Genetic control of resistance to salmonellosis and to Salmonella carrier-state in fowl: a review

Abstract: Salmonellosis is a zoonotic disease caused by the Gram-negative enteric bacterium Salmonella. More than 2500 serotypes have been described, mostly belonging to the species S. enterica [1]. Some Salmonella serotypes can infect a broad range of domestic animals including poultry, sheep, cattle and pigs and cause symptoms of varying severity ranging from mild gastro-enteritis to death. Some of these serotypes, such as S. Typhimurium and S. Enteritidis, can infect humans. Other serotypes are host-specific, infecting a single species and generally causing severe, typhoid-like symptoms sometimes leading to death (for instance, S. Gallinarum and S. Pullorum in poultry). These serotypes can be responsible for disease outbreaks leading to severe economic losses.Prophylactic measures, vaccination and use of antibiotics are insufficient to eradicate salmonellosis in poultry stocks, whatever the serotype involved. In this context, selection of more resistant chickens can be considered as an alternative solution to decrease occurrence of the disease. The first selection experiments at the beginning of the 20th century aimed to decrease disease occurrence in poultry production systems, which was mainly caused by S. Pullorum and S. Gallinarum. As food safety became an important concern and these host-specific serotypes were better controlled, the interest of researchers and breeders extended towards decreasing food contamination, mainly due to the serotypes Enteritidis and Typhimurium. S. Enteritidis alone, which infects the eggs of contaminated hens, is responsible for one third of the human food poisoning cases in France [2] and of about 15% in the UK in 2007 http:/ / www.defra.gov.uk/ foodfarm/ farmanimal/ diseases/ atoz/ zoonoses/ reports.htm webcite. It does not cause severe symptoms in poultry, but the eggs and meat of infected animals can become a reservoir of infection for the human consumer. Abstract: Since feed conversion ratio (FCR) is higher in slow-growing "Label Rouge" chickens than in broiler chickens, it is important to work on its improvement in this breed. However, this involves rearing animals in cages (C), an environment very different from that used for selection (in floor pens, S) and production (outdoor, E). The aim of this study was to evaluate the importance of genotype by environment (G × E) interactions between S, C, and E environments, to find the best way to select for FCR, using 2002 related animals. Growth curve parameters were estimated and body composition measured. Individual feed conversion ratios (FCR) were recorded between 8 and 10 weeks in C. The presence of G × E interactions was assessed by the genetic correlations between the same trait recorded in different environments. Moderate but significant G × E interactions were detected for carcass traits, a significant one was observed between E and S or C for growth curve parameters but none between C and S. If G × E interactions are set aside, i.e. selecting on traits recorded in C, abdominal fatness is the best indirect selection criterion for FCR but if they are taken in account then leg yield or growth curve parameters in S and growth curve parameters in E are better.

Abstract: In our case the QTL analysis method had an important impact on QTL detection. We were able to identify new genomic regions controlling resistance to carrier-state, in particular by testing the existence of two segregating QTLs. But some of the previously identified QTLs were not confirmed. Interestingly, two QTLs were detected on chromosomes 2 and 3, close to the locations of the major QTLs controlling resistance to colonization and to candidate genes involved in the immune response identified in other, independent studies.Due to the lack of stability of the QTLs detected, we suggest that interesting regions for further studies are those that were identified in several independent studies, which is the case of the QTL regions on chromosomes 2 and 3, involved in resistance to both Salmonella colonization and carrier state. These observations provide evidence of common genes controlling S. Typhimurium colonization and S. Enteritidis carrier-state in chickens.

Abstract: A bilogarithmic hyperbolic cosine method for the evaluation of overlapping formation constants at varying (or fixed) ionic strength is devised in this paper and applied to data reported in the analytical literature, i.e. succinic acid system, Cu(II)-glycine system and Ag(I)-aminobutan-1-ol system. The method is based on the linearization of the formation function ? = f(pH) or ? = f(pL) data. A theoretical slope of unity should be obtained thus proving the correctness of the assumed equilibria. An additional advantage of the bilogarithmic method proposed is that it provides a closed scale representation of Y and X unlike other plots. This paper forms part of an investigation into the uses of bilogarithmic methods and hyperbolic functions in parameter estimation. Methods based on the application of spectrophotometric measurements have been the subject of recent studies.
