CenH3/CID Incorporation Is Not Dependent on the Chromatin Assembly Factor CHD1 in Drosophila

Abstract: CHD1 is a SNF2-related ATPase that is required for the genome-wide incorporation of variant histone H3.3 in the paternal pronucleus as well as in transcriptionally active nuclei in Drosophila embryos. The S. pombe and vertebrate orthologs of CHD1 have been implicated in the assembly of the centromeric histone H3 variant CenH3CENP-A, which occurs in a DNA replication-independent manner. Here, we examined whether CHD1 participates in the assembly of CenH3CID in Drosophila. In contrast to the findings in fission yeast and vertebrate cells, our evidence clearly argues against such a role for CHD1 in Drosophila. CHD1 does not localize to centromeres in either S2 cells or developing fly embryos. Down-regulation of CHD1 in S2 cells by RNAi reveals unchanged levels of CenH3CID at the centromeres. Most notably, ablation of functional CHD1 in Chd1 mutant fly embryos does not interfere with centromere and kinetochore assembly, as the levels and localization of CenH3CID, CENP-C and BubR1 in the mutant embryos remain similar to those seen in wild-type embryos. These results indicate that Drosophila CHD1 has no direct function in the incorporation of the centromeric H3 variant CenH3CID into chromatin. Therefore, centromeric chromatin assembly may involve different mechanisms in different organisms.

Abstract: El artículo presenta resultados de una investigación sobre la constitución de algo como un deber ser femenino en las prescripciones de la revista Boa Forma. Se analizó sobretodo los editoriales, secciones y reportajes sobre los cuidados con el cuerpo, tomando como fuentes principales las ediciones de verano y las que para ello preparan (setiembre-febrero, 2001, 2006). Los resultados sugieren la existencia de un aparato de control que está centrado en el uso de discursos ambiguos; en el escuadrinamiento del cuerpo; en la valoración de la disciplina y del sacrificio; en el uso de métodos de legitimación orientados hacia los mecanismos de la industria cultural.
Effectiveness of a course in the control of tobacco use for university health students: A quasi-experimental community intervention trial  [PDF]

Abstract:

Introduction: The aim of the study was to evaluate the effectiveness of a tobacco control course in the reduction of prevalence and the improvement of knowledge and attitudes among university health students. Method: Quasi-experimental study on a community based intervention. Interventional campus students received a tobacco control course in the first year of the degree. Data were collected by Self-Answered Questionnaire during three academic years in two graduations years among health students in both campuses. Results: 84.4% of students participated in the study. In both campuses tobacco use was reduced, not significantly, above all in those joining courses from origins other than baccalaureate. In the intervention and control campus showed an increase in the nicotine dependency according to Fagerstr?m test and a little change in motivation according toRichmondtest. The regression model best explaining the improving of the knowledge included academic course mainly and campus and academic background too. With regard to opinions and beliefs tobacco use was not influenced, being associated in general to the academic course. Conclusions: A specific tobacco control course did not change prevalence of tobacco, nicotine dependence and motivation to give up. The improvement in knowledge was associated with academic course in both campuses. We recommend to commence smoking cessation strategies in university health students.

Transcription profiles of non-immortalized breast cancer cell lines

Abstract: Cultures of cells from pleural effusions or ascitic fluids from breast cancer patients (MSSMs) were used in addition to commercially-available normal breast epithelial cells (HMECs), established breast cancer cell lines (T-est) and established normal breast cells (N-est). The Atlas Human Cancer 1.2 cDNA expression array was employed. The data obtained were analysed using widely-available statistical and clustering software and further validated through real-time PCR.According to Significance Analysis of Microarray (SAM) and AtlasImage software, 48 genes differed at least 2-fold in adjusted intensities between HMECs and MSSMs (p < 0.01). Some of these genes have already been directly linked with breast cancer, metastasis and malignant progression, whilst others encode receptors linked to signal transduction pathways or are otherwise related to cell proliferation. Fifty genes showed at least a 2.5-fold difference between MSSMs and T-est cells according to AtlasImage, 2-fold according to SAM. Most of these classified as genes related to metabolism and cell communication.The expression profiles of 1176 genes were determined in finite life-span cultures of metastatic breast cancer cells and of normal breast cells. Significant differences were detected between the finite life-span breast cancer cell cultures and the established breast cancer cell lines. These data suggest caution in extrapolating information from established lines for application to clinical cancer research.The search for differentially expressed genes in tumours has made extensive use of array technology. Most studies have involved tumour biopsy samples or established tumour-derived cell lines [1]. Differentially expressed genes may help to identify tumours with high metastatic potential and pathways that might be therapeutic targets.As noted by Dairkee et al. Abstract: this article presents results of a research that sought to investigate aspects of bodily education, focusing in a program of school sports for boys and girls from a public school in florianópolis, brazil. systematic observations were made of the contexts of training sessions and competitions, apart from discursive interviews and some data gathering about the institution. results were organized in three major analysis categories. the first of them was called punishments, penalties and sacrifices. it relates to the way found by coaches to deal with the dynamics of reprehension and encouragement of their teams. the second category, entitled human formation versus the dream of professionalization, describes the role of representations and expectations about football (soccer) for children, in contrast to the formation that school and sport promise to afford them. the choice of the third category was a consequence of the constant presence of rituals in the organized bodily practices, with special attention here to sport. it was denominated rituals as technique. the final considerations of the article suggest an interpenetration of rituals, future expectations and moralist questions involved in sport, pointing to the prevalence of interests of control. additionally, they suggest the mimetism of the conventional sport practices with the mediation of the culture industry.
Planificación de la información y comunicación en centros e institutos de investigación: situación en la universidad del Zulia de la Republica Bolivariana de Venezuela.

Abstract: La información y comunicación se han convertido en herramientas esenciales de la actividad humana. En el caso específico de los centros e institutos de investigación, coadyuvan a interrelacionar e integrar la generación de conocimiento, acorde a las exigencias del entorno. De allí que el presente trabajo tuvo como principal objetivo el diagnosticar la planificación de la información y comunicación generada en los centros e institutos de investigación que aborden las ciencias sociales en la Universidad del Zulia, 2005-2006. Para la misma, se requirió de la identificación y determinación de elementos básicos de la planificación como, la filosofía de gestión, políticas y estrategias aplicadas en relación a la información y comunicación, entre otros.La metodología utilizada se basó en un tipo de investigación descriptivo y un dise o de investigación no experimental-transeccional, abarcándose todos los centros e institutos de investigación que abordan las ciencias sociales en la Universidad del Zulia, Núcleo Maracaibo. Entre las conclusiones obtenidas, se encuentran que existe una planificación de la información y comunicación básica, encontrando como limitantes la poca disponibilidad de tecnología innovadora e infraestructura suficiente para el eficaz funcionamiento de los entes de investigación.
Negative Feedback and Transcriptional Overshooting in a Regulatory Network for Horizontal Gene Transfer

Abstract: Horizontal gene transfer (HGT) is a major force driving bacterial evolution. Because of their ability to cross inter-species barriers, bacterial plasmids are essential agents for HGT. This ability, however, poses specific requisites on plasmid physiology, in particular the need to overcome a multilevel selection process with opposing demands. We analyzed the transcriptional network of plasmid R388, one of the most promiscuous plasmids in Proteobacteria. Transcriptional analysis by fluorescence expression profiling and quantitative PCR revealed a regulatory network controlled by six transcriptional repressors. The regulatory network relied on strong promoters, which were tightly repressed in negative feedback loops. Computational simulations and theoretical analysis indicated that this architecture would show a transcriptional burst after plasmid conjugation, linking the magnitude of the feedback gain with the intensity of the transcriptional burst. Experimental analysis showed that transcriptional overshooting occurred when the plasmid invaded a new population of susceptible cells. We propose that transcriptional overshooting allows genome rebooting after horizontal gene transfer, and might have an adaptive role in overcoming the opposing demands of multilevel selection.
Morfometría fluvial aplicada a una cuenca urbana en Ingeniero White, República Argentina

Abstract: the fluvial morphometry studies several variables that characterize the form of a drainage basin. the aim of this paper is to apply the natural watershed morphometric method to an urban watershed. the study area (6 km2) drains its waters into the waterfront and includes the town of ingeniero white, república argentina. hierarchical organization of the drainage network was done. three sub-basins that have low-gradient flood events, most dangerous extreme rainfall, were identified. the length relationship values showed greater concentration of power in event of major avenues. the elongation relationship values showed that one of the sub-basins is circular and has more speed in the avenues of runoff. the two elongated basins have greater delay in the avenues. the increased flow of the study area flows through the basin that has the largest number of order and the largest area. the other two basins are subject to increasing surface snapshot and immediate response. the slope and area values applied to hydrologic and hydraulic models determined the section of the ducts and channels. the calculated flow for the area drained was 22.41 m3/s.
Analisis geométrico y legal de la traza de "La Trochita" patrimonio histórico y cultural de la Comarca Serrana

Abstract: the recovering of "la trochita" original railway trail is a joint project with the city hall of tornquist. the current train station of sierra de la ventana and club hotel de la ventana were communicated through the 18 km narrow railway trail. the club hotel de la ventana, a former world-wide known hotel, was built in the early xx century. today the ruins of the hotel are part of the local history of the comarca serrana, along with the bridges, sewer systems, embankments - all of them built along the railway trail. the phrase "comarca serrana" refers to a popular touristic destination in the sistema de ventania located at the south of the province of buenos aires - between 38° 4′ and 38° 8′ south latitude, and 61° 47′ and 61°56′ west longitude - that comprises the small villages of sierra de la ventana and villa ventana, both within the city hall of tornquist. this research is intended to recover the original railway trail to organize a thematic tour. for this, the identification and analysis of the graphic and analytical background was organized, along with the survey of the area. the specific goal of this study is the analysis of the property rights of the lands through which the original railway trail runs. this research project aims at assisting people living in this area in understanding the importance of this cultural heritage and identifying the need to preserve it for promoting tourism in this region.

Abstract: the objective of this work was to evaluate the effect of cabbage and kale cultivars on the diamondback moth biology; to analyze the plant chemical composition in relation to glucosinolates and to apply the multivariate analysis to classify cultivar resistance. trials were conduct using: green cabbage cultivar chato-de-quintal and hybrid midori; purple cabbage roxo precoce and purple hybrid tpc00682; kale cultivar georgia (susceptibility standard) and hybrid hs20. the pest biological characteristics evaluated were: viability and the length of the larval and pupal period, sex ratio, female fertility, adult longevity and the length and viability of the egg phase; also it was calculated the corrected reproductive potential index (crp). pearson correlation was obtained, and the grouping and main components of multivariate analysis were applied. sinigrin and other glucosinolates were evaluated by liquid chromatography. 'georgia', 'roxo' and hs20 hybrids were classified as highly susceptible; 'roxo precoce' and midori hybrid, as susceptible, and 'chato-de-quintal' as moderately resistant to diamondback moth. multivariate analyses provided a better way to classify cultivars in relation to different resistance degrees. brassica genetic materials evaluated do not present sinigrin.
