak (Oral-soak HGWD), were tested by enzyme-linked immunosorbent assay. Results: IL-6, TNF-β, TXB2 levels were significantly higher (P<0.01) and 6-keto-PGF1α level was lower (P<0.01) in serum of rats in the untreated group than in the normal control group. Compared with the untreated group, the level of IL-6 obviously decreased (P<0.05) in serum of the rats treated by oral HGWD, while no significant decrease (P>0.05) was observed in the soak HGWD group, and there was no interaction (P>0.05) between the two administration methods in regulating the level of IL-6. The levels of TNF-β obviously decreased (P<0.01, P<0.05) in serum of the rats treated by oral and soak HGWD, and there was interaction between the two administration methods. The level of TNF-β in the oral HGWD group was significantly lower than that in the soak HGWD group (P<0.01). Compared with the untreated group, level of TXB2 in oral HGWD or soak HGWD group did not decrease significantly (P>0.05) and there was no interaction (P>0.05) between the two administration methods. The level of 6-keto-PGF1α was obviously increased (P<0.01) in serum of the rats treated by oral HGWD, while there was no significant decrease (P>0.05) in the soak HGWD group as compared with the untreated group, and there was interaction (P<0.05) between the two administration methods in regulating the level of 6-keto-PGF1α.Conclusion: Rats with frostbite has immunologic dysfunction and a state of forming thrombus easily. The oral-soak HGWD can improve frostbite of local skin in rats. The therapeutic mechanism of HGWD may be to regulate the dysfunction of immune system and the imbalance of TXB2-PGF1α.

Abstract: A series of second amines were used to catalyze the synthesis of methacrolein from formaldehyde and propionaldehyde via Mannich reactionBy using diethylamine,the highest yield of 94% was obtained.The kinetic model was obtained by analysis of the reaction process.The parameters in the model were determined:reaction order 2.456,activation energy 62.45 kJ/mol and frequency factor 8.182×107(mol/L)-1.456/s.In situ FT-IR spectroscopy was introduced to monitor the concentration of compounds in real time during the reaction.The samples taken in different reaction times were analyzed by GC-MS,and the intermediate Mannich base was detected,laying a foundation for the reaction mechanism.

Abstract: A series of supported intermetallic Pb-Pb catalysts were prepared with the impregnation method by changing the support (silica, molecular sieve or γ-alumina) and the pore size. The chemical states of the two metals were characterized by XPS analysis, the process for producing methyl methacrylate based on the direct oxidative esterification of methacrolein with methanol in the presence of oxygen was performed in a slurry reactor with the above-mentioned catalysts. The influence of the calcination temperature and the kinds of support as well as the pore size on catalytic activity had been extensively investigated. Under the conditions of temperature at 80 ℃, catalyst 3.8% (ω) and the reaction time 2 h, the conversion rate of methacrolein reached 85%, the selectivity and the yield of methyl methacrylate were 90% and 76.5%, respectively.

Abstract: We discuss the motions of curves by introducing an extra spatial variable or equivalently, moving surfaces in affine geometries. It is shown that the 2+1-dimensional breaking soliton equation and a 2+1-dimensional nonlinear evolution equation regarded as a generalization to the 1+1-dimensional KdV equation arise from such motions.

Abstract: 本文报道了担子菌菌种24属43种122株经过液氮超低温冻结后的结果。122株担子菌分别用5%或10%二甲亚砜(DMSO)和10%甘油作保护剂,经快速和慢速冻结其成活率分别是,以甘油作保护剂,快速冻结为81%;慢速冻结为94%;以DMSO作保护剂时,快速冻结为97%;慢速冻结为100%。14属23种41株担子菌经冻结保存不同时间后,进行结菇试验,除银耳 (Tremella fucifor-mis) 的对照和冻结样品均未形成子实体外,其余属种均保持着结菇这一特性。
