Abstract: The gradual growth of the flexography market generates different expectations in organizations of this segment. To ensure the fulfillment of the demand and ensure its market share, it is important to know the real productive capacity of its plant. The research aimed to identify the capacity of the sector recording a stereotype workshop the city of Blumenau/SC. Was defined as qualitative research, objective descriptive and as in the investigation, the research is characterized as action research. Data were collected from the daily monitoring of the processes performed in the recording industry, observing the development of activities, recording information in reports and informal interviews with employees. Thus, the data were triangulated and presented in tables and figures. The results of this investigation was identified the bottleneck of production as well as the existence of hours shutdowns for lack of service, and made suggestions to optimize production capacity, as well as strategies to increase earnings. O crescimento gradativo do mercado de flexografia gera diversas expectativas nas organiza es deste segmento. Para garantir o atendimento da demanda e garantir a sua fatia de mercado, é importante conhecer a real capacidade produtiva da sua unidade de produ o. A pesquisa teve como objetivo identificar a capacidade de produ o do setor de grava o de uma clicheria da cidade de Blumenau/SC. Foi definida como pesquisa qualitativa, de objetivo descritivo e quanto aos procedimentos de investiga o, a pesquisa se caracterizou como pesquisa-a o. Os dados foram coletados a partir do acompanhamento diário dos processos realizados no setor de grava o, observa o do desenvolvimento das atividades, registro das informa es em relatórios e entrevistas informais com os funcionários. Dessa forma, os dados obtidos foram triangulados e apresentados em figuras e tabelas. Como resultados desta investiga o, foi identificado o gargalo de produ o, bem como a existência de horas paradas programadas por falta de servi o, sendo feitas sugest es para otimizar a capacidade produtiva, assim como estratégias para aumentar os ganhos.
Turbulent Flowfield Analysis in a Bluff-Body Burner Using PIV  [PDF]

The structure of inert turbulent flows, stabilized in a Bluff-Body burner, is studied considering different volumetric flows for Nitrogen jet and annular air in coflow configuration. Flowfield analysis on Bluff-Body burner is essential to improve the knowledge about this burner, which plays an important role in industrial applications. Thus, vector velocity field is performed, employing Particle Image Velocimetry technique. Also, an uncertainty analysis is performed considering parameters involved in this technique yielding 6% to velocity measurements. The acquired information produces the results based in flowfield structure, which are presented in terms of statistical momentum and Reynolds stress, in which Boussinesq Hypothesis is considered to incompressible flows. However, this hypothesis fails in certain conditions. In this way, is possible to comprehend and provide experimental data from the turbulent effects on the flowfield and also contribute to predict the combustion flows, in order to enable the validation and develop numerical models.

Optimized Protein Extraction for Quantitative Proteomics of Yeasts

Abstract: Background The absolute quantification of intracellular protein levels is technically demanding, but has recently become more prominent because novel approaches like systems biology and metabolic control analysis require knowledge of these parameters. Current protocols for the extraction of proteins from yeast cells are likely to introduce artifacts into quantification procedures because of incomplete or selective extraction. Principal Findings We have developed a novel procedure for protein extraction from S. cerevisiae based on chemical lysis and simultaneous solubilization in SDS and urea, which can extract the great majority of proteins to apparent completeness. The procedure can be used for different Saccharomycetes yeast species and varying growth conditions, is suitable for high-throughput extraction in a 96-well format, and the resulting extracts can easily be post-processed for use in non-SDS compatible procedures like 2D gel electrophoresis. Conclusions An improved method for quantitative protein extraction has been developed that removes some of the sources of artefacts in quantitative proteomics experiments, while at the same time allowing novel types of applications.
A quantitative estimation of the global translational activity in logarithmically growing yeast cells

Abstract: An examination of the accuracy of genome-wide expression datasets generated for Saccharomyces cerevisiae shows that the available datasets suffer from large random errors within studies as well as systematic shifts in reported values between studies, which make predictions of translational activity at the level of individual genes relatively inaccurate. In contrast, predictions of cell-wide translational activity are possible from such datasets with higher accuracy, and current datasets predict a production rate of about 13,000 proteins per haploid cell per second under fast growth conditions. This prediction is shown to be consistent with independently derived kinetic information on nucleotide exchange reactions that occur during translation, and on the ribosomal content of yeast cells.This study highlights some of the limitations in published genome-wide expression datasets, but also demonstrates a novel use for such datasets in examining global properties of cells. The global translational activity of yeast cells predicted in this study is a useful benchmark against which biochemical data on individual translation factor activities can be interpreted.The rate of translation of eukaryotic messenger RNAs is often stringently controlled and can limit the overall amount of protein produced from a gene. Moreover, changes in mRNA-specific translation rates can occur, and such changes can alter protein production rates independently of transcriptional activity. Translational control mechanisms of this kind are known to contribute to cellular growth and proliferation, the development of organisms, and to the occurrence of disease [1].Mechanisms by which the translational apparatus can exert control over gene expression rates are being described in increasing detail. Abstract: in his encounter with don quijote the canonigo of toledo tries to convince him that the libros de caballerias have done him wrong and he uses arguments taken from neoaristotelic poetics. don quijote, on the other hand tries to show with his own example that this kind of books do not lead to madness or to abandon one self, but rather bring forth the best of an individual’s nature and makes him a better person. this is the only type of imitation that plato accepts in the republic, since more than a substitution of another it implies emulation. this paper seeks to show, having as a starting point the dialogue with the canonigo (i, 49-50), how in his mimetic conducts don quijote understands imitation as emulation; and it tries to prove that the effect of literature on don quijote seems to be the type of mimesis that plato approves in book iii of the republic.

Abstract: Resumen:Frente a argumentos tomados de las poéticas neoaristotélicas que esgrime el canónigo para condenar los libros de caballerías, don Quijote pretende mostrar con su propio ejemplo, que ese tipo de lecturas no llevan a la locura ni al abandono de sí mismo, sino que, por el contrario, sacan lo mejor de la naturaleza de un individuo y lo hacen mejor persona. Este es el único tipo de imitación que Platón acepta en la República, pues más que un remedo o una suplantación, implica emulación. Este escrito se propone mirar, a partir del diálogo con el canónigo de Toledo (I, 49-50), cómo en sus conductas miméticas desde el comienzo de la obra, don Quijote entiende la imitación como emulación; en consecuencia se intenta demostrar que el efecto de la literatura en el caballero manchego parece ser el tipo de mimesis que Platón acepta en el libro III de la República.Abstract:Don Quijote demonstrates with the example of his own life the error made by the Canon of Toledo when the latter uses the arguments of the neo-Aristotelian poets to condemn the novels of chivalric romance. Quijote tries to show that reading the chivalric romances leads to neither madness nor the abandonment of the self, but, on the contrary, brings out the best in a person’s nature, and makes one a better person. This is the only type of imitation that Plato accepts in the Republic, because, unlike the mere copying of a person, or the substitution of one person for another, it implies a type of emulation. Starting with the dialogue of the Canon of Toledo (I, 49-50), this paper shows how, in his mimetic behavior, Don Quijote understands imitation as emulation. As a result, the paper shows that the effect of literature on the gentleman from La Mancha is the type of mimesis of which Plato approves in book III of the Republic.

Reality Can Bite: Perspectives of Young People on the Role of Religion in Their World

Abstract: Despite the vast research on young people and identity issues in recent years, little is still known about the role of religion in adolescents’ lives across various ethnic, religious, and belief-related scenarios. The overall sense is that young people in general do not care about religion, and religion is of little or no importance in their daily lives. This article contributes to that discussion through extensive qualitative interviews with students from different ethnic, religious, and non-religious backgrounds. An analysis of these interviews yields explanations that complicate and sometimes even challenge widely held assumptions about young people and religion, and add new information and nuances to the general understanding of the interplay between ethnicity, religion, and skin colour. This study is part of a larger European project on young people and religion in education (REDCo). Focusing on students aged 14-16, and their discursive constructions on religion and diversity, the findings also contribute important knowledge to school in general, and for religious education teachers in particular.
