FORMULATION AND EVALUATION OF SUSTAINED RELEASE PELLETS OF TRAMADOL HYDROCHLORIDE

Abstract: The aim of the present research is to develop and evaluate a better sustained release multiple unit pellets (MUP) formulation of Tramadol hydrochloride. Dissolution and diffusion controlled systems have classically been of primary importance in oral delivery of medication because of their relative ease of production and cost compared with other methods of sustained or controlled delivery. Most of these systems are solids, although a few liquids and suspension have been recently introduced. The present work aimed at developing SR pellets of Tramadol HCl by Wurster process. FTIR studies showed no unacceptable extra peaks which confirm the absence of chemical interaction between the drug and polymer. Angle of repose, tapped density, bulk density values for the formulations were within the range which indicates that pellets prepared by Wurster process were satisfactory for further studies. The percentage drug content of Tramadol was determined by extraction with methanol and analyzed by using UV-visible spectrophotometer at 271nm.

Abstract: The persistent market failures and policy inertia due to the existence of carbon lock-in create barriers to the diffusion of carbon saving technologies. In spite of their apparent environmental and technological advantages, the renewable technologies cannot take off in the market. On an empirical level this paper studies the barriers to the market deployment of indigenously developed ArunTM 160 Solar Concentrator technology which has the potential to revolutionize the industrial restructuring by providing Solar Heat for Industrial Processes. This study classifies the barriers into micro, meso and macro barriers and analyses the impact of them by conducting two rounds of questionnaire: one with firms and other with experts.

Abstract:

This paper presents a new logical mechanism called as Cluster Based Hierarchical Routing (CBHR) to improve the efficiency of NoC. This algorithm comprises the following steps: 1) the network is segmented logically into clusters with same size or different sizes; 2) algorithms are assigned for internal and global routing; 3) routers working functions are modified logically to support local and global communication. The experiments have conducted for CBHR algorithm for two dimensional mesh and torus architectures. The performance of this mechanism is analyzed and compared with other deterministic and adaptive routing algorithms in terms of energy, throughput with different packet injection ratios.

Hydrodynamic study on gasification of biomass in a fluidized bed gasifier

Abstract: Current scenario of energy insecurity urges us to realize the importance of alternate energy sources. In country with variety of vegetation like India, Biomass finds its place of which fluidized bed gasification of biomass could be more effective. This paper emphasizes the importance of a fluidized bed gasifier for energy conversion of agro-residues for useful purposes. Coconut Shell and Ground nut shell of gross calorific value 19.43MJ/kg and 14.91 MJ/kg respectively are taken for the study. The particle size is restricted not to exceed 3mm. Various empirical correlations involved in fluidization are studied and their interdependence is detailed. From various published data, importance of inert materials and their relative proportions with biomass fuels are studied and optimum biomass to sand ratio is fixed as 10 to 15% by mass. Equations for predicting the minimum fluidization velocities of these mixtures are also discussed. Variations of Fluidization parameters such asminimum fluidization velocity, bubble rise velocity, expanded bed height with respect to temperature, equivalence ratio, particle size is studied and their quantification is analyzed. A 108 mm internal diameter and 1400 mm high FBG is used for the study. Fuel is fed through screw feeder and air is supplied through blower. In the down stream side cyclone separator is placed after which the sampling and burner lines are connected. A regression model is developed and the feasibility of gasifying coconut shell and groundnut shell are discussed. Earlier and present work of coconut shell gasification proves fluidized bed gasification is more appropriate for agro residues.
Effects of Chemical and Physical Enhancement Techniques on Transdermal Delivery of Cyanocobalamin (Vitamin B12) In Vitro

Abstract: Vitamin B12 deficiency, which may result in anemia and nerve damage if left untreated, is currently treated by administration of cyanocobalamin via oral or intramuscular routes. However, these routes are associated with absorption and compliance issues which have prompted us to investigate skin as an alternative site of administration. Delivery through skin, however, is restricted to small and moderately lipophilic molecules due to the outermost barrier, the stratum corneum (SC). In this study, we have investigated the effect of different enhancement techniques, chemical enhancers (ethanol, oleic acid, propylene glycol), iontophoresis (anodal iontophoresis) and microneedles (soluble maltose microneedles), which may overcome this barrier and improve cyanocobalamin delivery. Studies with different chemical enhancer formulations indicated that ethanol and oleic acid decreased the lag time while propylene glycol based formulations increased the lag time. The formulation with ethanol (50%), oleic acid (10%) and propylene glycol (40%) showed the maximum improvement in delivery. Iontophoresis and microneedle treatments resulted in enhanced permeation levels compared to passive controls. These enhancement approaches can be explored further to develop alternative treatment regimens.
Herniation of the anterior lens capsule

Abstract: Herniation of the anterior lens capsule is a rare abnormality in which the capsule bulges forward in the pupillary area. This herniation can be mistaken for an anterior lenticonus where both the capsule and the cortex bulge forward. The exact pathology behind this finding is still unclear. We report the clinical, ultrasound biomicroscopy (UBM) and histopathological findings of a case of herniation of the anterior lens capsule. UBM helped to differentiate this entity from anterior lenticonus. Light microscopy revealed capsular splitting suggestive of capsular delamination and collection of fluid (aqueous) in the area of herniation giving it a characteristic appearance.

MODELING OF HUMAN CASPASE-5 PROTEIN AND DOCKING STUDIES OF CASPASE-5 INHIBITORS AGAINST MODELED CASPASE-5

Abstract: The present study is aimed at building human caspase-5 protein structure by homology modeling. Human interleukin-1 beta convertase P20 (1RWN) and human caspase-1 (3E4C) crystal structures from blast results were taken as caspase-5 structural relatives (from protein data bank). The former showed 58% and the latter, 57% sequence identity with the target caspase-5 protein. The 3D-model was generated by using modeler in Accelrys discovery studio 2.5. The energy refinement was also been carried out in the modeler protocol. The Ramachandran plot showed 89.1% of residues in the favorable region. Further, binding site analysis was done to decipher the interacting residues at the active site region of the modeled protein. Then, docking of Pyrazinone mono amide analogues, peptide aldehydes, substituted P4 aryl sulfonamides on a pyridine scaffold analogues, P1 aspartyl ketone derivatives on a pyridine scaffold analogues and a P3 sulfonamide-modified thiophene derivatives against modeled human caspase-5 protein were performed using ligandFit. The results showed that Cys161, Arg55, Val214 and His113 were found to be among the interacting residues of caspase-5 with all the types of inhibitors. These 4 residues of caspase-5 active site were found to be interacting with the crystal ligand (copied from 1RWN) also. The dock scores of P4 aryl sulfonamides on pyridine scaffold analogues and P1 Aspartyl Ketone derivatives on Pyridine Scaffold analogues were higher indicating their high activity against caspase-5 compared to that of pyrazinone monoamide analogues and tetra peptides
