Large-Scale Conformational Changes of Trypanosoma cruzi Proline Racemase Predicted by Accelerated Molecular Dynamics Simulation

Abstract: Chagas' disease, caused by the protozoan parasite Trypanosoma cruzi (T. cruzi), is a life-threatening illness affecting 11–18 million people. Currently available treatments are limited, with unacceptable efficacy and safety profiles. Recent studies have revealed an essential T. cruzi proline racemase enzyme (TcPR) as an attractive candidate for improved chemotherapeutic intervention. Conformational changes associated with substrate binding to TcPR are believed to expose critical residues that elicit a host mitogenic B-cell response, a process contributing to parasite persistence and immune system evasion. Characterization of the conformational states of TcPR requires access to long-time-scale motions that are currently inaccessible by standard molecular dynamics simulations. Here we describe advanced accelerated molecular dynamics that extend the effective simulation time and capture large-scale motions of functional relevance. Conservation and fragment mapping analyses identified potential conformational epitopes located in the vicinity of newly identified transient binding pockets. The newly identified open TcPR conformations revealed by this study along with knowledge of the closed to open interconversion mechanism advances our understanding of TcPR function. The results and the strategy adopted in this work constitute an important step toward the rationalization of the molecular basis behind the mitogenic B-cell response of TcPR and provide new insights for future structure-based drug discovery.
Effect of propionic and lactic acids on in vitro ruminal bacteria growth

Abstract: the objective of this work was to evaluate the effect of the levels of lactic and propionic acids on in vitro fermentation of ruminal microorganisms. in experiment 1, the levels, in a total of 12 were the following: addition of 0 (control 1), 50, 100, 150, 200 and 250 mm of lactic acid and 0 (control 2), 50, 100, 150, 200 and 250 mm of propionic acid, respectively, in incubation flasks, which contained ruminal inoculum, glucose and synthetic culture medium, with two repetitions for each combination. in experiment 2, the combinations, in a total of 4, were the following: presence of 12 and 24 mm of propionic acid and 0 mg of glucose, respectively; presence of 12 and 24 mm of propionic acid and 40 mg of glucose, respectively, to the incubation flasks which contained ruminal inoculum, with or without glucose and in synthetic culture medium with two repetitions each. there was no effect on the specific growth velocity of ruminal microorganisms in the presence of lactic acid or propionic acid. however, when there were greater concentrations of these acids in the media, there was a longer lag phase in the microorganism phase. acid propionic at the concentration of 24 mm inhibited the production of acid acetic and butyric acid in a media with glucose. despite of not being used as a source of energy by the ruminal microorganisms, propionic acid affects their metabolism. lactic and propionic acids inhibit growth of some ruminal microorganisms at elevated concentrations.

Abstract: the southwestern region of paraná state is an area of great ecological relevance because it represents an ecotone between two important forest types of the brazilian south-southeastern region: the seasonal semideciduous forest (ssf) and the araucaria forest (af). despite its importance, there is a lack of floristic surveys and vegetation studies in this region. in this study, we assessed floristic and structural attributes of the vegetation at three different native forest sites, located in quedas do igua?u, paraná state. moreover, we made a phytogeografical characterization of the studied forests, based on the data we collected, and on the list of species surveyed in other 52 studies in ssf and af of brazil. samples were taken using the point-centered-quarter method. all individuals with diameter at breast height > 5 cm were identified to the species level. together, 128 species were found. within sites, the number of species varied from 63 to 78. the three areas are distinguished with respect to structure and most abundant species. in addition, the areas differed with respect to the degree of influence of ssf and af on their floristic composition. comparatively, the lower altitude area, located in igua?u river's valley, has more elements of ssf on its floristic composition, while the other two areas, located in higher altitude areas, are more influenced by af, with araucaria angustifolia (bertol.) kuntze on their list of species.
Evaluation of whey fermented by Enterococcus faecium in consortium with Veilonella parvula in ruminant feeding

Abstract: the objective of this study was to evaluate the whey fermented by enterococcus faecium in consortium with veilonella parvula on the in vitro growth of ruminal bacteria and as a supplement in the cattle diet. in the in vitro experiment, a randomized design, with the following combinations was used: ruminal bacteria; ruminal bacteria and inactive whey; ruminal bacteria and active whey; and active whey. in the in vivo experiment, five fistulated zebu holstein-zebu crossbred heifers were distributed in a 5 × 5 latin square. supplements were formulated without the addition of whey, with the addition of two levels of unfermented whey (2.5 and 5 l/day) or two levels of fermented whey (2.5 and 5 l/day). a positive effect of the whey fermentation was detected on the consumption of dry matter, organic matter, crude protein, ether extract, non-fiber carbohydrates and neutral detergent fiber, corrected for ash and protein in kg/day. no effects of whey were observed on the ph and concentration of rumen ammonia nitrogen, serum concentration of urea and glucose, urinary excretion of urea or nutrient digestibility, except for the total digestible nutrients. supplementation with whey improved the apparent nitrogen balance, but supplementation with fermented whey decreased the intestinal flow of microbial nitrogen and microbial synthesis efficiency in relation to the unfermented whey. the whey fermentation process does not optimize the physiological responses of heifers supplemented with 2.5 and 5.0 l of whey.

Abstract: spinal cord compressing syndrome due to synovial cyst (sc) of the thoracic spine is a rare clinic condition. we report a case of sc located in the thoracic spine causing spastic paraparesis in a 14 year-old female patient. the sc was removed thoroughly by laminectomy. the patient had an excellent recovery. the etiological and therapeutic aspects are discussed.

Abstract: introduction: several studies, including meta-analysis, have demonstrated the safety, effectiveness and oncologic equivalence of laparoscopic resections when compared to open procedures leading minimally invasive colorectal surgery to be adopted in crescent number of services around the world. this study aims to evaluate the quality of life of patients underwent laparoscopic and open colorectal resections in the early postoperative period. methods: this is a prospective study which evaluated 42 patients underwent laparoscopic and open colorectal resection between may to november 2008 followed up until 60th postoperative day. questionnaires of quality of life were applied in 3th, 7th and 30th postoperative days. statistical analysis consisted of descriptive analysis of global healthy status scores, functional scores and symptoms of eortc/qlq 30. shapiro-wilk, mann-whitney e t de student statistical tests were used to check the data, with level of significance in 0.05. results: most of patients were females (57.1%) with mean age of 61.5 years. it was observed significant difference of "global health status" score on the 30th postoperative between groups, with values of 75.0 and 58.3 for patients underwent laparoscopic and open procedures respectively (p = 0.005). there were no differences in terms of physical function and others as, accomplishment, emotional, cognitive and social functions. in relation to symptoms (fatigue, nausea, pain, dyspnea, insomnia, loss of appetite and constipation) and financial difficulties, there were also no differences between groups. conclusion: our results have demonstrated that patients underwent laparoscopic colorectal resections have better quality of life at the end of first postoperative month when compared to patients underwent to open colorectal resections.

Nitrogen dynamics in a soil-sugar cane system

Abstract: results of an organic matter management experiment of a sugar cane crop are reported for the first cropping year. sugar cane was planted in october 1997, and labeled with a 15n fertilizer pulse to study the fate of organic matter in the soil-plant system. a nitrogen balance is presented, partitioning the system in plant components (stalk, tip and straw), soil components (five soil organic matter fractions) and evaluating leaching losses. the 15n label permitted to determine, at the end of the growing season, amounts of nitrogen derived from the fertilizer, present in the above mentioned compartments.

Abstract: geophysical studies in impacted areas by petroleum products may be described by anomalies of both low and high resistivity, confirmed as contaminant by chemical analyses. this apparent contradiction can reflect processes of contaminants degradation, directly associated with its residence time in the soil, through the generation of by-products that change the physical properties of the soil and groundwater, principally for the mineral dissolution by action of organic acids and by neoformation minerals of oxides and hydroxides minerals. this paper presents the application of electrical resistivity and induced polarization methods in a contaminated industrial area for benzene, toluene, xylene, 1,2 dichloroethene and inorganic salts, in natural attenuation process. the results indicated the mineral neoformation in the center and adjacencies of contaminated area, as it suggested the alterations in physical parameters measured.

Abstract: the application of geophysical methods in environmental studies, essentially in the characterization and monitoring of contaminated areas, is growing and promising. this paper presents the application of the induced polarization method (ip), through the electrical profiling technique, in the determination of the leachate plume originated in the type ditch landfill. in absence of monitoring wells or other direct techniques of investigation in landfill area, the geological characterization together with the application of geophysical techniques they constitute an alternative for preliminary characterization and monitoring leached materials. however, is vital the appropriate understanding of the physical parameter due to possible alterations in the geological environment resultants of the contaminants presents. the results suggest a reduction tendency in the chargeability values in the leachate sense flow, possibly due to the dissolution for attack acid to the metallic materials present in the residues and oxides and hydroxides of iron present abundantly in the soil.
