Abstract: We present a patient with corticoid resistant nephrotic syndrome who developed marked juxtaglomerular apparatus hyperplasia and renin expression in the context of dual angiotensin system blockade.Although renin may have profibrotic effects mediated by (pro)renin receptor activation, this report raises questions on the potential consequences of local renin activation on chronic kidney disease in patients with dual angiotensin blockade.
Osteoprotegerin in Exosome-Like Vesicles from Human Cultured Tubular Cells and Urine

Abstract: Urinary exosomes have been proposed as potential diagnostic tools. TNF superfamily cytokines and receptors may be present in exosomes and are expressed by proximal tubular cells. We have now studied the expression of selected TNF superfamily proteins in exosome-like vesicles from cultured human proximal tubular cells and human urine and have identified additional proteins in these vesicles by LC-MS/MS proteomics. Human proximal tubular cells constitutively released exosome-like vesicles that did not contain the TNF superfamily cytokines TRAIL or TWEAK. However, exosome-like vesicles contained osteoprotegerin (OPG), a TNF receptor superfamily protein, as assessed by Western blot, ELISA or selected reaction monitoring by nLC-(QQQ)MS/MS. Twenty-one additional proteins were identified in tubular cell exosome-like vesicles, including one (vitamin D binding protein) that had not been previously reported in exosome-like vesicles. Twelve were extracellular matrix proteins, including the basement membrane proteins type IV collagen, nidogen-1, agrin and fibulin-1. Urine from chronic kidney disease patients contained a higher amount of exosomal protein and exosomal OPG than urine from healthy volunteers. Specifically OPG was increased in autosomal dominant polycystic kidney disease urinary exosome-like vesicles and expressed by cystic epithelium in vivo. In conclusion, OPG is present in exosome-like vesicles secreted by proximal tubular epithelial cells and isolated from Chronic Kidney Disease urine.
Regulation of the Activity of the Dual-Function DnaA Protein in Caulobacter crescentus

Abstract: DnaA is a conserved essential bacterial protein that acts as the initiator of chromosomal replication as well as a master transcriptional regulator in Caulobacter crescentus. Thus, the intracellular levels of active DnaA need to be tightly regulated during the cell cycle. Our previous work suggested that DnaA may be regulated at the level of its activity by the replisome-associated protein HdaA. Here, we describe the construction of a mutant DnaA protein [DnaA(R357A)]. The R357 residue in the AAA+ domain of the C. crescentus DnaA protein is equivalent to the R334 residue of the E. coli DnaA protein, which is required for the Regulatory Inactivation of DnaA (RIDA). We found that the expression of the DnaA(R357A) mutant protein in C. crescentus, but not the expression of the wild-type DnaA protein at similar levels, causes a severe phenotype of over-initiation of chromosomal replication and that it blocks cell division. Thus, the mutant DnaA(R357A) protein is hyper-active to promote the initiation of DNA replication, compared to the wild-type DnaA protein. DnaA(R357A) could not replace DnaA in vivo, indicating that the switch in DnaA activity once chromosomal replication has started may be an essential process in C. crescentus. We propose that the inactivation of DnaA is the main mechanism ensuring that chromosomal replication starts only once per cell cycle. We further observed that the R357A substitution in DnaA does not promote the activity of DnaA as a direct transcriptional activator of four important genes, encoding HdaA, the GcrA master cell cycle regulator, the FtsZ cell division protein and the MipZ spatial regulator of cell division. Thus, the AAA+ domain of DnaA may play a role in temporally regulating the bifunctionality of DnaA by reallocating DnaA molecules from initiating DNA replication to transcribing genes within the unique DnaA regulon of C. crescentus.

Abstract: We describe the Gevrey series solutions at singular points of the irregular hypergeometric system (GKZ system) associated with an affine monomial curve. We also describe the irregularity complex of such a system with respect to its singular support. We use restriction and some results in D-module theory to reduce our study to the two dimensional case.

Automatic Taxonomy Extraction from Query Logs with no Additional Sources of Information

Abstract: Search engine logs store detailed information on Web users interactions. Thus, as more and more people use search engines on a daily basis, important trails of users common knowledge are being recorded in those files. Previous research has shown that it is possible to extract concept taxonomies from full text documents, while other scholars have proposed methods to obtain similar queries from query logs. We propose a mixture of both lines of research, that is, mining query logs not to find related queries nor query hierarchies, but actual term taxonomies that could be used to improve search engine effectiveness and efficiency. As a result, in this study we have developed a method that combines lexical heuristics with a supervised classification model to successfully extract hyponymy relations from specialization search patterns revealed from log missions, with no additional sources of information, and in a language independent way.

Abstract: Tenofovir is an acyclic nucleotide analogue reverse-transcriptase inhibitor structurally similar to the nephrotoxic drugs adefovir and cidofovir. Tenofovir is widely used to treat HIV infection and approved for treatment of hepatitis B virus. Despite initial cell culture and clinical trials results supporting the renal safety of tenofovir, its clinical use is associated with a low, albeit significant, risk of kidney injury. Proximal tubular cell secretion of tenofovir explains the accumulation of the drug in these mitochondria-rich cells. Tenofovir nephrotoxicity is characterized by proximal tubular cell dysfunction that may be associated with acute kidney injury or chronic kidney disease. Withdrawal of the drug leads to improvement of analytical parameters that may be partial. Understanding the risk factors for nephrotoxicity and regular monitoring of proximal tubular dysfunction and serum creatinine in high-risk patients is required to minimize nephrotoxicity. Newer, structurally similar molecular derivatives that do not accumulate in proximal tubules are under study. 1. Tenofovir Tenofovir disoproxil fumarate is an orally bioavailable prodrug of tenofovir, an acyclic nucleotide analogue reverse-transcriptase inhibitor (NtRTI) structurally similar to adefovir and cidofovir [1] (Figure 1). Acyclic nucleotides differ in their side chains: hydroxy phosphonomethoxypropyl (HPMP) for cidofovir, phosphonomethoxyethyl (PME) for adefovir and phosphonomethoxypropyl (PMP) for tenofovir [2]. Tenofovir diphosphate is a structural analog of deoxyadenosine-5′-triphosphate, the usual substrate for viral RNA-directed DNA polymerase, and is a weak inhibitor of mammalian DNA α- and β-polymerases and mitochondrial DNA γ-polymerase [3]. A lesser uptake by proximal tubular cells can be achieved by either esterifying the compounds with an alkoxyalkyl group, in effect disguising them as lysophospholipids (hexadeciloxypropyl-tenofovir, CMX157) or by ribose-modification (GS-9148 and its oral prodrug GS-9131). Tenofovir was the first (2001), and remains the only, NtRTI approved by the US Food and Drug Administration (FDA) for the treatment of HIV infection [1]. Irregular Modified $A$-Hypergeometric Systems

Abstract: A modified $A$-hypergeometric system is a system of differential equations for the function $f(t^w \cdot x)$ where $f(y)$ is a solution of an $A$-hypergeometric system in $n$ variables and $w$ is an $n$ dimensional integer vector, which is called the weight vector. We study the irregularity of modified systems by adapting to this case the notion of umbrella introduced by M. Schulze and U. Walther. Especially, we study slopes and Gevrey series solutions. We develop some applications of this study. Under some conditions we give Laplace integral representations of divergent series solutions of the modified system and we show that certain Gevrey series solutions of the original $A$-hypergeometric system along coordinate varieties are Gevrey asymptotic expansions of holomorphic solutions of the $A$-hypergeometric system.
Remission of HIV-associated myelopathy after highly active antiretroviral therapy

Abstract: HIV-associated myelopathy is the leading cause of spinal cord disease in HIV-infected patients. Typically, it affects individuals with low CD4 T cell counts, presenting with slowly progressive spastic paraparesis associated with dorsal column sensory loss as well as urinary disturbances. Other aetiologies must be first ruled out before establishing the diagnosis. We report here the case of a 37-year-old woman with advanced HIV disease, who developed HIV-associated myelopathy. The patient showed a gradual improvement after beginning with highly active antiretroviral therapy and, finally, she achieved a complete functional recovery. In addition, neuroimaging and neurophysiological tests normalized.

Abstract: El artículo presenta resultados de una investigación sobre la constitución de algo como un deber ser femenino en las prescripciones de la revista Boa Forma. Se analizó sobretodo los editoriales, secciones y reportajes sobre los cuidados con el cuerpo, tomando como fuentes principales las ediciones de verano y las que para ello preparan (setiembre-febrero, 2001, 2006). Los resultados sugieren la existencia de un aparato de control que está centrado en el uso de discursos ambiguos; en el escuadrinamiento del cuerpo; en la valoración de la disciplina y del sacrificio; en el uso de métodos de legitimación orientados hacia los mecanismos de la industria cultural.
