Biophysical Mechanism of the SAHA Inhibition of Zn2+-Histone Deacetylase-Like Protein (FB188 HDAH) Assessed via Crystal Structure Analysis  [PDF]

Abstract: The zinc-containing enzyme HDAC-like amidohydrolase (FB188 HDAH), identified in the Bordetella alcaligenes bacteria, is similar to enzymes that participate in epigenetic mechanisms such as histone modifications. The X-ray crystal structure of FB188 HDAH complexed with the antagonist SAHA (suberoylanilide hydroxamic acid) has been solved (PDB ID: 1ZZ1). Notably, the complex crystallizes as a tetramer in the asymmetric unit cell of the crystal. The crystal yielded a suitable structure to analyze the dynamics of the inhibitory mechanism of SAHA on this histone deacetylase. Applying computational chemistry techniques and quantum mechanics theory, several physicochemical properties were calculated to compare the active site of the enzyme of the four monomers. Significant differences were observed in the areas and volumes of the binding pocket, as well as hydrophobic interactions, dipole moments, atomic charges and electrostatic potential, among other properties. Remarkably, a free-energy curve resulting from the evaluation of the energies of SAHA and the interacting amino acids of the four crystal monomers unveiled the biophysical mechanism of the FB188 HDAH inhibition exerted by SAHA to a greater extent. The biophysical mechanism of SAHA inhibition on FB188 deacetylase was clearly observed as a dynamic process. It is possible to define the physicochemical dynamics of the molecular complex by the application of computational chemistry techniques and quantum mechanics theory by studying the crystal structures of the interacting molecules.
Maternal Olfactory Cues Synchronize the Circadian System of Artificially Raised Newborn Rabbits

Abstract: In European newborn rabbits, once-daily nursing acts as a strong non-photic entraining cue for the pre-visual circadian system. Nevertheless, there is a lack of information regarding which of the non-photic cues are capable of modulating pup circadian system. In this study, for the first time, we determined that the mammary pheromone 2-methylbut-2-enal (2MB2) presented in the maternal milk acts as a non-photic entraining cue. We evaluated the effect of once-daily exposure to maternal olfactory cues on the temporal pattern of core body temperature, gross locomotor activity and metabolic variables (liver weight, serum glucose, triacylglycerides, free fatty acids, cholecystokinin and cholesterol levels) in newborn rabbits. Rabbit pups were separated from their mothers from postnatal day 1 (P1) to P8 and were randomly assigned to one of the following conditions: nursed by a lactating doe (NAT); exposed to a 3-min pulse of maternal milk (M-Milk), mammary pheromone (2MB2), or water (H2O). To eliminate maternal stimulation, the pups of the last three groups were artificially fed once every 24-h. On P8, the rabbits were sacrificed at different times of the day. In temperature and activity, the NAT, M-Milk and 2MB2 groups exhibited clear diurnal rhythmicity with a conspicuous anticipatory rise hours prior to nursing. In contrast, the H2O group exhibited atypical rhythmicity in both parameters, lacking the anticipatory component. At the metabolic level, all of the groups exhibited a diurnal pattern with similar phases in liver weight and metabolites examined. The results obtained in this study suggest that during pre-visual stages of development, the circadian system of newborn rabbits is sensitive to the maternal olfactory cues contained in milk, indicating that these cues function as non-photic entraining signals mainly for the central oscillators regulating the expression of temperature and behavior, whereas in metabolic diurnal rhythmicity, these cues lack an effect, indicating that peripheral oscillators respond to milk administration.

Abstract:

The impacts of excess Body mass index (BMI) on physical health are well known and widely studied, but little is known about the consequences of being overweight or obese on cognitive function. The aim of this study was to correlate the BMI with executive functions in overweight middle- aged women. Seventy overweight middle-aged women between 48 and 64 years of age who are otherwise healthy participated in the study. Demographic and anthropometric variables were evaluated. Four standard neuropsychological tests were applied to assess executive functions, sustained attention, selective attention, and verbal fluency. The mean BMI of the participants was 29.35 and was negatively correlated with categories reached (r = -0.41, p = 0.007) and positively correlated with the number of errors (r = 0.43, p = 0.005) of executive function test. Scores of other neuropsychological tests applied showed no correlation with BMI. These findings suggest that BMI impacts the executive functions in overweight women who do not present any signs of disease.

Abstract: the flora of four fog oases in the northern chilean coastal desert, visited between october 1997 and january 1998, is described. these botanical collections are new for alto patache, punta lobos and alto chipana. a total of 72 vascular plant species were registered, contributing to the area′s floristic knowledge, where till now only about 20 species were registered. three species are new records for the chilean flora: nolana adansonii (roem. et schult.) johnst., solanum cfr. montanum l. and ophryosporus cfr. floribundus (d.c.) king et rob., and another one alstroemeria lutea (mu?oz 2000) is a new species. this last one, together with eulychnia aricensis ritter, pyrrhocactus saxifragus ritter (both cactaceae) and nolana intonsa johnst. (nolanaceae) are local endemics. of the total of registered species, 43 % are endemics, 52.7 % are non endemic natives and a low percentage (4.1 %) are foreign species; 47.2 % share their distribution with perú. the families asteraceae, nolanaceae and solanaceae, are the best represented in the area, each one with more than seven species. information about special climatic conditions due to el ni?o 1997-1998 event is discussed in relationship to the development of the flora and vegetation. in spite of a non significant occurrence of rainfall events, an increase in temperature and in the fog's water content is detected. these conditions favor the development of more exuberant vegetation and the expansion of the distribution range of some floristic elements

Abstract: Se describe la flora de cuatro sitios de "oasis de neblina" del desierto costero del Norte de Chile, entre Iquique y la desembocadura del río Loa, muestreados entre los meses de octubre de 1997 y enero de 1998. No existían muestreos anteriores en tres de los sitios: Alto Patache, Punta Lobos y Alto Chipana. Se registraron en esta ocasión 72 especies de plantas vasculares, lo que amplía notablemente el conocimiento florístico de este sector, que poseía a la fecha apenas una veintena de especies registradas. De las especies recolectadas, tres constituyen nuevos registros para la flora chilena: Nolana adansonii (Roem. et Schult.) Johnst., Solanum cfr. montanum L. y Ophryosporus cfr. floribundus (DC.) King et Rob., mientras que Alstroemeria lutea (Mu oz 2000) constituye una especie nueva para la ciencia. Esta última, junto a Eulychnia aricensis Ritter, Pyrrhocactus saxifragus Ritter y Nolana intonsa Johnst., son endémicas locales. Del total de especies, un 43 % son endémicas y un 52,8 % son nativas no endémicas, existiendo un bajo porcentaje (4,2 %) de especies adventicias; 34 especies (47,2 %) comparten su área de distribución con el Perú. Las familias Asteraceae, Nolanaceae y Solanaceae son las mejor representadas en el área con más de siete especies cada una. Las condiciones climáticas derivadas del evento El Ni o 1997-1998 han favorecido el desarrollo de la vegetación en los oasis de neblina al sur de Iquique. Abstract: measurement of intra-access venous pressure is a vascular monitoring method that facilitates early detection of dysfunctions in arteriovenous fistulas. our aim was to show how we use this detection method and describe our experience in monitoring 31 patients. in 9 patients a one-off increase in intra-access venous pressure was observed, and in 4 patients an effective preventive angioplasty was carried out. in 7 patients who continually showed an increase in intra-access pressure, vascular access was thrombosed and not recoverable by angio radiology. the increase in intra-access pressure can facilitate the early detection of problems in vascular access.

Abstract: La medición de la presión venosa intraacceso es un método de vigilancia vascular que facilita la detección temprana de las disfunciones de las fístulas arteriovenosas. Nuestro objetivo fue describir como utilizamos este método de detección y exponer nuestra experiencia en el seguimiento de 31 pacientes. En 9 pacientes se observó un aumento puntual de la presión venosa intraaceso, y a 4 se les realizó una angioplastia preventiva efectiva. En 7 pacientes que continuadamente presentaban un aumento de la presión intraaceso se trombosó el acceso vascular no siendo recuperable por angiorradiología. El aumento de la presión intraaceso puede facilitar la detección precoz de problemas en el acceso vascular. Abstract: Se sintetizaron siete compuestos como drogas potencialmente anti-inflamatorias, inhibidores selectivos de la enzima ciclooxigenasa- 2 (COX-2). El dise o estructural se basó en el conocimiento del sitio activo de la enzima. Los compuestos sintetizados presentan estructuras combinadas de naproxen y celecoxib.
