Quantification of the Expression of HIF-1alpha by Real-Time PCR in Rat Hepatocytes Cultures Invaded by Shigella flexneri under Normoxic and Hypoxic Conditions  [PDF]

Abstract: Aim: Shigella flexneri (S. flexneri) is a gram-negative enterobacterium responsible for severe intestinal end systemic infection in humans. The bacteria can reach the liver due to degeneration of the colonic epithelium. Hypoxia is present in many human diseases and can induce the expression of the transcription factor HIF-1alpha that may have a cell protective role. The influence of hypoxia and HIF-1alpha on bacterial infection, studied in this work, is unclear. Hypoxia inducible factor-1alpha (HIF-1alpha) is a transcription factor that acts as a master regulator of gene expression induced by hypoxia. Methods: We compared the ability of S. flexneri to invade rat hepatocytes in primary culture both in normoxic and hypoxic conditions. We evaluated TNF-alpha released by hepatocytes, apoptosis rate and HIF-1alpha expression by confocal microscopy as well as real time PCR technique. Results: We showed that S. flexneri invaded less hepatocytes previously submitted to 24 h hypoxia (6.5% O2) than those cultivated in normoxia (21% O2). S. flexneri also induced HIF-1α expression in hepatocytes, TNF-α secretion and apoptosis. Conclusion: a) Hypoxia alone was not a stimulus to TNF-α secretion, but induced cell apoptosis and HIF-1α expression; b) S. flexneri was able to invade rat hepatocytes and hypoxia apparently influenced significantly bacterial cell invasiveness; c)

Abstract: background: during a period of the embryonic life, the sciatic vein is the main lower limb collector. in vascular embryogenesis, there is a differentiation of the angioblasts in a primitive vascular plexus, with posterior remodeling and expansion. consequently, anomalies may occur during this process. when there is persistence of the sciatic vein, it may communicate with the small saphenous vein or with the popliteal vein during its route, being anastomosed to the superior perforating vein and to the medial circumflex femoral vein. objective: to report a case of bilateral persistent sciatic vein on the lower limbs in comparison to the literature. methods: thirty-two lower limbs from 16 corpses preserved in formaldehyde were dissected at the laboratory of anatomy of the discipline of topographic anatomy of the medical school of universidade santo amaro (unisa), during 2006 and 2007, and the sciatic vein was observed in 2 lower limbs of one single corpse. results: on the left lower limb of a corpse that presented bilateral anomaly, the vein had 37 cm, emerging on the popliteal vein, accompanying the sciatic nerve, perforating the long adductor muscle and leading into the deep femoral vein. on the right lower limb, it measured 36 cm, emerged receiving the veins of the anterior tibial compartment, accompanied the sciatic nerve, perforated the long adductor muscle and led into the internal iliac vein. conclusion: the anatomical variations of the lower limb venous system are the most common ones. the persistent sciatic vein may cause chronic venous failure in the lower limbs and, in this manner, must be investigated aiming at a better clinical or surgical management.
Profile of Patients Treated in Outpatient Vestibular Rehabilitation at a Hospital in São Paulo  [PDF]

Abstract: Introduction: Currently it is very common that the search for diagnosis and treatment for curing diseases can cause vertigo or dizziness. Objective: This study sought to characterize the profile of patients seen in the clinic for vestibular rehabilitation in a tertiary hospital in the last 10 years. Methods: Survey questionnaires of patients with dizziness. Results: The prevalence of treated subjects was female 65.3% (N = 439). The average age was 54.9 years old. Dizziness type roundabout was more prevalent 33.04% (N = 222). Tinnitus occurred in 58.33% of the population. The time of most observed dizziness was less than 5 years 70.68% (N = 475). Neurovegetative symptoms appeared in 63.98% (N = 430) of the population. The bilateral normal hearing was more standard 35.26% (N = 237). The topographic diagnosis of peripheral origin was the most usual 65.47% (N = 440). Conclusion: The peripheral otoneurologic involvement was more prevalent in this population, affecting more women at an average age of 54.9 years old. The most prevalent symptoms were vertigo, nausea, tachycardia, sweating, vomiting and tinnitus, characterizing the peripheral vestibular impairment. The time of dizziness was less than five years. Audiological standard normal curve was predominant and the symptom of tinnitus was the most reported. The most prevalent comorbidity was Hypertension.

Abstract: The adoption of performance indicators promotes knowledge of quantitative and qualitative material wastes in businesses and, when inserted in a collaborative process, provides a comparative evaluation of results between companies and, thereafter, an identification of best practices (bench-marking). The purpose of this paper is to present the best practices identified by performance indicators, related to the measurement of wastes and associated to construction companies participation in the benchmarking research process “Implementation of a system of performance indicators of cement-based construction technologies of the Community of Construction of Recife city in Brazil-PROGRIDE”, coordinated by the Brazilian Association of Portland Cement-ABCP. Therefore, it was sought to characterize best practices that led to the benchmarking of performance indicators related to the wastes of concrete, industrialized mortar for masonry settling and blocks/ bricks. As a contribution, a set of factors that characterize the best practices for each technology and conducted to benchmarking were identified.
Exchange, experimentation and preferences: a study on the dynamics of manioc diversity in the Middle Solim es, Amazonas

Abstract: The paper presents a study on the manioc diversity in the Middle Solim es region, focusing largely on communities located in the Sustainable Development Reserves of Mamirauá and Aman , state of Amazonas, Brazil. The study combines quantitative and ethnographic data. The analysis of survey data collected in 13 communities in the 'várzea' and in the 'terra firme' revealed the following pattern of manioc diversity: a total richness of 54 varieties, demonstrating a broad distribution of a small number of varieties and a local occurrence of the majority; an average richness of ten varieties per community; and an average of three varieties maintained per household. A temporal analysis of survey data collected at the household level illustrates the dynamic nature of this regional diversity. Over the course of five years, almost all the 55 accompanied families altered the composition of manioc varieties in their collections; however, the size of these collections showed little variation. To discuss the dynamics of diversity, we conducted qualitative research in three communities. This analysis sought to understand the social and environmental conditions with which farmers contend, patterns of manioc management, and the logic behind farmers' preferences for certain manioc varieties. The research demonstrates that maniva diversity is a result of active experimentation, and that collections of manivas maintained by farmers are dynamic and ever-changing. This dynamism is defined by a series of factors that include economic practices, environmental conditions, and the historical relationship of the regional population with manioc.

Abstract: this paper presents a numerical study for obtaining cooling curves of orange valência (citrus sinensis o.) and banana prata (mussa balbisiana colla), in different positions along the bed and conditioned in package with 40% of effective opening area. the fruits were pre-cooled in a forced-air system (1.933m3/h), at 1°c and 7°c, rh= 88,4 ± 2,0% and air velocity around 1m/s. a two-dimensional mathematical model was applied, assuming prolate spheroid coordinate, to characterize the transient heat transfer process inside the fruits considering convective profile conditions at the surface of the body. the generated equations were numerically solved by finite-volume method. the convective heat transfer coefficients were obtained using the least square method by comparison between experimental and numerical data. the comparative analysis of the theoretical and experimental curves presented a satisfactory agreement, with error between 5% and 7%. it was noted that the convective heat transfer coefficient changes with the fruits position in the bed and cooling time present a variation of approximately 38% among the different points. the spatial distributions of temperature inside the fruits, for three times, demonstrate the existence of a temperature differential between the center and the surface of 30%, approximately. the model predicts that the cooling rate is in the extremes of the banana. the bi and fo numbers obtained represent very well the heat transfer process in the fruits under different relationships of volume/area.

Abstract: Neste trabalho apresenta-se um estudo numérico para a obten o das curvas de resfriamento de laranja Valência, (Citrus sinensis O.) e banana prata (Mussa balbisiana Colla), em posi es diferentes ao longo do leito e acondicionadas em embalagens com 40% de área efetiva de abertura. Os frutos foram resfriados num sistema de circula o com ar for ado (1.933m3/h), à temperatura de 1°C e 7°C, umidade relativa de 88,4 ± 2,0% e velocidade do ar em torno de 1m/s. Um modelo matemático bidimensional, em coordenadas esferoidais prolato, foi aplicado para predizer a condu o de calor transiente dentro das frutas, assumindo-se condi o de contorno convectiva na superfície do corpo. As equa es geradas foram resolvidas numericamente pelo método de volumes finitos e o coeficiente convectivo de transferência de calor foi obtido aplicando o método de ajuste do erro quadrático mínimo, entre os dados experimentais e numéricos. A análise comparativa das curvas teóricas e experimentais mostrou uma concordancia satisfatória, com valores de erro entre 5% e 7%. Notou-se que o coeficiente de transferência de calor varia com a posi o dos frutos no leito e que o tempo de resfriamento apresenta uma varia o de aproximadamente 38% entre os diferentes pontos. A distribui o espacial de temperatura no interior dos frutos, para três instantes de tempo, demonstrou a existência de um diferencial de temperatura entre o centro e a superfície de aproximadamente 30%. O modelo matemático prediz que a taxa de resfriamento nas bananas é muito mais alta nas pontas que nas outras regi es do fruto. O número de Bi e de Fo mostraram-se adequados e representativos do processo estudado, caracterizando adequadamente o resfriamento de corpos em qualquer rela o de área/volume diferente.

Abstract: this study has as objectives, analyze the scientific and technical publications about the standard procedures of precautions in national and international newspapers and to highlight important aspects of articles about the nurse's paper in the prevention and control of hospital infection. it constituted as sample of the research the secondary sources in form of books and newspaper, with the purpose of putting us in direct contact with articles raised about the use of the precautions standard for the hospital infections in the nursing care, between 1999 and 2005. 10 articles related to the use of standard precautions had been found: 70% had been gotten in bank of teses, 20% in the site scielo and 10% in the library of a public university of rio de janeiro, being 40% published in the year of 2003. the results showed that, in this period, in spite of few publications, there was a continuous renewal of the theme, such as national level, as international.

Abstract: civility is a prosocial behavior regulated by tacit social norms. however, few studies have investigated the factors determining civility in urban contexts. the purpose of this paper was to test the influence of gender, pedestrian density, and social categorization on civility. three field experiments simulated everyday social situations. results showed an overall help frequency above 65%. study 1 revealed that civility is influenced by the gender of the researcher and density of pedestrians. studies 2 and 3 found no support for an endogroup favoritism hypothesis, as predicted by social identity theories. the theoretical and practical implications of theses findings for civility are discussed.

Abstract: Na busca por competitividade, as empresas buscam estratégias logísticas que elevem seu nível de servi o sem que seu lucro seja penalizado. Neste contexto, a ado o de Centros de Distribui o (CD) surge como uma alternativa eficaz para tratar o trade-off entre custos logísticos e nível de servi o ao cliente. O objetivo do trabalho é mostrar como diferentes modelos de CD podem auxiliar na busca por solu es logísticas eficientes, para diferentes produtos, com características logísticas diversas. Para tanto, s o analisadas as estratégias adotadas por três empresas de diferentes segmentos, que utilizam CD, além de uma quarta, que embora ainda n o possua um CD, estuda a viabilidade da ado o de um.
