A Comprehensive In Silico Analysis of the Functional and Structural Impact of Nonsynonymous SNPs in the ABCA1 Transporter Gene

Abstract: Disease phenotypes and defects in function can be traced to nonsynonymous single nucleotide polymorphisms (nsSNPs), which are important indicators of action sites and effective potential therapeutic approaches. Identification of deleterious nsSNPs is crucial to characterize the genetic basis of diseases, assess individual susceptibility to disease, determinate molecular and therapeutic targets, and predict clinical phenotypes. In this study using PolyPhen2 and MutPred in silico algorithms, we analyzed the genetic variations that can alter the expression and function of the ABCA1 gene that causes the allelic disorders familial hypoalphalipoproteinemia and Tangier disease. Predictions were validated with published results from in vitro, in vivo, and human studies. Out of a total of 233 nsSNPs, 80 (34.33%) were found deleterious by both methods. Among these 80 deleterious nsSNPs found, 29 (12.44%) rare variants resulted highly deleterious with a probability >0.8. We have observed that mostly variants with verified functional effect in experimental studies are correctly predicted as damage variants by MutPred and PolyPhen2 tools. Still, the controversial results of experimental approaches correspond to nsSNPs predicted as neutral by both methods, or contradictory predictions are obtained for them. A total of seventeen nsSNPs were predicted as deleterious by PolyPhen2, which resulted neutral by MutPred. Otherwise, forty two nsSNPs were predicted as deleterious by MutPred, which resulted neutral by PolyPhen2. 1. Introduction Nonsynonymous single nucleotide polymorphisms (nsSNPs) are single base changes in coding regions that cause an amino acid substitution in the correspondent proteins. These missense variants constitute the most identifiable group of SNPs represented by a small (<1%) proportion [1]. The nsSNPs might alter structure, stability, and function of proteins and produce the least conservative substitutions with drastic phenotypic consequences [2–5]. Studies suggest that about 60% of Mendelian diseases are caused by amino acid exchanges [6]. Thousands of associations between Mendelian and complex diseases reveal a phenotypic code that links each complex disorder to a unique set of Mendelian loci [7]. Discriminating disease-associated from neutral variants would help to understand the genotype/phenotype relation and to develop diagnosis and treatment strategies for diseases. Abstract: Lenny Kaye has been Patti Smith’s long term guitarist, friend and collaborator, ever since they first began together in the early 1970s. He grew up between New York and New Jersey, graduating in American History from Rutgers University, where he later taught a course in the Department of American Studies on the History of American Rock, which became famous because of the large number of students who wanted to enroll in it. A very prolific writer and musician, he has produced an important number of records, as well as collaborated with numerous music magazines. He is the author of two books, Waylon Jennings: An Autobiography (1996) and You Call it Madness, The Sensuous Song of the Croon (2004). Nuggets (1972), his anthology of 60s garage music, is famous for defining the genre. This interview took place when he was visiting Spain in November 2012 with the Patti Smith Group. In it, we discussed the New York scene of the 70s, music, literature, drugs, politics, and many other things.

Abstract: a descriptive and cross-sectional study on mortality due to cancer in santiago de cuba province during 2010 was carried out, taking into account 1 819 deceased at a rate of 173,9 every 100 000 inhabitants. this rate increased compared to the previous year with a male mortality predominance of elderly aged 75 and over, as well as mortality in mella and ii frente municipalities. most common malignancies in women were detected in lungs, breast, and colon; while in men, they were preponderantly diagnosed in prostate, lungs, and colon. for this reason, it is recommendable addressing and planning health actions according to the stratified mortality risk in each municipality of this province.

Abstract: Se realizó un estudio descriptivo y transversal sobre la mortalidad por cáncer en la provincia de Santiago de Cuba durante 2010, dada por 1 819 fallecidos por esa causa, para una tasa de 173,9 por cada 100 000 habitantes; cifra aumentada respecto al a o anterior, con predominio de la sobremortalidad masculina, de los ancianos de 75 y más a os, así como también de los municipios de Mella y II Frente. Los tumores malignos más comunes en las mujeres se localizaron en pulmón, mama y colon, mientras que en los hombres se diagnosticaron preponderantemente en próstata, pulmón y colon, por lo cual se recomienda dirigir y planificar las acciones sanitarias según el riesgo de mortalidad estratificado en cada municipio de esta provincia. Abstract: We presented in this paper results of a research which aimed to investigate how the community of practice context of pre-service mathematics teacher education collaborates for learning on algebraic thinking by these future teachers. We analyzed, taking into account the Social Theory of Learning developed by Wenger (1998) as a theoretical frame, processes of negotiation of meanings present in participants' algebraic thinking in the development of tasks in one of the actions of the project "Mathematical Education of Teachers of Mathematics" inside the program "Universidade sem Fronteiras". This analysis allowed us to define some forms of member participation and explicit reification of algebraic thinking, due to some interactions in the processes of negotiation of meanings, which revealed changes in the identity of participants in become teachers of mathematics.

Abstract:

Cucumber mosaic virus (CMV) has the broadest host range, infecting more than 1300 species in more than 500 genera from over 100 botanical families. In ornamental plants, CMV can cause mosaic and distortion of leaves, stunting, color break, and malformation of flowers. CMV coat protein (CP) sequences obtained from seven ornamental plants and other homologous sequences available in GenBank were compared, and phylogenetic relationships were established. Total RNA from virus-infected ornamental species were extracted, submitted to RT-PCR with specific primers, and amplicons obtained were sequenced. A nucleotide substitution model and phylogenetic analyses were carried out using the PAUP program. The seven sequences of CMV CP obtained showed similar identity percentages and close relationships with subgroup I isolates from other countries and hosts. Abstract: in this work we assume the level of education as an indicator of political elite's quality. our hypothesis is that legislators' education level in some of the latin american countries has not a positive impact on the characteristics of democracy. instead, under social and economic inequalities, a high level of education shows individual or collective privileges and, therefore, it does not have an effect on the general condition of social and political system.

Abstract: injuries of the ischemia and reperfusion (i/r) syndrome in the different pathophysiological states from clinical situations, as well as in surgical procedures of high vascular and tissue manipulation, are of great importance in equine medicine; for that reason, most studies are made in the particularities of the equine specie due to the increase in frequency of presentation in intestinal accidents (white et al. 1980; moore et al. 1995; faleiros, 1997; kooreman et al. 1998; faleiros et al. 2002). in spite of the recent advances in therapeutics and surgical clinic, mortality in equines carrying strangling diseases is elevated, having great importance the intestinal ischemia. similarly an efficient drug for the treatment of the injury by reperfusion in the clinical routine of the acute abdomen in equines is not available (hinnebusch et al. 2002; dabareiner et al. 1998). they either do not know absolutely the class of injuries that a problem in the vascularization could cause in organs highly employees in the vascular provision as heart, lungs, liver and kidney, because it is verified that the damage by reperfusion brings in consequence cellular death and endothelial dysfunction caused by the restoration of the blood flow (alfonso et al. 2000).

Abstract: lupus anticoagulants (la) are immunoglobulins which interfere in in vitro phospholipid-dependent coagulation tests. various methods have been proposed; however, activated partial thromboplastin time (aptt) is the most used screening test for lupus anticoagulant. previous studies have shown that sensitivity to the lupus anticoagulant defect varies considerably with different aptt reagents. in view of the undoubted clinical importance of lupus anticoagulants, the sensitivity of 19 commercial aptt reagents has been evaluated. the study raises several important conclusions: no difference was found in sensitivity associated with a narrower distribution of liposomes' diameter, considering the latter as a marker of uniformity in phospholipid distribution. the source of the platelet substitute and the nature of the contact phase activator are unlikely to determine such varied sensitivity. finally, a significant negative correlation between aptt sensitivity and total phospholipid concentration was found. the lower the phospholipid concentration, the higher the aptt sensitivity to al.
Recurrent pneumonia caused by genetic immunodeficiency: a prophylactic and rehabilitative approach

Abstract: recurrent infections are a consequence of a series of genetic diseases characterized by deficiency in the immunological response. one of these diseases is the agammaglobulinemia, which is characterized by the basic defect in the maturation of lymphocytes b. the carrier of this kind of immunodeficiency, which is linked to the x (xla) chromosome, has had primary pneumonias that have evolved into secondary pneumonias (chronic lungs with sequelae) after the third or fourth year of life. the clinical and rehabilitative quest for prophylaxis against the xla immunodeficiency is accomplished in order to avoid the evolution of the bacterial infection into sequelae and loss of pulmonary function, which propitiates the recurrence of the disease and deteriorates the life quality of the patient. forty cases of recurrent respiratory infections were studied. some of them were associated with primary respiratory diseases without investigation of serum immunoglobulins and some were not. casuistics was performed according to data from medical records with pertinent treatments collected from january 1997 to september 2004 at the specialized physiotherapy center. age average was 2.7 years of life. it is statistically impossible to precise results concerning only the immunosuppressed patients due to the lack of specific diagnosis. that is explained by the fact that recurrent xla pneumonias may be attributed to the gastroesophageal reflux disease or to bronchial asthma. however, the improved results showed by the pulmonary function as preventive strategy were attributed to the respiratory physiotherapy, since intravenous immunoglobulin replacement therapies were not performed. respiratory physiotherapy acts as a supportive factor in the healing process and occupies a fundamental role in the prophylaxis against recurrent respiratory clinical features, especially those of obstructive and secretionary characteristics.
