Precise identification of photosynthetic glycerolipids in microalga Tetraselmis chuii by UPLC-ESI-Q-TOF-MS

Abstract: Precise structural identification of photosynthetic polar glycerolipids in microalga Tetraselmis chuii has been established using Ultra Performance Liquid Chromatography-Electrospray ionization-Quadrupole-Time of Flight Mass Spectrometry (UPLC-ESI-Q-TOF-MS) by direct analysis of the total lipids extract. The mass spectrometry was performed in reflective time-of-flight using electron spraying ionization in both positive and negative modes. The structural determination was based on the characteristic product ions yielded by different glycerolipids under ESI-MS/MS mode, and confirmed the molecular species by the carboxylate anions produced by glycerolipids in the negative mode. As a result, more than 40 lipid molecular species, including 11 monogalactosyldiacylglycerols (MGDG), 7 digalactosyldiacylglycerols (DGDG), 16 sulfoquinovosyldiacylglycerols (SQDG), and 9 phosphatidylglycerols (PG), were detected in Tetraselmis chuii, which had never been identified before in this microalga. Furthermore, some intact lipid molecules with hydroxylated fatty acids that could not be detected by the traditional GC-MS method were found this time, providing novel information for the photosynthetic lipidome of Tetraselmis chuii. Comparative studies on fatty acids at the sn-2 position showed that SQDG and MGDG are dominantly biosynthesized through the prokaryotic pathway, PG is a typically mixed biosynthetic pathway, while DGDG is somewhat peculiar with C14:0 and C16:0 at its sn-2 position. This method could provide a full structural profile of intact photosynthetic lipid molecular species, which may be applied to study the physiological and ecological functions of lipid by monitoring their individual changes.

Predictive Encoding of Contextual Relationships for Perceptual Inference, Interpolation and Prediction

Abstract: We propose a new neurally-inspired model that can learn to encode the global relationship context of visual events across time and space and to use the contextual information to modulate the analysis by synthesis process in a predictive coding framework. The model learns latent contextual representations by maximizing the predictability of visual events based on local and global contextual information through both top-down and bottom-up processes. In contrast to standard predictive coding models, the prediction error in this model is used to update the contextual representation but does not alter the feedforward input for the next layer, and is thus more consistent with neurophysiological observations. We establish the computational feasibility of this model by demonstrating its ability in several aspects. We show that our model can outperform state-of-art performances of gated Boltzmann machines (GBM) in estimation of contextual information. Our model can also interpolate missing events or predict future events in image sequences while simultaneously estimating contextual information. We show it achieves state-of-art performances in terms of prediction accuracy in a variety of tasks and possesses the ability to interpolate missing frames, a function that is lacking in GBM.

Abstract: 为了解斜纹夜蛾Spodoptera litura(Fabricius)配对后雌、雄成虫日活动行为节律,在室内人工气候室中配对饲养斜纹夜蛾,连续6 d观察成虫移动、静息、补充营养、产卵和交配5个行为类型。结果表明,在配对后6 d内,斜纹夜蛾的交配、产卵、移动、补充营养和停息行为所占比例均有明显变化,交配行为主要发生在配对后的前2 d,在配对后第6 天成虫除偶尔补充营养外基本处于停息状态;交配行为有2个明显高峰期,即00:00~01:00(3.89%)和04:00~05:00(3.89%),雌、雄成虫移动行为日节律所占比例在03:00~06:00和补充营养行为日节律在03:00~05:00差异显著;成虫各行为分配频次最多的是停息行为,远高于其它行为,且各行为所占比例差异极显著,但雌、雄成虫之间显著差异;雌、雄成虫运动行为几乎全部发生在暗期,且在不同时间段显著差异;光、暗期各行为活动发生的比率差异显著;在暗期不同时间段,除雌蛾的移动行为、雄蛾的补充营养行为和交配行为显著差异外,其它各种行为均差异显著。表明斜纹夜蛾雌、雄配对后移动、静息、补充营养、产卵和交配行为受成虫日龄影响较大,也受性别影响,表现出明显的日节律性。
To study the diurnal behavioral rhythm and time budgets of Spodoptera litura (Fabricius) adults after paired, the adult behaviors, including moving (MV), resting (RT), mating (MT), supplementary nutrition (SN) and oviposition (OP), were observed for six consecutive days in artificial climatic chambers. The results showed that all behaviors of S. litura adults changed apparently in six consecutive days. MT-behavior occurred mainly in the 1st and 2nd day after paired. All were nearly RT in the 6th day except occasional SN. Two distinct MT behavioral peaks existed at 00:00-01:00 and 04:00-05:00. The rates of MV-behavior at 03:00-06:00 and SN-behavior at 03:00-05:00 were significantly different between females and males. RT-behavior accounted for the largest proportion and was far more than other behaviors. There was a significant difference in the diurnal time budget of adult behaviors, but no difference was observed between females and males. No significant difference was found in the motional behavior occurring mostly in scotophase between females and males in different periods. The proportion of all behaviors was significantly different between scotophase and photophase, not significantly different in different periods of scotophase except for MV-behaviors of females and MT- and SN-behavior of males. This study indicated that all behaviors of S. litura after paired were affected by adult age, and behavior rhythm was obvious and affected by gender.

Abstract: 为摸清斜纹夜蛾成虫在不同寄主上的行为节律变化, 在室内分别用烟草和白菜饲养3代, 建立烟草种群和白菜种群, 比较分析了成虫的羽化、交配、产卵等行为节律。结果表明:烟草种群雌虫有2个羽化高峰期, 分别在熄灯前2 h和熄灯后1 h, 雄虫只有1个高峰期, 出现在熄灯后2 h;白菜种群雌、雄虫均有2个高峰期, 主、次高峰期分别在熄灯前后1 h内和熄灯后4~5 h。2种群均有2个交配高峰期, 烟草种群第1高峰期在熄灯后2 h, 而白菜种群在熄灯后5 h, 第2高峰期均在开灯前2 h;烟草种群和白菜种群一生的交配持续时间分别为54.78±3.09 min和60.00±2.58 min, 差异不显著;烟草种群一生交配1~3次, 而白菜种群只交配1次, 差异极显著;2种群均只有1个产卵高峰期, 烟草种群在熄灯后4 h, 白菜种群则在熄灯后5 h。表明斜纹夜蛾取食烟草和白菜2种不同寄主其成虫繁殖节律会发生较大的改变。
In order to find out the differences in the behavioral rhythm of Spodoptera litura adults on different host plants, diurnal rhythms of adult emergence, mating and ovulating between tobacco population and cabbage population were carefully researched for three generations indoors. The results showed that the diurnal rhythm of two host populations were different. Female adults of tobacco population had two emergence peaks: one appeared two hours before lights out and the other one hour after lights out; males had only one peak that appeared two hour after lights out. Both male and female adults of cabbage population had two emergence peaks: the main peak appeared one hour before and after lights out and the subordinate peak appeared between four and five hours after lights out. Mating and ovulating behaviors of both populations appeared at night. Both populations had two mating peaks: the first peak of tobacco population appeared two hours after lights out, while the cabbage population appeared five hours after lights out; the second appeared two hours before lights on. Two populations had only one ovulating peak: the tobacco population appeared four hours after lights out, while the cabbage population appeared five hours after lights out. The lifetime mating duration of the tobacco population and cabbage population were 54.78±3.09 min and 60±2.58 min, respectively, and they were not significantly different. Tobacco population mated 1-3 times, but cabbage population mated only one time. The results indicated that the reproductive diurnal rhythm of S. litura varied after they fed on tobacco and cabbage.

Abstract: Developing large- and intermediate-scale biogas project is an important gripper ofBeijingrural energy construction and “Green Beijing” construction. The existing projects have made obvious energy and environment benefits, but the overall effects have not been fully exerted. There is still a large gap betweenBeijing’s ecological civilization goal and the operation effect. This paper takes biogas projects ofBeijingseven mountainous counties as examples, the development status, ecological environment construction and existing problems of operating are explained respectively. Finally, some countermeasures and suggestions are proposed for promoting the sustainable development of biogas project inBeijing, such as strengthening technical innovation and setting standards, innovating development patterns and planning reasonable layout, improve the maintenance mechanism and intensify policy support.

Abstract: Ethanol fermentation from Jerusalem artichoke tubers by recombinant Saccharomyces cerevisiae strains expressing the inulinase gene (inu) from Kluyveromyces marxianus was investigated. The inu native and pgk promoters were used to drive the expression of the inu gene, and the inulinase was expressed as an extracellular enzyme. All positive clones (confirmed by PCR) were able to express inulinase as measured by enzyme activity in the culture supernatant, among which two clones HI6/6 and HPI6/3 were selected, and their inulinase activity and ethanol fermentation performance were compared with their wild type. The inulinase activities of 86 and 23.8 U/mL were achieved, which were 4.6-fold and 1.5-fold higher than that of the wild type. Furthermore, ethanol fermentation was carried out with the recombinants and medium containing 200 g/L raw Jerusalem artichoke meal, and ethanol concentrations of 55 g/L and 52 g/L were obtained, with ethanol yields of 0.495 and 0.453, respectively, equivalent to 96.9% and 88.6% of the theoretical value.

Abstract: Traditional ECG acquisition system lacks for flexibility. To improve the flexibility of ECG acquisition system and the signal-to-noise ratio of ECG, a new ECG acquisition system was designed based on DAQ card and Labview and oversampling was implemented in Labview. And analog signal conditioning circuit was improved on. The result indicated that the system could detect ECG signal accurately with high signal-to-noise ratio and the signal processing methods could be adjusted easily. So the new system can satisfy many kinds of ECG acquisition. It is a flexible experiment platform for exploring new ECG acquisition methods.
