An improved method for measuring the stability of a three-state unfolding protein

Abstract: In the current three-state protein unfolding model, the two transitions are considered to be independent and each transition is fitted to a two-state unfolding model. This three-state unfolding process is therefore composed of two sequential two-state unfolding processes. In this paper, a modified method is presented to determine the value of the unfolding free energy [δG total 0 (H2O)] for the three-state unfolding equilibrium of proteins. This method is demonstrated on the apoCopC protein mutant, Y79W-W83F-Cu, which unfolds via a three-state process. The value of ΔG total 0 (H2O) calculated using the modified method was found to be more accurate in determining ΔG total 0 (H2O) than the previously reported method.
Nonhost Interaction of Phytophthora sojae and Arabidopsis thaliana and Genetic Analysis of a Susceptible Mutant

Abstract: Phytophthora sojae and Arabidopsis thaliana were used in this study as a nonhost plant-oomycete interaction system to investigate the genetic basis of nonhost resistance against oomycete pathogens in plants. A collection of more than 40 000 T3 A. thaliana T-DNA mutant plants representing 12 000 independent insertion lines were screened by inoculating detached leaves with P. sojae zoospores, and the susceptible mutant was re-confirmed by pathogen inoculation and cytological characterization. A number of P. sojae-susceptible A. thaliana mutants were successfully obtained, and one of them, mutant 581-51, was shown to be stably susceptible to P. sojae infection. Water-soaked lesions formed on the detached leaves within 3 days, as did oospores and sporangia 4–5 days after inoculation with P. sojae zoospores. Cytological characterization revealed the formation of haustoria-like structures. Southern analysis showed the presence of four T-DNA insertion events in the mutant. Genetic analysis indicated that the susceptibility to infection by the nonhost pathogen P. sojae in the mutant 581-51 was likely controlled by a single recessive gene.
Fluorescence study on the interaction between apoCopC and cupric

Abstract: The interaction between apoCopC and cupric was investigated by fluorescence spectra, in phosphate (20 mmol/L) buffer at pH 6.0. Results suggest that the environment is measured to be hydrophobic completely around tryptophan (83). At the same time, apoCopC fluorescence at 320 nm was significantly quenched with the addition of cupric and the 1:1 stoichiometric ratio of apoCopC to cupric was confirmed by fluorescence. In addition, the conditional binding constants were calculated to be KCu_Copc = (1.8±0.58)× 1013 mol 1 L on the basis of the results of fluorescence titration curves. The apoCopC has the ability to bind specifically cupric ion.

Abstract: In the title binuclear iron(III) complex, [Fe2(C14H10N2O2)3]·4C4H8O or [Fe2(salda)3]·4THF [H2salda = 2,2′-(azinidimethylene)diphenolate and THF is tetrahydrofuran], the ligand possesses a rotationally flexible single N—N bond. Three dinucleating O,N:N′,O′-donor ligands provide three diazine (=N—N=) bridges between the metal ions, yielding a binuclear triple helicate structure with crystallographic C2 symmetry, the rotation axis bisecting one N—N bond.
H-RBAC: A Hierarchical Access Control Model for SaaS Systems

Abstract: SaaS is a new way to deploy software as a hosted service and accessed over the Internet which means the customers don’t need to maintain the software code and data on their own servers. So it’s more important for SaaS systems to take security issues into account. Access control is a security mechanism that enables an authority to access to certain restricted areas and resources according to the permissions assigned to a user. Several access models have been proposed to realize the access control of single instance systems. However, most of the existing models couldn’t address the following SaaS system problems: (1) role name conflicts (2) cross-level management (3) the isomerism of tenants' access control (4) temporal delegation constraints. This paper describes a hierarchical RBAC model called H-RBAC solves all the four problems of SaaS systems mentioned above. This model addresses the SaaS system access control in both system level and tenant level. It combines the advantages of RBDM and ARBAC97 model and introduces temporal constraints to SaaS access control model. In addition, a practical approach to implement the access control module for SaaS systems based on H-RBAC model is also proposed in this paper.
Metal ion-binding properties of wild-type and mutant D37K of ciliate Euplotes octocarinatus centrin

Abstract: Ciliate Euplotes octocarinatus centrin (EoCen) is an EF-hand calcium-binding protein closely related to the prototypical calcium sensor protein calmodulin. The first amino acid of the Ca2+-binding loops found in the EF-hand calcium-binding proteins is a highly conserved aspartic acid residue. The D37K mutant was produced to elucidate the metal binding role of the first aspartic acid of the EF-loop I of EoCen. Aromatic-sensitized Tb3+ fluorescence results indicated that the metal binding ability of loop I was lost due to the D37K mutation. Based on fluorescence titration curves of Lu2-D37K, the conditional binding constants of the EoCen loop II were quantitatively found to be K II = (1.61 ± 0.04) × 105 L mol 1 and K II = (3.52 ± 0.08) × 102 L mol 1 with Tb3+ and Ca2+, respectively. Using 2-p-toluidinylnaphthalene-6-sulfonate as a hydrophobic probe, exposure of the hydrophobic surface upon metal binding was found to be significantly reduced for the metal ion-saturated EoCen D37K mutant.
Binding of Euplotes octocarinatus centrin with target peptide melittin

Abstract: Interactions between model target peptide melittin (ME) and Euplotes octocarinatus centrin (EoCen) were investigated by fluorescence spectra, circular dichroism (CD) spectra and native polyacrylamide gel electrophoresis (PAGE). In 0.1 mol/L N-2-hydroxyethylpiperazine-N-2-ethanesulfonic acid (Hepes) and 150 mmol/L NaCl at pH 7.4, EoCen and isolated short C-terminal domain of EoCen (SC-EoCen) form 1:1 peptide: protein complexes. However, no detectable signal changes can be observed while isolated N-terminal domain of EoCen (N-EoCen) or isolated long C-terminal domain of EoCen (LC-EoCen) was added into solution of ME. The interaction between EoCen and ME is specified exclusively for the short C-terminal domain of EoCen. On the basis of fluorescence titration curves, the conditional binding constants of ME with EoCen and SC-EoCen were calculated to be logK ME-EoCen = 6.81±0.33 and logK ME-SC-EoCen = 6.51±0.45, respectively.
Binding and endocytosis of monoterbium transferrin by K562 cells

Abstract: Using isotopic labeling of human serum apotransferrin, the binding and the endocytosis of monoterbium transferrin (Tbc-apotransferrin, Tbc-apotransferrin-FeN) by K562 cells, a human leukemic cell line, have been investigated. There are about (8.58 ± 2.41) × 105 binding sites per cell surface at 0°C. The association constant for Tbc-apotransferrin binding is 4.1 × 10 mol 1· L, for Tbc-apotransferrin-FeN 2.7×107 mol 1 · L at 0°C. At pH 7.4, upon warming cells to 37°C, endocytosis starts. The rate constants for the endocytosis are about 0.97 min 1 and 0.31 min 1 and the endocytosis ratio reaches 56% and 80% for Tbc-apotransferrin and TbC-apotransferrin-FeN, respectively.
The role of cupric in maintaining the structure of CopC

Abstract: The CopC protein from pseudomonas syringae pathovar tomato is expressed as one of four proteins encoded by the operon CopABCD that is responsible for copper resistance. And there are one tryptophan (83), one tyrosine (79), and three phenylalanines (35, 43, 99) in apoCopC. The fluorescence peak of apoCopC is located near 320 nm, and the peak shifts toward 353 nm in the presence of 10 mol·L 1 urea with excitation at 280 nm. Using urea as a chemical agent, the conformational stabilities of apoCopC and Cu N 2+ -CopC were monitored by fluorescence spectrum in 20 mmol·L 1 phosphate buffer and 100 mmol·L 1 sodium chloride at pH 6.0. The free energy of stabilization for apoCopC and Cu N 2+ -CopC is 16.29±0.65 kJ·mol 1 and 26.26±0.35 kJ·mol 1, respectively. The distance between the tryptophan residue and the Cu2+ in Cu N 2+ -CopC has been studied by observing F rster type nonradiative energy transfer. And it is calculated to be 11.6 .

Abstract: 闽北万木林是我国中亚热带一处以常绿阔叶林为保护主体的自然保护区。植物种类丰富,计有维管束植物160科580属1199种。种子植物区系地理成分复杂,属的分布区类型有13个,热带亚热带分布、温带分布和中国特有分布分别占本区野生种子植物属总数的60.6%、38%和1.4%。本区种子植物的93.2%与华东区系共有,82.7%与华南区系共有。据属的相似性系数和模糊聚类法分析,万木林与武夷山区系的关系最为密切,其次为庐山和九龙山等,再次为罗浮山和其他山地。万木林属泛北极植物区中国-日本森林植物亚区华东地区的一部分,但同时也显示了闽北由华东植物区系向华南植物区系过渡的特征。
