Development of Sequence-Based Microsatellite Marker for Phalaenopsis Orchid

Abstract: Phalaenopsis is one of the most interesting genera of orchids due to the members are often used as parents to produce hybrids. The establishment and development of highly reliable and discriminatory methods for identifying species and cultivars has become increasingly more important to plant breeders and members of the nursery industry. The aim of this research was to develop sequence-based microsatellite (eSSR) markers for the Phalaenopsis orchid designed from the sequence of GenBank NCBI. Seventeen primers were designed and thirteen primers pairs could amplify the DNA giving the expected PCR product with polymorphism. A total of 51 alleles, with an average of 3 alleles per locus and polymorphism information content (PIC) values at 0.674, were detected at the 16 SSR loci. Therefore, these markers could be used for identification of the Phalaenopsis orchid used in this study. Genetic similarity and principle coordinate analysis identified five major groups of Phalaenopsis sp. the first group consisted of P. amabilis, P. fuscata, P. javanica, and P. zebrine. The second group consisted of P. amabilis, P. amboinensis, P. bellina, P. floresens, and P. mannii. The third group consisted of P. bellina, P. cornucervi, P. cornucervi, P. violaceae sumatra, P. modesta. The forth group consisted of P. cornucervi and P. lueddemanniana, and the fifth group was P. amboinensis.
β-Glucan production of Saccharomyces cerevisiae in medium with different nitrogen sources in air-lift fermentor

Abstract: β-Glucan is one of the most abundant polysaccharides in yeast Saccharomyces cerevisiae cell wall. The aim of this research is to explore an alternative nitrogen sources for β-glucan production. S. cerevisiae were grown in fermentation medium with different nitrogen sources. Peptone 2%, glutamic acid 0,5%, urea 0,2%, and diammonium hydrogen phosphate (DAHP) 0,02% were used for nitrogen source in the medium. A two liter air-lift fermentor was used in the fermentation process for 84 hours (T = 300C, pH 7, and 1.5 vvm for the aeration). During the fermentation, optical density, extraction of β-glucan, glucose and protein in hydrolisate cultured were determined. β-glucan production level is similar with the growth rate of yeast and followed by decreasing glucose and protein content in hydrolysis cultured. The highest and lowest β-glucan content were obtained from peptone (933.33 mg/L) and glutamic acid (633.33 mg/L) as a nitrogen source in cells cultured after fermentation completed respectively. Yeast cells cultured with urea and DAHP as a nitrogen source give the same content of β-glucan about 733.33 mg/L. β-glucan concentration produced in medium with urea was a higher than that produced using glutamic acid and DAHP as a nitrogen source. The result indicated that urea can be used as an alternative nitrogen source for the production of β-glucan. Urea is easily available and cheaper than peptone, glutamic acid and DAHP.

Abstract: As we know, in mid July 1999, Indonesia is suffering an economic crisis that shocked every economist in Indonesia, economic cycle jammed and many enemployment and "Pemutusan Hubungan Kerja" (PHK). The change of economical pattern either in the society or among the enterpreneur has gone in a drastic way and causes a very big trouble to the Indonesian government to stabilize the economic cycle. Many economist from may kind of proffesion or any work field try to save there own business in their own way and limited only in a micro scale. Even in the economic crisis that coused by the weakness in rupiah to US $, the economist behavior make the rupiah weaker, becouse they with hold cash rupiah and cash US $. To over come it, Indonesian government give a secure guarantee to the banks consumers so they will agree to release cash at home and deposit the money in the bank. In that way the economic cycle will run again and ever better. The purpose of this research is to see the differences of the economist behavior through their financial assets coused by the differences of proffesion and economical condition and how for they have been participated with the government policy in banking in January 1998. Untuk menanggulangi hal ini pemerintah Indonesia memberikan jaminan keamanan bagi nasabah bank agar mereka bersedia melepas uang tunai di rumah dan mendepositokannya di bank. E-TRANSLATOR WITH RULE BASED INDONESIA – MINANG DAN MINANG – INDONESIA

Abstract: Bahasa Minang sebagai salah satu bahasa daerah di Indonesia mulai terancam keberadaannya. Hal ini disebabkan oleh pengaruh bahasa Indonesia yang sudah menyebar jauh ke pelosok daerah di Sumatra Barat, yang dibawa oleh media audio visual seperti televisi dan radio. Berbagai usaha telah banyak dilakukan, salah satunya adalah pembuatan kamus untuk bahasa Minangkabau, namun kamus masih memiliki banyak keterbatasan, yaitu jika ingin mengetahui arti sebuah kata harus membaca satu persatu kata yang terdaftar, kata-kata yang terdapat dalam kamus juga dalam jumlah yang terbatas, padahal dalam bahasa Minang ternyata banyak memiliki kesamaan dengan bahasa Indonesia yang dapat dipolakan dengan aturan tertentu, sehingga kata-kata yang dapat dipolakan tidak harus didaftarkan dalam basis data. Tujuan dari penelitian ini adalah membangun aplikasi terjemahan kamus bahasa Indonesia – Minang dan sebaliknya yang dapat menterjemahkan kata dan kalimat menggunakan metode rule based berbasis web. Penelitian dimulai dengan mengumpulkan data berupa kosa kata bahasa Indonesia dan bahasa Minang, kemudian secara berturut-turut membuat algoritma rule based, membuat rancangan tampilan, membuat kamus, mengimplementasikan algoritma, dan langkah terakhir adalah melakukan pengujian dari aplikasi yang telah dibuat yaitu dengan Black Box Test dan Alpha Test. Penelitian yang dibuat menghasilkan sebuah aplikasi yang dapat menerjemahkan kata dan kalimat dari Bahasa Indonesia – Bahasa Minang serta bahasa Minang – Bahasa Indonesia. Prey diversity for Three Species Herons in Pulau Dua Nature Reserve Serang Distrik, Banten Province

Abstract: The diversity of prey consumed by three species herons (Casmerodius albus, Egretta garzetta and Bubulcus ibis) was studied in Pulau Dua Nature Reserve Serang, Banten Province using video tapes by following a focal individual for five minutes. In the laboratory the film was analysed every minutes to select complete sequences of feeding behavior in foraging site. There were differences on diversity and size of prey that consumed by three species herons among the different feeding locations. Casmerodius albus which was feeding on mudflat and fisheries consumed Chanos chanos (15.6%), Oreochromis sp (9.4%) and Periophtalmus sp. (7.3%), and 36.5% couldn’t be identified. Egretta garzetta was feeding on mudflat, fisheries and rice-field, consumed Periophtalmus sp (15.4%), Mugil dussumeieri (7.9%), Chanos chanos (4.7%), Oreochromis sp (0.5%), including small frog and crustacean (0.5%). Bubulcus ibis was feeding on rice-field and grassland, consumed many variety of insects (92 %) and many variety of snail (1.9%) in their diet. The insects that B. ibis consumed including grasshopper (26.8%), caterpillar (0.9%) and other invertebrates (worm, crustacean) that could not be identified (64.3%).
The feeding location of three species herons Casmerodius albus, Egretta garzetta, and Bubulcus ibis in Pulau Dua Nature Reserve, Serang, Banten Province

Abstract: This research is to know feeding location for three species herons that lived in Pulau Dua Nature Reserve (CAPD), Serang, Banten Province. This research has been done at feeding area for the herons around Pulau Dua Nature Reserve. The data were taken from film shooting use Focal Observation Methods, while the film shooting analysed use Scan Sampling Methods. Result showed that Casmerodius albus and Egretta garzetta were using the mudflat, fisheries and rice-fields around CAPD as feeding location. Bubulcus ibis was using rice-field and grassland as feeding location. The preference for C. albus to feeding in mudflat was ( 2=21691,29; db=2, =0,01). E. garzetta have preference for feeding in fisheries ( 2=18221,27; db=2, =0,01) and the preference for B. ibis to feeding in grassland is ( 2=27,57; db=1, =0,01). The selections of feeding location for three species herons were depend on the specific preys for them. C. albus and E. garzetta were select fish as their food, and B. ibis choose insect and other small animals as his food.
Type of intraspecific and interspecific interaction among three heron species, while foraging around Pulau Dua Nature Reserve, Serang, Banten's province

Abstract: This research intends to know interaction type among three heron species while foraging around Pulau Dua Nature Reserve, Serang Banten's Province. Observational method that is utilized is direct observing up to 6 months, pictures taking supported watch utilize Handycam by focal observation method, meanwhile data analyze was done by using scan sampling method. The research’s result points out that interaction during foraging for three species herons, namely Casmerodius albus, Egretta garzetta, and Bubulcus. Ibis, cover neutralism, competition and cooperation. C. albus showed cooperative's interaction as much as 66.67%. This interaction is happening between C. albus and Phalacrocorax sp. The competition and neutralism on C. albus each of them are 16.67%. E. garzetta showed neutralism as much as 50%, between E. garzetta and other waterbird which are Threskiornis melanocephalus and Plegadis falcinellus. Meanwhile, cooperative as high as 37.50% and competitions as high as 12.50%. B. ibis showed neutralism, as high as 44.44% between B. ibis and both Ardeola speciosa and P. falcinellus. Neutralism and competition interaction each of them are 33.33% and 22.22%.

Abstract: This paper is to argue that literature studies may help reveal the formation of young Indonesian female Muslim identity by looking at the books they read and write. It will particularly discuss two popular Islamic Chick Lit Santri Semelekete [Funky Islamic Boarding School Girl] (2005) by Ma’rifatun Baroroh and Jilbab Britney Spears: Catatan Harian Sabrina [Britney Spears’ Headscarf: Sabrina’s Diary] (2004) by Herlinatiens. The first part of the discussion will examine some external aspects such as physical presentation, biographical details of the authors, and publication-related matters. Then, using such side-line information, the next part will discuss the novels’ contents to see in what way they offer some cultural analysis of youth identity in contemporary Indonesian society.
Bringing American Popular Culture to the English Departments in Indonesia*

Abstract: One outcome of the globalization process is the growing influence and dominance of American popular culture. The speed with which American music, films, and goods have flooded the markets worldwide is remarkably high, thanks to the advancement of telecommunication technologies and the Internet. Increased cultural transfer or, more precisely, internationalization of American culture has posed both fear and fascination to other cultures. How do people in the academia respond to this conundrum of cross-cultural contacts? What do we teach when we teach popular culture? What viable research in American popular culture is encouraged so as to result in impartially beneficial impacts for society at large? This paper is to argue that one can become an avid learner or critic of a certain culture when s/he finds meaningful connections between that culture and life itself. The teaching of American popular culture in the English Department, for instance, has to be locally contextualized, learner-participant oriented, and socially self-actualized. In this way, American Studies outside the U. S. may in turn become less centralized as the interchange of cross-cultural understanding takes place concurrently.
