Immunohistochemistry Analysis of Proteins Related with Apoptosis as Prognostic Factor inEpidermoid Carcinomaof Penis

Abstract: the aim was to analyze the protein expression of apoptotic genes caspase-3, caspase-8 and bcl-2 with the immunohistochemistry technique, correlating with tumor grade (i, ii and iii) and with the patient survival in order to understand the basic mechanism of tumoral transformation. the immunohistochemistry reactions on 50 samples of squamous cell carcinoma were carried out with the avidin-biotin immunoperoxidase method and antigen recovery. the analyses were made using the graduation method "in crosses" (0 to 4 crosses - no stain to more than 75% of positives cells) and in categories (low, intermediate, high) of the cytoplasm immunoreactivity of the epidermoid penile carcinoma cells. it was observed a statistically significant difference when the expression of caspase-3 were compared with the grades i and ii of the tumor (p=0.0010) and when comparing the patient survival with the grades i and ii of the tumor (p=0.0212). the protein bcl-2 was more expressed than caspase-3 and caspase-8 proteins, suggesting that the apoptotic rate in this carcinoma is low. the higher expression of the anti-apoptotic protein bcl-2 suggests a higher preservation of the tumoral cells.
Developing Concrete Research Proposals and Facilitating Self-Directed Learning via Concept Mapping  [PDF]

Self-direction in learning and writing viable research proposals are considered by higher education institutions as essential skills for graduate students to start their careers as researchers. This is an evidence-based practice study focuses on the use of concept mapping to facilitate self-directed learning and enhance research proposal writing in teacher education. An action mixed methods research design was used in this study with quantitative and qualitative data. Participants were 29 graduate students who were enrolled in a core course aimed to provide learners with an in-depth understanding of research methods. All students, at the beginning of the course, were asked to write a research proposal and complete the Self-Directed Learning Readiness Scale (SDLRS). They then were given training in the use of concept mapping technique throughout the course to develop research proposals. Students’ scores prior to and after the intervention were compared. Results indicate that students developed significantly more concrete research proposals, and displayed higher scores at post-intervention assessment. Findings of this study value and support the use of concept mapping to provide students with a comprehensive understanding of the knowledge of their area of study as they reflect on every element of their proposals.

Teachers’ Knowledge and Misconceptions of Attention Deficit/Hyperactivity Disorder  [PDF]

Teachers can play a key role in identifying and supporting students with Attention Deficit/Hyperactivity Disorder (ADHD). In order to fulfill this important role, it is imperative for teachers to have explicit knowledge about ADHD. The overall aim of this study is to investigate teachers’ knowledge and misconceptions of ADHD. Four hundred and twenty-nine (429) teachers participated. The Knowledge of Attention Deficit Disorder Scale (KADDS) along with a demographic questionnaire was used as the survey instruments to collect data. Descriptive statistics and correlation test were used to analyze the data. Results indicated that teachers’ knowledge of ADHD was insufficient. Teachers’ level of knowledge of ADHD was positively related to their prior training and experience with ADHD (i.e., the number of ADHD courses taken in college or graduate level, and the number of workshops pertaining to ADHD). Teachers’ level of knowledge of ADHD also correlated positively with their level of confidence in teaching a student with ADHD. In spite of a few limitations, the results of this study are valuable for identifying areas where there is a misperception or lack of knowledge among teachers.

Identification and Quantification of Microbial Contaminations Present in Herbal Medicines Commonly Consumed by Women in Riyadh, Saudi Arabia  [PDF]

The present study aimed to investigate the microbial populations such a bacteria and fungi contamination present in the herbal medicinal preparations (Lepidium sativum, Nigella sativa, Cuminum cyminum, Foeniculum vulgare, Pimpinella anisum, Trigoneela foenum-graecum, Cinnamomum verum, peel coffee, Alchemilla valgaris, Vitex agnus-castus) commonly available in different parts of Riyadh, Saudi Arabia. To determine the contamination of the herbal products, experiments such as total aerobic bacterial plate counts were evaluated by various plating techniques. The isolation and identification of fungi were done using czapek dox agar and potato dextrose agar. The results showed that a total of herbal remedies were contaminated with bacteria and fungi (100%). Among the contaminated products, peel of coffee and Vitex angnus-castus were noted more contamination than the other herbal medicinal plants. The results confirmed that the traditionally prepared herbal medications in Riyadh city are likely to be contaminated with a wide variety of potentially pathogenic bacteria and fungi. Therefore, before consumption, the quality assurance of these products should be thoroughly enforced and monitored in the production and distribution of herbal preparations. In conclusion, the present study gives proper evidence that the consumed herbal products contain different levels of pathogenic microbes.
Multilocus Sequence Typing of the Guangdong Isolates of Riemerella anatipestifer from Ducks in China  [PDF]

Abstract: It is a very important significance to explore the bacterial typing methods rapidly, accurately and understand the genetic relation between isolates for controlling effectively the disease and preventing the dissemination and diffusion of the pathogen. The changes in the genetic material nucleotide sequences result in variation and evolution of bacteria, the development of molecular biology and genomics make typing of bacteria from phenotype to molecular typing. Riemerella anatipestifer is the causative agent of polyserositis of ducks and geese. We studied 54 isolates of Riemerella anatipestifer from Guangdong in China by multicolor sequence typing (MLST). The result showed that 54 isolates of Riemerella anatipestifer were divided into 14 STs, and among E3, b12xiao and Cb1 three isolates have with independent of type STs. It was found that there was high homology of RA and low genetic variability in the same area. The synonymous mutation and non synonymous mutation rate: dN/dS in seven housekeeping genes was lower than 0.25. Cluster analysis showed that the 54 isolates were clustered into 5 groups. eBRUST analysis showed that all isolates were clustered into one Group. It also proved that the genetic relationship was very closed in duck plague pathogen in this area; the source may be the same ancestors.
A Practical and Automated Hall Magnetometer for Characterization of Magnetic Materials  [PDF]

Abstract: A versatile Hall magnetometer has been developed, manufactured, calibrated, and turned operational for measurements of the magnetic properties of bulk materials and magnetic micro- and nanoparticles. The magnetometer was constructed from the combination of various equipments, which was usually available in most laboratories, such as a Hall effect sensor, an electromagnet, a current source, and a linear actuator. The achieved sensitivity to the magnetic moment was approximately 10-8 Am2. The results were compared to measurements performed with commercial vibrating-sample magnetometers and superconductor quantum interference devices (SQUID) and showed errors of around 1.7% and a standard deviation of 1.2% in relation to measures themselves. The constructed Hall magnetometer records a magnetic hysteresis loop of up to 1.2 T at room temperature. This magnetometer is cost-effective, versatile, and suitable for research.

Abstract: purpose: to demonstrate the economical impact of surgical castration in comparison to the medical castration for patients with advanced prostate cancer. material and methods: between january 2001 and december 2001, 32 patients with advanced prostate cancer underwent bilateral sub-capsular orchiectomy at our hospital. the costs of this procedure were compared to the costs of medical castration with lh-rh analogues. results: the costs of the surgical procedure were extremely reduced when compared to published data on the medical treatment. surgical castration did not have any stronger negative impact on the evolution of these patients when compared to medical castration. conclusion: surgical castration is an efficient and low cost treatment for advanced prostate cancer.

Abstract: Research questions. How does a virtual bronchoscopy navigation system (VBNS) improve prediction of candidate bronchus across a range of doctors investigating a range of lesions with Endobronchial ultrasound (EBUS) guide sheath? To what extent do benefits of virtual bronchoscopic pre-procedure navigation apply to experienced versus inexperienced bron- choscopists? Methods: Using archived EBUS Guide sheath cases, a comparison was made between identified candidate 4th order bronchus by Computerised tomography (CT) evaluation versus that identified after virtual path creation. Results: From 7 archived cases, 14 doctors identified the correct bronchus in 94 of 98 assessments (95%). Percentage of cases where there was an improvement in localisation by 2 or more 4th order bronchi was 39.8% overall (28.6% – 51.0%), 26.6 for experienced and 53.1 for inexperienced bronchoscopists (p < 0.02). The absolute mean number of 4th order bronchi different between CT and VBNS was 2.0 ± 2.6 overall, 1.2 (range 0-6) for experienced, and 2.8 (range 0-11) for inexperienced bronchoscopists. Virtual Path software calculation time was 8.1 ± 2.7 minutes, compared to 3.6 ± 2.1 minutes by CT. Conclusion: VBNS allowed rapid accurate assessment with minimal software training. Greatest benefits in reduction of procedure time were obtained in inexperienced bronchoscopists, and VBNS could allow more rapid skill development in EBUS GS in these doctors.

Abstract: Background: Insulin resistance is linked to dyslipidemia, characterized by a decrease in high density lipo-proteins and an increase in low density lipoproteins. Thiazolidinediones (TZDs) are insulin-sensitizing agents used to improve glycemic control in patients with type 2 diabetes. Recently, the safety of certain TZD regimens has been questioned because of associated adverse effects on the plasma lipid profile. We examined the effect of a TZD, Ciglitazone, on apolipoprotein synthesis and secretion in human liver HepG2 cells. Methods and Results: The effect of Ciglitazone treatment on apolipoprotein synthesis was addressed at the level of transcription, translation and secretion. RT-PCR showed that Ciglitazone increased the transcription of apoE and apoAI but reduced the levels of apoCI and apoB mRNA. Western blot analysis showed an increase in apoAI and apoE secreted in the cell culture media, whereas the amounts of apoB100 and apoCI were reduced. To confirm that Ciglitazone regulates apolipoprotein translation, its effect on de novo protein synthesis was evaluated by metabolic labeling with [35S]-methionine/cysteine, and a similar pattern of regulation was observed. The change in apolipoprotein levels was not secondary to cholesterol biosynthesis or clearance, since Ciglita-zone did not regulate the transcription of HMGCoA reductase, or the LDL receptor. However, mRNA levels for both PPAR-γ and LXRα were induced, suggesting a role for either or both receptors in modulating the hepatic apolipoprotein profile. The involvement of these nuclear receptor transcription factors was confirmed since direct activation of these receptors by endogenous PPAR-γ ligand, 15d-prostaglandin J2, or LXRα ligand, 22(R)hydroxycholesterol, similarly upregulated apoAI and apoE, but down-regulated apoB100 protein synthesis. Conclusion: Our results suggest that Ciglita-zone treatment results in an atheroprotective lipoprotein profile in liver cells. Thus, while the adipose and muscle tissues may be primary targets in TZD-mediated glucose homeostasis, liver PPAR-γ contributes significantly to the regulation of plasma lipoprotein profile.
