Abstract: the organization for tropical studies (ots)/organización para estudios tropicales (oet) has evolved in many ways since its founding in 1963 as a non-profit consortium offering graduate courses and facilitating research in tropical ecology in costa rica. by 2002, its international membership included about 65 institutions, including four from costa rica. it had developed three costa rican field stations (la selva, las cruces, and palo verde) with excellent facilities for teaching and research, and it was constructing a new costa rican office at the university of costa rica. combinations of internal and external pressures influenced ots to develop in new directions in the 1980s and 1990s. it became more diversified and more concerned with applied science in its traditional areas of graduate education and research facilitation. the organization also evolved into new niches: more applied biology, professional education, environmental education and policy, conservation efforts, and an expanded geographic distribution to other latin american countries. ots was composed of changing combinations of people (boards, members, staff) with evolving and competing priorities for limited financial resources. external environmental changes also shaped ots?s evolution. new problems of increased tropical deforestation, the emergence of the biodiversity "crisis" and conservation biology, global climate change, and calls for sustainable development affected ots constituents and funding priorities of governments and foundations. both internal and external pressures have in some cases demanded for ots to improve its relationship with: costa rican biologists and their institutions, the costa rican government, and costa ricans living around the three ots field stations

A GIS Pre and Post Processing Contribution to a Multi Techniques Test Survey for an Ancient Railway Restoration in the Brembana Valley  [PDF]

Abstract: Stemming from the renewed desire to requalify the former Brembana Valley railway route (inside one of the valleys of the Province of Bergamo and close to the important city of Milan), from Bergamo to Villa d’Almè, in 2017, the Bergamo Transport Company and other partners have signed a memorandum of understanding. They have also entrusted a project company to develop a technical and economic viability study, which would be an update of a preliminary one presented in 2009. A private proposal for the realization of a test survey, useful for the required engineering project, has been submitted to the project company and accepted. For this reason, a team, comprising different companies, and among them also the Geomatics group at the University of Bergamo, was formed so as to perform this task and develop it in a degree thesis. The aim of the test survey is to evaluate a joint use of the new available technologies, so as to possibly get faster, more economic and thorough results, by performing a 3D metric analysis over two sub-areas selected along the route. The paper describes this experience and the achieved results. In particular, the areas of interest in the test project have been analyzed first thing with a GIS software and then surveyed with a multi-sensor approach, i.e. terrestrial laser scanner, GNSS techniques and UAV. The final integrated results allowed providing a metric model of the morphological and urban aspect for the territorial layout, useful also for further advanced GIS analyses. The analysis of the reached precisions has provided satisfactory results which agree with the metric requirements of the project. Further meaningful considerations can be derived from this test survey: the implementation of different techniques has helped to overcome problems due to hidden parts in complex objects, it has allowed to provide a faster survey and to test different technologies and software packages. The results have been quite satisfactory.
Primary antiphospholipid syndrome evolving into systemic lupus erythematosus: may antinucleosome antibodies be predictive?

Abstract: Objective: It was reported by several groups that patients diagnosed as primary antiphospholipid syndrome (PAPS) had developed a full-blown systemic lupus erythematosus (SLE) even after many years of follow-up. Little is known about clinical and/or serological factors that may help predict such evolution. Antinucleosome antibodies (anti-NCS) were described to appear in early stages of SLE, in particular before anti-dsDNA antibodies. The aim of the study is to evaluate the prevalence of anti-NCS in a large cohort of PAPS patients. Methods: IgG and IgM anti-NCS antibodies were detected using a home made assay with H1-stripped chromatin as antigen. Sera from 106 PAPS patients were tested; 52 of them were also tested during the follow-up, at least 2 years apart form the basal sample. Results: Medium-high titre anti-NCS were found in nearly half of the patients (49/106, 46%), more frequently in those presenting features of “lupus like disease”. Most of patients displayed an unchanged pattern of anti-NCS over time. We describe three cases of PAPS patients that developed SLE after many years of follow-up; high titre and low titre anti-NCS were present in two and one of them respectively several years before evolving into SLE. Conclusions: A significant proportion of PAPS patients displayed medium-high titre anti-NCS, suggesting that the autoimmune response against chromatin may be a relevant event not only in patients with SLE. Further studies are warranted to explore the predictive value of anti-NCS with respect to the evolution from PAPS to SLE.
A Minimal Anaphase Promoting Complex/Cyclosome (APC/C) in Trypanosoma brucei

Abstract: The anaphase-promoting complex/cyclosome (APC/C) is a multi-subunit E3 ubiquitin ligase that initiates chromosome segregation and mitotic exit by targeting critical cell-cycle regulators for proteolytic destruction. Previously, seven APC/C subunit homologues were identified in the genome of Trypanosoma brucei. In the present study, we tested five of them in yeast complementation studies and found none of them capable of complementing the yeast mutants lacking the corresponding subunits, suggesting significant discrepancies between the two APC/C’s. Subunit homologues of mitotic checkpoint complex (MCC) have not yet been identified in T. brucei, raising the possibility that a MCC-APC/C complex equivalent may not exist in T. brucei. We performed tandem affinity purification of the protein complex containing a APC1 fusion protein expressed in the cells enriched in different phases of the cell cycle of procyclic form T. brucei, and compared their protein profiles using LC-MS/MS analyses. The seven putative APC/C subunits were identified in the protein complex throughout the cell cycle together with three additional proteins designated the associated proteins (AP) AP1, AP2 and AP3. Abundance of the 10 proteins remained relatively unchanged throughout the cell cycle, suggesting that they are the core subunits of APC/C. AP1 turned out to be a homologue of APC4. An RNAi knockdown of APC4 and AP3 showed no detectable cellular phenotype, whereas an AP2 knockdown enriched the cells in G2/M phase. The AP2-depleted cells showed stabilized mitotic cyclin B. An accumulation of poly-ubiquitinated cyclin B was indicated in the cells treated with the proteasome inhibitor MG132, demonstrating the involvement of proteasome in degrading poly-ubiquitinated cyclin B. In all, a 10-subunit APC/C machinery with a conserved function is identified in T. brucei without linking to a MCC-like complex, thus indicating a unique T. brucei APC/C.
Eliciting Guilty Feelings: A Preliminary Study Differentiating Deontological and Altruistic Guilt  [PDF]

Abstract: Guilt has been identified as both an intrapsychic and an interpersonal emotion. The current study presents evidence of the existence of two senses of guilt, deontological and altruistic guilt, induced through different experimental paradigms. Deontological guilt evolves from having slighted moral authority or norms, while altruistic guilt arises from selfish behavior and the distress of others. We hypothesize that specific stimuli would evoke, separately, deontological guilt and altruistic/interpersonal guilt feelings. Two different procedures were used to test our hypothesis, adding two emotions as control conditions (i.e. anger and sadness). Results clearly indicate that two different guilt emotions can be evoked separately, by appropriate stimulation. Findings and possible clinical implications are discussed.

Abstract:

Gut Fermentation Syndrome also known as Auto-Brewery Syndrome is a relatively unknown phenomenon in modern medicine. Very few articles have been written on the syndrome and most of them are anecdotal. This article presents a case study of a 61 years old male with a well documented case of Gut Fermentation Syndrome verified with glucose and carbohydrate challenges. Stool cultures demonstrated the causative organism as Saccharomyces cerevisiae. The patient was treated with antifungals and a low carbohydrate diet and the syndrome resolved. Helicobacter pylori was also found and could have been a possible confounding variable although the symptoms resolved post-treatment of the S. cerevisiae.
