Bactérias diazotróficas em mudas de bananeira

Abstract: the objective of this work was to select strains of diazotrophic bacteria associated with banana plants and to determine micropropagated plantlet-growth-promotion by their inoculation with diazotrophs. isolates of herbaspirillum-like and burkholderia cepacia related bacteria were inoculated in plantlets of banana cv. prata an? and the cultivar caipira. plantlets of banana cv. prata an? grown in vitro on a poor nitrogen substrate presented higher fresh weight when herbaspirillum-like bacteria were used as inoculum, while the banana cv. caipira grew better with the inoculation of burkholderia cepacia related bacteria. nevertheless, plants of the cultivar caipira cultivated in plastic bags filled with sand and vermiculite (1:2) and supplied with hoagland's solution containing 5 mg l-1 of n grew faster with the inoculation of herbaspirillum-like and burkholderia cepacia related bacteria. this effect was similar to 50 mg l-1 of n fertilizer added to the positive control. however, the control plants showed higher n content in their shoots when compared to bacterial inoculated plants. this work constitutes the first announcement of banana plant-growth-promotion by diazotrophic bacteria.

Abstract: the objective of this study was to evaluate the diversity of gluconacetobacter diazotrophicus isolates from different sugarcane (saccharum spp.) varieties. this study was developed using 123 strains from the internal tissues of stems and roots, isolated from 80 species and hybrids of sugarcane, originated from different countries and maintained in two germoplasm collections localized in bahia and rio de janeiro states. five isolates obtained from coffee plants (coffea arabica), two from pineapple (ananas comosus), one from pennisetum purpureum and 10 strains, which present different electrophoretic patterns, were used in the comparison. cluster analysis of enzyme-linked immunosorbent assay (elisa) results suggested that variations were not correlated with the plant species, sugarcane variety, geographic origin, parts of plants nor with sampling time. high nitrogen doses lowered the diversity of g. diazotrophicus.

Abstract: Este trabalho objetivou selecionar bactérias diazotróficas isoladas de bananeira (Musa spp.) e avaliar sua influência no crescimento de mudas micropropagadas. Bactérias do tipo Herbaspirillum e relacionadas a Burkholderia cepacia foram inoculadas em plantulas de banana cv. Prata An e cv. Caipira. As bananeiras cv. Prata An , cultivadas in vitro com substrato pobre em N, apresentaram maior crescimento na presen a de bactérias do tipo Herbaspirillum, ao passo que as bananeiras cv. Caipira cresceram melhor com o inóculo contendo bactérias do gênero Burkholderia. Em casa de vegeta o, as bananeiras cv. Caipira crescidas em sacolas de plástico contendo areia e vermiculita (1:2), suplementada com a solu o de Hoagland contendo 5 mg L-1 de N, apresentaram maior crescimento quando da inocula o simultanea dos dois gêneros de bactérias, em compara o à inocula o individual. A inocula o simultanea proporcionou um crescimento nas bananeiras equivalente ao observado nas plantas-controles adubadas com 50 mg L-1 de N no substrato, porém o teor e o acúmulo de N na parte aérea das bananeiras foram menores. A contribui o de bactérias diazotróficas no crescimento de bananeiras é demonstrada pela primeira vez.
Sele??o de inoculantes à base de turfa contendo bactérias diazotróficas em duas variedades de arroz

Abstract: the study consisted of a comparison of two-peat materials (brazil and canada) containing different c-total content. the peats were inoculated with strains of herbaspirillum seropedicae zae 94, burkholderia sp. m130, and azospirillum brasilense sp109, and monitored during a period of six months in relation to variation on humidity and survival of bacteria in the inoculant. the quantification of viable cells in the inoculant was measured by the most probable number (mpn) method. the rice seeds were pelleted with the respective inoculants, grown in pots containing soil and maintained outside a greenhouse. the dry mass accumulation, n percent, n-total and yield were determined during the plant cycle. the humidity content varied with storage period. the survival measurement showed that only burkholderia sp. m130 maintained the number of viable cells around 108 g-1 of peat, while there was a reduction in population of other strains. an increase of yield and total n of 13 and 19.4%, respectively in comparison to treatment fertilized with 40 kg n ha-1 was observed for variety iac4440, inoculated with strain zae94. there was no difference in yield of the ir42 inoculated with either zae94 or m130, as compared to n control treatment. no significant difference in development of both rice varieties was observed for both peat used. the results suggest that peat can be used as a carrier for production of an inoculant based on diazotrophic bacteria, since it allowed maintenance of a bacterial population up to 108 cells g-1 peat during the storage period of up to 100 days. the results obtained encourage the practice of inoculation of non-leguminous plants.

Abstract: the objective of this work was to evaluate the diversity of endophytic diazotrophic bacteria of the genera herbaspirillum and burkholderia, in two rice varieties, considered of high (ir 42) and low (iac 4440) contribution on bnf. two experiments were conducted in greenhouse conditions, in order to study the association of endophytic diazotrophic bacteria with wetland rice varieties, which were planted in two types of soil: one from rio de janeiro state and another from goiás state, brazil. bacterial population (in different parts) and physiological stages of the plants were evaluated, followed by the both genera strains isolation using culture media. the isolated bacteria were characterized based on morphological and physiological aspects. high bacterial counts were detected (106 cells g-1 fresh weight), especially in root samples of both rice varieties. the majority of the isolates were used for characterization, and the data obtained from the similarity matrix showed the presence of isolates belonging to herbaspirillum seropedicae, while other isolates were clustered into the burkholderia genus.
Ocorrência e diversidade de bactérias diazotróficas associadas a gramíneas forrageiras do Pantanal Sul Matogrossense

Abstract: this study was carried out to verify the occurrence and genetic diversity of diazotrophic bacteria associated to the native graminaceus plants elyonurus muticus (carona grass) and axonopus purpusii (mimoso grass) and the exotic brachiaria humidicola (brachiaria) that form the pastures in the nhecolandia region of the pantanal in the state of mato grosso do sul, brazil. the plants were harvested in the dry and rainy seasons and the diazotrophic bacteria populations in the soil and on roots and leaves of the three grasses were determined. strains belonging to the species azospirillum brasilense, a. lipoferum, a. amazonense; herbaspirillum spp., and burkholderia spp. were isolated. the number of diazotrophic bacteria associated to the three forage grass species during the rainy season was smaller than in the dry season. genetic diversity in the isolated bacteria was studied by amplified rdna restriction analysis (ardra) using the amplified 16s rdna region. the isolates were divided into five different genotype groups. a. brasilense and a. lipoferum presented 50 % similarity while a. amazonense was included in another group with 25 % similarity to other species of the genus. the bacteria of genus herbaspirillum formed a separate group with only 25 % similarity to genus azospirillum. the fifth group consisted of only one isolate with 25 % similarity to the other groups.
Sele o de inoculantes à base de turfa contendo bactérias diazotróficas em duas variedades de arroz = Selection of peats inoculants with diazotrophic bacteria in two rice varieties

Abstract: O estudo consistiu em avaliar a sobrevivência e a eficiência das bactérias diazotróficas Herbaspirillum seropedicae, Burkholderia sp. e Azospirillum brasilense, inoculadas em dois tipos de turfas. A sobrevivência das bactérias foi determinada pelo método do NúmeroMais Provável (NMP), durante o armazenamento de até seis meses. A eficiência foi avaliada pela contribui o destas bactérias nos parametros agron micos de acúmulo de massa seca e N-total dos gr os nas variedades de arroz IR42 e IAC4440. O experimento foi conduzido emvasos com solo, em delineamento inteiramente casualizado com três repeti es, em arranjo fatorial 2 x 4. Os dados obtidos mostraram que o inoculante contendo a estirpe de Burkholderia sp. manteve o número de células viáveis em torno de 108 cel. g-1 de turfa no período testado, enquanto nos demais inoculantes este número ficou em torno de 106. Os ensaios de inocula o n o mostraram diferen as significativas quanto à origem das turfas usadas como veículo sobre os parametros agron micos e sobrevivência das bactérias. O tratamento que continha H. seropedicae manteve o acúmulo de massa seca e N-total dos gr os estatisticamente igual ao tratamento que recebeu 40 kg N ha-1, embora tenham sido observados aumentos de 13 e 19,4% nestes parametros na variedade IAC4440. Os resultados mostraram-se promissores quanto à utiliza o da prática de inocula o na cultura do arroz. Abstract: the objective of this work was to evaluate the germination and vigour of rice seeds affected by inoculation with diazotrophic bacteria. two experiments were conduced. in the first one, a completely randomized design with four replications arranged in a factorial scheme (2 lots x 10 treatments) was used. for this, rice seeds of cultivar ic4440 were inoculated with eight strains of diazotrophic (ar1122, m130, bf1358, zae94, ar3122, ar2112, cd e br2113), submitted to coating with peat soil humidified with a sterile medium and not inoculated. in the second study, a completely randomized design with four replications arranged in a factorial scheme (2 lots x 6 treatments) was used. for this, recently harvested seeds of cultivars ir42 and zebu branco were inoculated with six strains (bf1358, ar3122, ar1122, zae94, m130 e ar2112), submitted to coating with peat soil humidified with a sterile medium and not inoculated. the bacteria ar1122, m130, bf1358, zae94 and ar3112 can be selected for inoculation of rice seeds. the inoculation of diazotrophic bacteria increases the speed of germination of rice seeds of cultivars ir42 e zebu branco, with low contamination by fungi.

Abstract: the objective of this work was to evaluate the effect of the inoculation of diazotrophic bacteria associated with the molybdenum in two rice cultivars. two experiments were performed: the first one was lead under gnotobiotics conditions, using herbaspirillum seropedicae, strain br 11417 and burkholderia sp., strain br 11340, two doses of molybdenum (1.12 and 2.25 g) and two different sources of mo - sodium molybdate and ammonium molybdate. the seeds were pelleted with peat containing diazotrophic bacteria and molybdenum. after inoculations were seeds planted in to test tubes with capacity for 120 ml content 60 ml hoagland solution without nitrogen. seedlings were harvested after 30 days and plant dry weight was evaluated. the second experiment was held in greenhouse with the same strains, sources and amount of molybdenum. the production and the total n of the rice grains were evaluated. statistical design was completely randomized with four replication and four plants per pot were planted. grain production increased with the inoculation in the cultivars ir42 and iac4440 up to 88 and 57%, respectively.
Use of endophytic diazotrophic bacteria as a vector to express the cry3A gene from Bacillus thuringiensis

Abstract: the goal of this study was to evaluate the potential of endophytic diazotrophic bacteria as a vector to express a cry gene from bacillus thuringiensis, envisaging the control of pests that attack sugarcane plants. the endophytic nitrogen-fixing bacteria gluconacetobacter diazotrophicus strain br11281 and herbaspirillum seropedicae strain br11335 were used as models. the cry3a gene was transferred by conjugation using a suicide plasmid and the recombinant strains were selected by their ability to fix nitrogen in semi-solid n-free medium. the presence of the cry gene was detected by southern-blot using an internal fragment of 1.0 kb as a probe. the production of d-endotoxin by the recombinant h. seropedicae strain was detected by dot blot while for g. diazotrophicus the western-blot technique was used. in both cases, a specific antibody raised against the b. thuringiensis toxin was applied. the d-endotoxin production showed by the g. diazotrophicus recombinant strain was dependent on the nitrogen fixing conditions since the cry3a gene was fused to a nif promoter. in the case of h. seropedicae the d-endotoxin expression was not affected by the promoter (rhi) used. these results suggest that endophytic diazotrophic bacteria can be used as vectors to express entomopathogenic genes envisaging control of sugarcane pests.
