Effects of zinc on carbon metabolism and its enzymes activities in leaves of balsam pear

Abstract: Effects of zinc on carbon metabolism and its enzymes activities in leaves of balsam pear were studied by field trial . The results show that in the control plants stressed under a low level of zinc, there appear lower contents of sucrose, soluble sucrose ,starch and zinc, lower activities of carbonic anhydrase(CA) and sucrose phosphate synthase (SPS). How- ever , the activities of amylase, acid invertase(AI)and neutral invertase (NI)in the upper and lower leaves are higher in CK than in other treatments. There is a significant correlation between the physiological properties.

Abstract: 苦瓜(株洲长白Momordica charantia cv.Zhuzhou Changbai)叶片喷施硫酸锌和钼酸钠均可显著增加苦瓜产量,提高叶片多胺(PAs)、抗坏血酸(ASA)、吲哚乙酸(IAA)、赤霉素(GA3)含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)的活性,减少脱落酸(ABA)、丙二醛(MDA)含量和自动氧化速率(AOR),从而抑制膜脂过氧化,延缓叶片衰老。苦瓜产量与叶片IAA、GA3、PAs、SOD、POD、CAT呈极显著正相关,而与MDA、AOR、ABA呈极显著负相关,叶片MDA含量与ABA、AOR呈极显著正相关,而与PAs、IAA、GA3、SOD、POD、CAT呈显著负相关。
Differential Gene Expression in Sugarcane in Response to Challenge by Fungal Pathogen Ustilago scitaminea Revealed by cDNA-AFLP

Abstract: Differential gene expression in sugarcane during sugarcane-Ustilago scitaminea interaction was conducted in a smut-resistant genotype. Using cDNA-AFLP along with silver staining, a total of 136 transcript-derived fragments (TDFs) were found to be differentially expressed in response to challenge by U. scitaminea. Forty TDFs, 34 newly induced plus six with obvious upregulated expression after infection, were sequenced and validated by RT-PCR analysis. These results demonstrated that the expression of 37 out of these TDFs in RT-PCR analysis was consistent with that in cDNA-AFLP analysis. Based on BlastX in NCBI, 28 TDFs were assumed to function in sugarcane under U. scitaminea stress. Analysis of expression profile of three TDFs revealed that they responded differently after infection with U. scitaminea, and the transcription was significantly enhanced. The response of two TDFs, SUC06 and SUC09, occurred before that of SUC10. This study enriches our knowledge of the molecular basis for sugarcane response to U. scitaminea infection.

Abstract: 分析了在CuCl2作用下辣椒叶片倍半萜环化酶活性,倍半萜环化酶mRNA表达,细胞GSH和GSSG代谢及膜脂过氧化,结果表明,Cu^2 能诱导辣椒叶片表达倍半萜环化酶活性,酶活性的表达与相应的基因转录有关,辣椒叶片在CuCl2作用下合成了含-SH的螯合肽,相应地GSH含量下降,GSSG的含量有所上升,细胞膜过氧化作用加剧,推测细胞膜脂过氧化产物或GSH氧化产物可能参与了CuCl2诱导辣椒倍半萜环化酶基因表达的信号传递作用。

Abstract: 根据已发表的同源基因序列,利用RT-PCR技术分离了斑茅(Erianthus arundinaceus)的GAPDH和APRT两个看家基因片段,用它们作为cDNA芯片阳性参照,以未经聚乙二醇(PEG)胁迫处理的斑茅叶片为对照,和PEG胁迫的4组材料同cDNA芯片进行杂交分析。杂交结果显示,GAPDH杂交的Cy5与Cy3平均信噪比(Signal/Noise,S/N)分别为56.12和60.8,APRT杂交的Cy5与Cy3平均信噪比分别为51.06和47.25,信噪比均很高;同时两个看家基因的杂交都显示出极强的信号,其中GAPDH的杂交信号值大于10000,APRT也在8000以上,杂交结果可靠。分析了PEG胁迫4个时段BADH与两个看家基因的表达,BADH的表达有明显变化,而看家基因表达均较稳定。上述结果表明所克隆的两个看家基因在斑茅中表达量高,且PEG胁迫下表达较为稳定,是基因芯片理想的阳性参照。

Abstract: 酿酒酵母乙醇合成代谢过程中, 阻断或削弱乙醛至乙酸代谢流不但能增强乙醇合成流, 同时还能降低发酵乙酸含量。本研究以乙醇脱氢酶Ⅱ(adh2)基因缺陷型酿酒酵母YS2-Dadh2为出发菌株, 应用长侧翼同源两步PCR(LFH-PCR)策略构建乙醛脱氢酶Ⅵ(ald6)基因敲除组件, 转化酿酒酵母YS2-Dadh2敲除ald6基因, 之后转入表达质粒pSH65到阳性克隆中, 半乳糖诱导表达Cre重组酶切除Kanr基因筛选标记, 最后, 传代丢失质粒pSH65获得单倍体ald6基因缺失突变株。利用同样的敲除组件和技术再次敲除其等位基因, 最终获得双基因缺失突变株YS2-△adh2-Dald6。发酵实验表明与出发菌株YS2相比, 突变株乙酸合成量降低18%, 乙醇最高产量提高12.5%。
Cloning and Expression Analysis of Cytosolic Malate Dehydrogenase Gene from Sugarcane

Abstract: A full-length cDNA sequence of sugarcane (Saccharum officinarum L.) cytosolic malate dehydrogenase (cMDH) gene, named as Sc-cMDH, was obtained from sugarcane full-length cDNA library by EST sequencing and bioinformatics analysis. The full-length of Sc-cMDH was 1314 bp with 999 bp open reading frame, encoding 332 amino acids. Sc-cMDH shared 86.5%~97.0% of amino acids with cMDH from other 9 species. Sc-cMDH contains typical NAD+ binding motif (T11GAAGQI17) and catalytic motif (I184WGNH188). Real-time PCR analysis showed that the expression of Sc-cMDH was higher in leaves and roots than that in stems of sugarcane.
Reservoir Characteristics of the Xujiahe Formation in Central-west Sichuan Analogous Foreland Basin

Abstract: Reservoirs of the upper Triassic Xujiahe Formation has characteristics of low compositional maturity, low cement content and medium maturity in Sichuan analogous foreland basin. Lithology is regularly distributed in plane, dominated by lithic sandstone in the northwest region. lithic sandstone and feldspathic sandstone mixed in central west region, lithic feldspathic sandstones in southwest region, feldspathic lithic and lithic quartz sandstone mixed in central-south region. Reservoir has poor physical properties in general, which are mainly low porosity and permeability reservoir. Only a few reservoirs with middle porosity and low permeability develop in some regions. Because of the sedimentation, diagenesis and tectonic difference in thrust belt, foredeep depression zone, foreland slope and foreland uplift, of the reservoir porosity has zoning characteristics. Reservoir space in Longmen Mountain thrust belt are mainly fracture with small amount of emposieu and original intergranular pore. The pore type in foredeep depression zone is fracture-secondary dissolved pore, dominated by secondary dissolved pore with little primary pore. In foreland slope, the pore type is mainly secondary dissolved pore and primary pore. While in foreland uplift, the pore is mainly dissolved pore and primary pore. The development of reservoirs is controlled by deposition, diagenesis and tectonic disruption.
Improved Successive Cancellation Decoding of Polar Codes

Abstract: As improved versions of successive cancellation (SC) decoding algorithm, successive cancellation list (SCL) decoding and successive cancellation stack (SCS) decoding are used to improve the finite-length performance of polar codes. Unified descriptions of SC, SCL and SCS decoding algorithms are given as path searching procedures on the code tree of polar codes. Combining the ideas of SCL and SCS, a new decoding algorithm named successive cancellation hybrid (SCH) is proposed, which can achieve a better trade-off between computational complexity and space complexity. Further, to reduce the complexity, a pruning technique is proposed to avoid unnecessary path searching operations. Performance and complexity analysis based on simulations show that, with proper configurations, all the three improved successive cancellation (ISC) decoding algorithms can have a performance very close to that of maximum-likelihood (ML) decoding with acceptable complexity. Moreover, with the help of the proposed pruning technique, the complexities of ISC decoders can be very close to that of SC decoder in the moderate and high signal-to-noise ratio (SNR) regime.
