In vitro and in vivo toxicity studies of Agrobacterium radiobacter k84 biopolymer (ARB) Estudos in vitro e in vivo de toxicidade de biopolímero de Agrobacterium radiobacter k84 (ARB)

Abstract: Sugar cane molasses is a cheaper carbon source alternative than glucose traditionally used in fermentation processes. In the present study a biopolymer soluble from Agrobacterium radiobacter k84 (ARB) was obtained by fermentation using sugar cane molasses as a carbon source in a process with yield of 10.0 g.L-1. The ARB is composed by minerals (40%), carbohydrate (35%) and protein (15%). In vitro test of the cytotoxic effect of ARB at concentrations 2.5 mg/mL, 5.0 mg/mL and 10.0 mg/mL in LLC MK2 (Rhesus Monkey Kidney) cells revealed a 50% cytotoxic concentration (CC50) of 9.32 mg/mL. In a 30-day in vivo oral toxicity study, Swiss mice were administered ARB by gavage at 5 mg/mL, 15 mg/mL, 50 mg/mL and 150 mg/mL (approximately 25 mg/kg/day, 75 mg/kg/day, 250 mg/kg/day and 750 mg/kg/day). The results did not present any hematological or histopathological signs of adverse effects, leading us to define the no observed adverse effect level (NOAEL) as 150 mg/mL (approximately 750 mg/kg/day). O mela o de cana-de-a úcar é uma fonte de carbono alternativa de menor custo que a glicose tradicionalmente utilizada em processos fermentativos. No presente estudo, um biopolímero solúvel de Agrobacterium radiobacter k84 (ARB) foi obtido por fermenta o utilizando mela o de cana de a úcar como fonte de carbono em um processo com rendimento de 10,0 g.L-1. O ARB é composto de minerais (40%), carboidratos (35%) e proteínas (15%). O teste do efeito citotóxico do ARB in vitro nas concentra es de 2,5 mg/mL, 5,0 mg/mL e 10,0 mg/mL em células LLC MK2 (Rim de Macaco Rhesus) revelou uma concentra o citotóxica 50% (CC50) de 9,32 mg/mL. Em estudo in vivo de toxicidade oral durante 30 dias, camundongos Swiss receberam por gavagem solu es de ARB nas concentra es de 5 mg/mL, 15 mg/mL, 50 mg/mL e 150 mg/mL (aproximadamente 25 mg/kg/dia, 75 mg/kg/dia, 250 mg/ kg/dia e 750 mg/kg/dia). Os resultados n o apresentaram sinais hematológicos ou histopatológicos de efeitos adversos, levando a definir a dose sem efeito adverso observado (NOAEL) como 150 mg/mL (aproximadamente 750 mg/kg/dia).

Abstract: In this study, a biopolymer composed of carbohydrates (35%), protein (15%) and minerals (40%) was obtained through fermentation using sugar cane molasses as the sole carbon source for Agrobacterium radiobacter k84. The process yield was 10 gL-1 of biopolymer, which showed high solubility in water, neutral pH in aqueous solution and low water activity (0.52). The analysis in Scanning Electron Microscopy revealed microstructure characteristic of an amorphous solid, with particles of irregular shapes and sizes. In the evaluation of technological properties, the biopolymer showed formation of viscous solutions at room temperature from concentration of 0.5% in aqueous solution, gelling activity in solution at 2%, emulsifying (56.11±1.39%) and stabilizing activity (98.02±0.78%). The results suggest that the biopolymer from Agrobacterium radiobacter k84 is a promising candidate for industrial use. No presente estudo, utilizando mela o de cana-de-a úcar como única fonte de carbono para Agrobacterium radiobacter k84 foi obtido, em processo fermentativo, um biopolímero composto por carboidratos (35%), proteínas (15%) e minerais (40%). O rendimento do processo foi de 10 g.L-1 do biopolímero que apresentou elevada solubilidade em água, pH neutro em solu o aquosa e baixa atividade de água (0.52). As análises em Microscopia Eletr nica de Varredura revelaram microestrutura característica de um sólido amorfo, com partículas de formas irregulares e tamanhos variáveis. Na avalia o das propriedades tecnológicas, o biopolímero mostrou viscosidade à temperatura ambiente a partir da concentra o 0.5% em solu o aquosa, atividade geleificante em solu o a 2%, atividade emulsificante (56.11±0.78%) e estabilizante (98.02±1.39%). Os resultados sugerem o biopolímero de Agrobacterium radiobacter k84 como um candidato promissor para uso industrial.
Avalia??o da mutagenicidade e antimutagenicidade de um biopolímero extraído do microorganismo Agrobacterium radiobacter em camundongos Swiss

Abstract: this study evaluated the mutagenic and ant mutagenic action of a biopolymer of glucose extracted from agrobacterium radiobacter (biopolymer of agrobacterium radiobacter). the experiment was conducted with swiss male mice divided into eight groups. treatment with the biopolymer was performed in a single dose by gavage at a dose of concomitant phosphate buffer groups in the evaluation of mutagenicity, or the agent of inducing dna damage, cyclophosphamide, the concentration of 50 mg/kg (body weight --b.w.), in groups of assessment ant mutagenic. we used the micronucleus test in peripheral blood. the blood sample was held 24 and 48 h after application of the test substances. statistical analysis showed that the biopolymer has no mutagenic activity and it is effective in preventing damage to dna. the percentages of damage reduction in groups of ant mutagenic were 83.9%, 89.1% and 103.1% in 24 h and 101.24%, 98.14% and 120.64% at doses of 48 to 75, 150 and 300 mg/kg (b.w.) respectively. the high percentage of damage reduction associated with the absence of mutagenic effects indicates the possibility of biopolymer chemoprotection action. it can also be considered a functional food candidate to be used as co-adjuvant chemotherapy to prevent side effects.

Abstract: bathymetric, sedimentological and seabed acoustic backscatter data were integrated into a 3d gis (geographic information system). 38 khz echo sounders were used to collect data over 14,596 nautical miles during six fish biomass assessment cruises over the continental shelf, slope and adjacent oceanic regions between chuí-rs (34°s) and cape s?o tomé-rj (22°s), brazil. the bathymetric set of data were complemented with satellite altimetry and also with data from general bathymetric charts in order to build a bathymetric grid with 5,397,090 points. from this grid, a 3d virtual representation of the seabed was generated, using the language for description of three-dimensional interactive environments x3d. bottom features such as rio grande cone and terrace, s?o paulo plateau and others, were identified in the bathymetric maps. 3d gis implementation allowed to analyze backscatter and sedimentological information over the bathymetric grid, demonstrating to be a useful tool to highlight areas with characteristically phosphatic deposits. 3d gis proved to be more valuable than two-dimensional representations because allowed observation of details and interpretation of information from different not static viewpoints.

Abstract: approximately 10% of thyroid cancers are present in patients less than 21 years of age, representing 3% of all cancers of children and adolescents, with predominance in females 2:1 in relation to males. thyroid cancers in this age group are usually papillary (90%), bilateral, multifocal and bigger in size compared to adults. capsule invasion and lymphatic and pulmonary metastases are more frequent in children. radiation sensitivity seems to represent an important factor in prepubertal patients. familial history is reported in 5% of the cases. genes such as ret/ptc, ras and braf are usually involved in thyroid carcinogenesis in this age group. cervical adenomegaly is a common clinical presentation, but does not represent a poor prognostic factor in children. ultrasound and fine needle aspiration biopsy are valuable diagnostic procedures. surgery is the preferred treatment including thyroidectomy and ganglionary excision, followed by ablative radioiodine therapy. l-thyroxine replacement with suppressive dosage should be employed targeting chronic tsh suppression. long-term prognosis is usually better in children when compared with adults. plasma thyroglobulin measurement is also useful to detect residual thyroid cancer disease.

Abstract: background: endovascular approach to the aortic arch is an appealing solution for selected patients. objective: to compare the technical and clinical success recorded in the different anatomical settings of endografting for aortic arch disease. methods: between june 1999 and october 2006, among 178 patients treated at our institution for thoracic aorta disease with a stent-graft, the aortic arch was involved in 64 cases. according to the classification proposed by ishimaru, aortic zone 0 was involved in 14 cases, zone 1 in 12 cases and zone 2 in 38 cases. a hybrid surgical procedure of supra-aortic debranching and revascularization was performed in 37 cases. results: zone 0. proximal neck length: 44±6 mm. initial clinical success was 78.6%: two deaths (stroke), one type ia endoleak. at a mean follow-up of 16.4±11 months the midterm clinical success was 85.7%. zone 1. proximal neck length: 28±5 mm. initial clinical success was 66.7%: 0 deaths, four type ia endoleaks. at a mean follow-up of 16.9±17.2 months the midterm clinical success was 75.0%. zone 2. proximal neck length: 30±5 mm. initial clinical success was 84.2%: two deaths (one cardiac arrest, one multiorgan embolization), three type ia endoleaks, one case of open conversion. two cases of delayed transitory paraparesis/paraplegia were observed. at a mean follow-up of 28.0±17.2 months the midterm clinical success was 89.5%. conclusions: this study and a literature review demonstrated that hybrid procedure for aortic arch pathology is feasible in selected patients at high risk for conventional surgery. our experience is still limited by the relatively small sample size. we propose to reserve zone 1 for patients unfit for sternotomy or in cases with aortic neck length > 30 mm following left common carotid artery debranching. we recommend to perform complete aortic rerouting of the aortic arch in cases with lesser comorbidities and shorter aortic neck.

Abstract: cystic fibrosis (cf) is the most common recessive autosomal disease among caucasian. children with cf have benefitted from advances in medical and nutritional treatments, and this can be gleaned from the improvement in the survival of these patients. the increase in the survival rate brought with it the appearance of co-morbidities related to cf. nowadays cystic fibrosis-related diabetes (cfrd) is considered the most common complication associated with cf. it can appear as early as infancy or adolescence, and its prevalence can be as high as 50% in adult patients. because of its high prevalence, difficulties in early detection and the risks involved, in recent years several studies and consensuses have focused on this condition, adding information about the epidemiology, pathophysiology, prognosis and treatment of cfrd. the main aspects of these new concepts, as well as the current recommendations for its diagnosis and follow-up, will be presented in this study.

Abstract: pollen, spores and other palynomorphs preserved in sediments are basic reference material for palaeoenvironmental studies. the aim of this research is to present the taxonomy of fungal and cryptogamic palynomorphs contained in sediments from two araucaria forests on the eastern plateau of rio grande do sul, thus contributing to palaeoenvironmental reconstitution in the region. the analysis involved samples from two holocene sedimentary profiles from s?o francisco de paula (profile 1, alpes de s?o francisco, 132 cm; profile 2, banhado amarelo, 101 cm), collected with a hiller sampler. chemical processing of the samples followed standard methodology, using hcl, hf, koh, acetolysis and slide mounts in glycerol-jelly. light microscopy was used in the analysis. palynomorphs of eight fungi, six algae, five bryophytes and 19 pteridophytes are presented. the description of each palynomorph is followed by illustrations and, whenever possible, by ecological data from the original organism. distinct palynomorphs preserved in the sediments reveal the potential of this material in providing relevant environmental information, in association with pollen grains, in the study of genesis and expansion of the araucaria forest.
