Hypothermic Machine Perfusion Reduces Delayed Graft Function and Improves One-Year Graft Survival of Kidneys from Expanded Criteria Donors: A Meta-Analysis

Abstract: Background Expanded criteria donors (ECDs) are currently accepted as potential sources to increase the donor pool and to provide more chances of kidney transplantation for elderly recipients who would not survive long waiting periods. Hypothermic machine perfusion (HMP) is designed to mitigate the deleterious effects of simple cold storage (CS) on the quality of preserved organs, particularly when the donor is in a marginal status. Methods We compared the transplant outcomes in patients receiving ECD kidneys with either HMP or CS graft preservation. Articles from the MEDLINE, EMBASE and Cochrane Library databases were searched and all studies reporting outcomes from HMP versus CS methods of kidney preservation were included in this meta-analysis. The parameters analyzed included the incidence of delayed graft function (DGF), primary non-function (PNF) and one-year graft and patient survival. Results A total of seven studies qualified for the review, involving 2374 and 8716 kidney grafts with HMP or CS preservation respectively, all from ECD donors. The incidence of delayed graft function (DGF) was significantly reduced with an odd ratio(OR) of 0.59 (95% CI 0.54–0.66, P<0.001) and one-year graft survival was significantly improved with an OR of 1.12 (95% CI 1.03–1.21, P = 0.005) in HMP preservation compared to CS. However, there was no difference in the incidence of PNF (OR 0.54, 95% CI 0.21–1.40, P = 0.20), and one-year patient survival (OR 0.98, 95% CI 0.94–1.02, P = 0.36) between HMP and CS preservation. Conclusions HMP was associated with a reduced incidence of DGF and an with increased one-year graft survival, but it was not associated with the incidence of PNF and one-year patient survival.

Green fluorescent protein (GFP) as a vital marker for studying the interaction of Phytophthora sojae and soybean

Abstract: Transgenic Phytophthora sojae strains that produce green fluorescent protein (GFP) were obtained after stable DNA integration using the Hsp70 promoter and the Ham34 terminator of Bremia lactucae. The expression of GFP during different developmental stages of P. sojae was observed using fluorescent microscopy. Based on this reporter system, the histopathologic events caused by the pathogen in soybean leaves, hypocotyls and roots were monitored. Meanwhile, the difference in resistance between different soybean cultivars against P. sojae was analyzed microscopically in roots. The results indicate that GFP can be stably expressed in zoosporangia, zoospores, cysts, hyphae and oospores of P. sojae. Using the GFP marker, the infecting pathogens in leaves, hypocotyls and roots of host could be distinctly visualized. The germ tube length of cysts germinating on the roots of resistant cultivar Nannong 8848 was longer than that on the roots of susceptible cultivar Hefeng 35. These results show for the first time that this eukaryotic reporter can be used in P. sojae as a stable and vital marker, allowing the study of genetics of this hemibiotrophic pathogen.

Abstract: Previous studies of precariously balanced objects in seismically active regions provide important information for aseismatic engineering and theoretical seismology. They are almost founded on an oversimplified assumption: any 3-dimensional (3D) actual object with special symmetry could be regarded as a 2D finite object in light of the corresponding symmetry. To gain an actual evolution of precariously balanced objects subjected to various levels of ground accelerations, a 3D investigation should be performed. In virtue of some reasonable works from a number of mechanicians, we derive three resultant second-order ordinary differential equations determine the evolution of 3D responses. The new dynamic analysis is following the 3D rotation of a rigid body around a fixed point. A computer program for numerical solution of these equations is also developed to simulate the rocking and rolling response of axisymmetric objects to various levels of ground accelerations. It is shown that the 2D and 3D estimates on the minimum overturning acceleration of a cylinder under the same sets of half- and full-sine-wave pulses are almost consistent except at several frequency bonds. However, we find that the 2D and 3D responses using the actual seismic excitation have distinct differences, especially to north-south (NS) and up-down (UD) components. In this work the chosen seismic wave is the El Centro recording of the 18 May 1940 Imperial Valley Earthquake. The 3D outcome does not seem to support the 2D previous result that the vertical component of the ground acceleration is less important than the horizontal ones. We conclude that the 2D dynamic modeling is not always reliable.

Abstract: Dynamic rupture propagation along an interface between two different elastic solids under shear dominated loading is studied numerically by a 2-D lattice particle model (LPM). The configuration of the lattice particle model consists of two solid blocks of different elastic properties connected along a planar interface. Each block is characterized as an isotropic elastic material and the interface strength is described as a composite elastic modulus of a mismatch function of the elastic properties of two dissimilar materials. The particle interaction between the two blocks with pair inter-particle potential also takes account of normal stress variations.
Global wellposedness of the equivariant Chern-Simons-Schr?dinger equation

Abstract: In this article we consider the initial value problem for the m-equivariant Chern-Simons-Schr\"odinger model in two spatial dimensions with real-valued coupling parameter g. This is a covariant NLS type problem that is L^2-critical. We prove that at the critical regularity, for any integer-valued equivariance index m, the initial value problem in the defocusing case (g < 1) is globally wellposed and the solution scatters. The problem is focusing when g >= 1, and in this case we prove that for nonnegative integer-valued equivariance indices m there exist constants c = c_{m, g} such that, at the critical regularity, the initial value problem is globally wellposed and the solution scatters when the L^2 initial data phi_0 is m-equivariant and has L^2-norm less than the square root of c_{m, g}. We also show that c_{m, g}^{1/2} is equal to the minimum L^2 norm of a nontrivial m-equivariant standing wave solution. In the self-dual g = 1 case, we have the exact numerical values c_{m, 1} = 8*pi*(m + 1).

Abstract: The functional formula diet AHP, containing polysaccharides from Auricularia auricular, polyphenolic compounds from Hawthorn (Crataegus) and Pueraria radix, has been recently developed as a dietary intervention against dyslipidemia in our previous study. In the present study, its antioxidant activities and protective effects against proliferation of vascular smooth muscle cells (VSMCs) were investigated. AHP possessed the potent radical-scavenging effects against hydroxyl and superoxide radicals, and also the inhibitory effects against peroxidation of low density lipoprotein induced by Cu2+ in vitro. The protective effects of AHP against proliferation of VSMCs were evaluated through the methodology of serum pharmacology. The serum containing AHP significantly inhibited the proliferation of VSMCs induced by oxidized low density lipoprotein in a time- and dose-dependent manner, and also promoted the nitric oxide production of VSMCs. Our study indicated that this functional formula diet would be a potent alternative as a functional diet to prevent atherosclerosis at early stage.
