Characterization of New Otic Enhancers of the Pou3f4 Gene Reveal Distinct Signaling Pathway Regulation and Spatio-Temporal Patterns

Abstract: POU3F4 is a member of the POU-homedomain transcription factor family with a prominent role in inner ear development. Mutations in the human POU3F4 coding unit leads to X-linked deafness type 3 (DFN3), characterized by conductive hearing loss and progressive sensorineural deafness. Microdeletions found 1 Mb 5′ upstream of the coding region also displayed the same phenotype, suggesting that cis-regulatory elements might be present in that region. Indeed, we and others have recently identified several enhancers at the 1 Mb 5′ upstream interval of the pou3f4 locus. Here we characterize the spatio-temporal patterns of these regulatory elements in zebrafish transgenic lines. We show that the most distal enhancer (HCNR 81675) is activated earlier and drives GFP reporter expression initially to a broad ear domain to progressively restrict to the sensory patches. The proximal enhancer (HCNR 82478) is switched later during development and promotes expression, among in other tissues, in sensory patches from its onset. The third enhancer (HCNR 81728) is also active at later stages in the otic mesenchyme and in the otic epithelium. We also characterize the signaling pathways regulating these enhancers. While HCNR 81675 is regulated by very early signals of retinoic acid, HCNR 82478 is regulated by Fgf activity at a later stage and the HCNR 81728 enhancer is under the control of Hh signaling. Finally, we show that Sox2 and Pax2 transcription factors are bound to HCNR 81675 genomic region during otic development and specific mutations to these transcription factor binding sites abrogates HCNR 81675 enhancer activity. Altogether, our results suggest that pou3f4 expression in inner ear might be under the control of distinct regulatory elements that fine-tune the spatio-temporal activity of this gene and provides novel data on the signaling mechanisms controlling pou3f4 function.

Abstract: En este artículo se presenta una visión de la ense anza de las matemáticas en las primeras edades que prioriza que los ni os y ni as aprendan a usar las matemáticas en su vida cotidiana. Se argumenta que para aprender a usar las matemáticas es necesario partir de un currículo de matemáticas que contemple dos tipos de conocimientos: los contenidos matemáticos (razonamiento lógico-matemático; numeración y calculo; geometría; medida; y estadística y probabilidad) y, sobre todo, los procesos matemáticos (la resolución de problemas; el razonamiento y la demostración; la comunicación; las conexiones; y la representación), ya que estos procesos ponen de relieve las formas de adquisición y uso de los contenidos matemáticos. Se ofrecen orientaciones didácticas para planificar y gestionar actividades que contemplen las conexiones entre los contenidos y los procesos matemáticos mediante la presentación de dos experiencias implementadas en diferentes centros escolares de la geografía espa ola.Beyond contents, mathematical processes in Early Childhood EducationABSTRACTThis article presents a view of teaching mathematics at an early age that gives priority to boys and girls learning to use mathematics in their everyday lives. It is argued that learning to use mathematics must be based on a mathematics curriculum that considers two types of knowledge: mathematical contents (logical mathematical reasoning; numbering and calculation; geometry; measuring; and statistics and probability) and, especially, mathematical processes (problem solving; reasoning and demonstration; communication; connections; and representation), since they emphasise ways of acquiring and using mathematical contents. Instructional guidelines are offered for the planning and management of activities that consider the connections between contents and the mathematical processes through the presentation of two experiences in different schools around Spain.
Competencias profesionales y creatividad docente en los futuros profesores de música de secundaria: Un estudio a partir del contexto universitario inglés

Abstract: The article presents a research on professional standards, music teaching skills and creative teaching practices in the context of a secondary music teachers training course in England. In order to get reliability, a wide range of qualitative data was collected through multiple techniques –analysis of documents, observations, questionnaires and interviews– and analysed. The results show that skills related with music performance and personal attitudes are the most valued by future teachers. Also, both pre-service secondary music teachers and their supervisors recognize school mentors’ major role in modelling and giving support to the development of creative teaching practices. The relationship between the Standards for Qualified Teacher Status and creative teaching is controversial since Standards are seen to be too pre-scriptive and not related enough to the teacher’s subject specialisation.El artículo presenta una investigación sobre las competencias en la pedagogía musical y la creatividad docente en el contexto de un curso en Inglaterra para la formación de profesores de música de secundaria. Percepciones y actitudes sobre las competencias profesionales docentes son a su vez examinadas. Se han analizado datos cualitativos proceden-tes de múltiples métodos de recogida: estudio de documentos, observaciones, cuestionarios y entrevistas. En los resul-tados se concluye que las habilidades mejor valoradas por los futuros docentes son las relacionadas con la interpreta-ción musical y las actitudes personales. Tanto los docentes en formación como sus formadores reconocen como figura fundamental la desempe ada por los tutores en los centros de prácticas, quienes dan soporte y ejemplo a los alumnos en prácticas. La relación entre las competencias profesionales y la creatividad docente presenta cierta controversia, puesto que las competencias se valoran como demasiado restrictivas y poco relacionadas con la especialidad de los docentes.
Media Art Wiki. Uso de Wikis para la ense anza interdisciplinar y multimedia del arte de los nuevos medios de comunicación en entornos virtuales de aprendizaje

Abstract: RESUMEN Este capítulo expone y analiza una experiencia piloto de uso de Wikis para la ense anza de las prácticas artísticas vinculadas a los nuevos medios de comunicación digital. El denominado “Media Art Wiki” constituye un recurso para la ense anza del Arte y la Estética de los nuevos medios en un contexto de ense anza-aprendizaje virtual como es la Universitat Oberta de Catalunya (UOC). Los objetivos iniciales del proyecto consistieron en generar un entorno dinámico de documentación multimedia de las prácticas de Arte de los nuevos medios, y a su vez, que ese espacio de contenidos abiertos pudiera alimentarse de la acción participativa y colaborativa de los estudiantes de las diferentes asignaturas implicadas, contribuyendo de este modo a un enfoque interdisciplinar de la materia común. El Media Art Wiki pretende innovar dentro del aula virtual en cuatro aspectos diferentes: proceso, formato, estructura y contenidos tratando de responder asimismo a un planteamiento pedagógico, que vincule cada una de las actividades implicadas en el Wiki con varios programas y asignaturas. En este capítulo intentaremos dar cuenta de las implicaciones del uso de este tipo de herramientas en la ense anza universitaria, estructurando el texto en diferentes apartados que, por un lado, trazan el contexto a partir del cual surgió el proyecto, sus necesidades, objetivos y motivaciones dentro del marco teórico del aprendizaje significativo- y el contexto -modelo de ense anza-aprendizaje- de la UOC. Por otro lado procedemos a la descripción técnica y de contenido del proyecto, así como el dise o de la estrategia didáctica y de las actividades a desarrollar por los estudiantes a corto y medio plazo. Finalmente exponemos la evaluación de dicha experiencia desde la perspectiva de los docentes y los estudiantes para llegar a plantear nuestras conclusiones y posibilidades de desarrollos de futuro. Abstract: in this study we analize the application of the reflective learning during initial formation mathematics teachers. this model is based on the sociocultural theories of the human learning and assumes that the interaction and the contrast make possible the coconstruction and the active reconstruction of knowledge. in order to make the study, it was left from a sample of 29 teaching students. the qualitative analysis allowed to identify factors that facilitate the incorporation of the reflective learning in university teaching, as well as the degree of effectiveness of this model to learn to teach mathematics.

Abstract: uruguay shares a long border with brazil. there, inhabitants from both sides of the border shop in whichever country they find the cheapest prices. as a consequence of severe changes in the bilateral real exchange rate, the demand moves alternately from side to the other of the border, affecting the level of employment in two uruguayan border provinces. the theoretical model outlined in this article intends to capture the main elements of the analyzed situation. the econometric estimates carried out validate the stated model and show that the labor markets on both sides of the border are not integrated enough to smooth the impact of demand movements.

Abstract: two new species of melitoma lepeletier and serville are described: m. fulviscopis n. sp. from matto grosso, brazil, and m. torquata n. sp. from southeastern and southern brazil, and northern argentina. the males have a peculiar hind basitarsus, unique among emphorine bees, that suggests a close relationship of the two species. the basitarsus bears, on the inner surface, a longitudinal channel-like polished depression fringed with long hairs.

Abstract: a revision of the bee genus nomada scopoli in argentina is presented. nine species are recognized in this region, five of which are described as new: n. mesopotamica, n. longula, n. chacoana, n. missionica, and n. turrigera. lectotypes are designated for n. pampicola holmberg, 1886, and n. costalis brèthes, 1909. a new name, n. holmbergiana, is proposed for hypochrotaenia parvula holmberg, 1886, preoccupied in nomada, and a neotype is designated for h. parvula holmberg. a key to the species, descriptions, distributional data, and illustrations are provided.

Abstract: the albidohirta group of ptilothrix smith includes three species: ptilothrix albidohirta brèthes, p. concolor sp. nov., and p. nemoralis sp. nov. the group is known from northern and western argentina and from central bolivia. one of its species, p. albidohirta, reaches high altitudes in the andes mountains, up to 3600 m a.s.l., while the other two species occur in the lowlands. a characterization of the group, descriptions of the species, and illustrations are provided.
