Bone marrow progenitor cells do not contribute to liver fibrogenic cells

Abstract: AIM: To investigate the contribution of bone marrow (BM) cells to hepatic fibrosis. METHODS: To establish a model of chimerism, C57Bl/6 female mice were subjected to full-body irradiation (7 Gy) resulting in BM myeloablation. BM mononuclear cells obtained from male transgenic mice expressing enhanced green fluorescent protein (GFP) were used for reconstitution. Engraftment was confirmed by flow cytometry. To induce liver injury, chimeric animals received carbon tetrachloride (CCl4) 0.5 mL/kg intraperitoneally twice a week for 30 d (CCl4 30 d) and age-matched controls received saline (Saline 30 d). At the end of this period, animals were sacrificed for post mortem analysis. Liver samples were stained with hematoxylin and eosin to observe liver architectural changes and with Sirius red for collagen quantification by morphometric analysis. α-smooth muscle actin (α-SMA) was analyzed by confocal microscopy to identify GFP+ cells with myofibroblast (MF) characteristics. Liver tissue, BM and peripheral blood were collected and prepared for flow cytometric analysis using specific markers for detection of hepatic stellate cells (HSCs) and precursors from the BM. RESULTS: Injury to the liver induced changes in the hepatic parenchymal architecture, as reflected by the presence of inflammatory infiltrate and an increase in collagen deposition (Saline 30 d = 11.10% ± 1.12% vs CCl4 30 d = 12.60% ± 0.73%, P = 0.0329). Confocal microscopy revealed increased reactivity against α-SMA in CCl4 30 d compared to Saline 30 d, but there was no co-localization with GFP+ cells, suggesting that cells from BM do not differentiate to MFs. Liver flow cytometric analysis showed a significant increase of CD45+/GFP+ cells in liver tissue (Saline 30 d = 3.2% ± 2.2% vs CCl4 30 d = 5.8% ± 1.3%, P = 0.0458), suggesting that this increase was due to inflammatory cell infiltration (neutrophils and monocytes). There was also a significant increase of common myeloid progenitor cells (CD117+/CD45+) in the livers of CCl4-treated animals (Saline 30 d = 2.16% ± 1.80% vs CCl4 30 d = 5.60% ± 1.30%, P = 0.0142). In addition the GFP-/CD38+/CD45- subpopulation was significantly increased in the CCl4 30 d group compared to the Saline 30 d group (17.5% ± 3.9% vs 9.3% ± 2.4%, P = 0.004), indicating that the increase in the activated HSC subpopulation was not of BM origin. CONCLUSION: BM progenitor cells do not contribute to fibrosis, but there is a high recruitment of inflammatory cells that stimulates HSCs and MFs of liver origin.
Functional Analysis of PGRP-LA in Drosophila Immunity

Abstract: PeptidoGlycan Recognition Proteins (PGRPs) are key regulators of the insect innate antibacterial response. Even if they have been intensively studied, some of them have yet unknown functions. Here, we present a functional analysis of PGRP-LA, an as yet uncharacterized Drosophila PGRP. The PGRP-LA gene is located in cluster with PGRP-LC and PGRP-LF, which encode a receptor and a negative regulator of the Imd pathway, respectively. Structure predictions indicate that PGRP-LA would not bind to peptidoglycan, pointing to a regulatory role of this PGRP. PGRP-LA expression was enriched in barrier epithelia, but low in the fat body. Use of a newly generated PGRP-LA deficient mutant indicates that PGRP-LA is not required for the production of antimicrobial peptides by the fat body in response to a systemic infection. Focusing on the respiratory tract, where PGRP-LA is strongly expressed, we conducted a genome-wide microarray analysis of the tracheal immune response of wild-type, Relish, and PGRP-LA mutant larvae. Comparing our data to previous microarray studies, we report that a majority of genes regulated in the trachea upon infection differ from those induced in the gut or the fat body. Importantly, antimicrobial peptide gene expression was reduced in the tracheae of larvae and in the adult gut of PGRP-LA-deficient Drosophila upon oral bacterial infection. Together, our results suggest that PGRP-LA positively regulates the Imd pathway in barrier epithelia.

Abstract: We obtain new families of (1,2)-symplectic invariant metrics on the full complex flag manifolds F(n). For n≥5, we characterize n−3 different n-dimensional families of (1,2)-symplectic invariant metrics on F(n). Each of these families corresponds to a different class of nonintegrable invariant almost complex structures on F(n).
Directorios cruzados: un estudio sobre la cúpula empresarial en la Argentina actual

Abstract: the aim of this article is to analyze, through the interlocking directorates method, the networks that are established between some of the highest revenues corporations in argentina. most of the research studies focused on the analysis of large corporations are based on economic indicators, such as studies on concentration of wealth. in the present case, through the concept of social networks, we focus on the social relations that are developed among big corporations in our country. we observe that the corporations studied represent a highly interconnected sector; in addition, through network analysis, we give evidence that supports the resources dependence theory, as well as the financial control theory: the interlocks among corporations respond to the type of resources under their control, in particular, the significant role of financial resources over the others. the firms were selected from the revenue company ranking published by the mercado magazine for the year 2005; we have added to the sample some of the most important banks that operate today in argentina and other four entities of supreme relevancy in the economic system, with the intention of knowing their relationship with the rest of the companies.

Abstract: this paper describes the co-authorship networks of mauricio lópez and ignacio ellacuría in tierra nueva's publications in 1970. tierra nueva was a great diffuser of latin american christian progressivism in that period. to rebuild the networks of ellacuría and lopez, it was analyzed 63 books co-authored published in the 1970's and the magazines: fichas latinoamericanas (1974), cristianismo y sociedad (1970-1975) y revista cuadernos de cristianismo y sociedad (1976- 1981). it's the aim of this study to make a contribution to the reconstruction of the political-religious network in latin america of the seventies, from a different analytical perspective. to this end, in the networks mauricio lópez and ignacio ellacuría were analyzed: the origin, the number of publications and careers of each member.

Abstract: the objective of this work is to take the development from the industry of the wine in mendoza like an indicator of the imbrication of the social dynamics of the chilean central valley and mendoza in argentina. the history of this province is very united to chile from its foundation. with respect to the history of wine-growing, in this work we will distinguish four moments: the wine elaboration of chile in mendoza before the creation of the virreinato of the river plate; the conformation of families argentine-chilean wine-producers during the xixth century; the relation between the relation between wine-producing families of mendoza and groups antipro-government during the government of the popular unit in chile and finally, the wine done in mendoza with chilean capitals as of the decade of 1990.

Abstract: mauricio lopez, philosopher and doctor of theology, was secretary for latin america of the universal federation of student christian movements (wscf), secretary of the department of church and society of the world council of churches (wwc) and chancellor of the universidad nacional de san luis (argentina). in 1977 he was kidnapped is a desaparecido (missing) of the last military dictatorship. this article seeks to reconstruct an ecumenical network of intellectuals from the analysis of the mauricio lopez's coauthorship network. we will analyze 29 publications, written between 1945 and 1972, which involved 92 co-authors using the methodology of social network analysis (sna) and the software ucinet 6 and netdraw.

Abstract: this article seeks to show how different conceptions of aesthetics can determine the relationship between art and politics. to achieve this, it first explores the link found by walter benjamin between l'art pour l'art and the "aestheticization of politics." it then compares this idea to the "politics of aesthetics" and "aesthetics of politics," which jacques rancière locates in the heart of what he calls the "distribution of the sensible." the article highlights how autonomous aesthetics leads to an aestheticization of politics, while an inherently political aesthetics illuminates the liberating potential of art.
