Determination of rheological properties using hybrid optimisation method

Abstract: Purpose: of this paper is description of special software developed for identification of parameters values of rheological models used by ProCAST package for simulation of state of stress in the solidifying castings.Design/methodology/approach: The PyTensionTest uses a hybrid optimization algorithm. This algorithm includes the stochastic method which searches the starting point for direct search optimization method. The identification of parameters values is based on measurement results obtained within the confines of the material tests carried out using the testing machine.Findings: This paper presents the results of approximation of stress-strain relationship for two grades of steel using the elasto-plastic rheological model with power law hardening where its parameters are linearly dependent on temperature.Research limitations/implications: This paper presents the special software that allows user to define any rheological model.Practical implications: Application of obtained values of rheological parameters in the ProCAST software allows to determine stress and strain distribution in continuous castings. It can help to optimize important process parameters.Originality/value: Developed software can approximate the results of static tension tests using rheological model depended on material temperature. This software makes possible an approximation of several stressstrain curves simultaneously. Application of the hybrid optimization method allows to obtain very precise approximation due to determination of the global minimum of the goal function.
Electrostatics controls the formation of amyloid-like superstructures in protein aggregation

Abstract: The possibility for proteins to aggregate in different superstructures, i.e. large-scale polymorphism, has been widely observed, but an understanding of the physico-chemical mechanisms behind it is still out of reach. Here we present a theoretical model for the description of a generic aggregate formed from an ensemble of charged proteins. The model predicts the formation of multi-fractal structures with the geometry of the growth determined by the electrostatic interactions between single proteins. The model predictions are successfully verified in comparison with experimental curves for aggregate growth. The model is general and is able to predict aggregate morphologies occurring both in vivo and in vitro. Our findings provide for the first time a unifying and general framework where the physical interactions between single proteins, the aggregate morphology and the growth kinetics are connected into a single model in excellent agreement with the experimental data.
Oxidation Enhances Human Serum Albumin Thermal Stability and Changes the Routes of Amyloid Fibril Formation

Abstract: Oxidative damages are linked to several aging-related diseases and are among the chemical pathways determining protein degradation. Specifically, interplay of oxidative stress and protein aggregation is recognized to have a link to the loss of cellular function in pathologies like Alzheimer's and Parkinson's diseases. Interaction between protein and reactive oxygen species may indeed induce small changes in protein structure and lead to the inhibition/modification of protein aggregation process, potentially determining the formation of species with different inherent toxicity. Understanding the temperate relationship between these events can be of utmost importance in unraveling the molecular basis of neurodegeneration. In this work, we investigated the effect of hydrogen peroxide oxidation on Human Serum Albumin (HSA) structure, thermal stability and aggregation properties. In the selected conditions, HSA forms fibrillar aggregates, while the oxidized protein undergoes aggregation via new routes involving, in different extents, specific domains of the molecule. Minute variations due to oxidation of single residues affect HSA tertiary structure leading to protein compaction, increased thermal stability, and reduced association propensity.
Unlocked Concanavalin A Forms Amyloid-like Fibrils from Coagulation of Long-lived “Crinkled” Intermediates

Abstract: Understanding the early events during amyloid aggregation processes is crucial to single out the involved molecular mechanisms and for designing ad hoc strategies to prevent and reverse amyloidogenic disorders. Here, we show that, in conditions in which the protein is positively charged and its conformational flexibility is enhanced, Concanavalin A leads to fibril formation via a non-conventional aggregation pathway. Using a combination of light scattering, circular dichroism, small angle X-ray scattering, intrinsic (Tryptophan) and extrinsic (ANS) fluorescence and confocal and 2-photon fluorescence microscopy we characterize the aggregation process as a function of the temperature. We highlight a multi-step pathway with the formation of an on-pathway long-lived intermediate and a subsequent coagulation of such “crinkled” precursors into amyloid-like fibrils. The process results in a temperature-dependent aggregation-coagulation pathway, with the late phase of coagulation determined by the interplay between hydrophobic and electrostatic forces. Our data provide evidence for the complex aggregation pathway for a protein with a highly flexible native conformation. We demonstrate the possibility to generate a long-lived intermediate whose proportion and occurrence are easily tunable by experimental parameters (i.e. temperature). As a consequence, in the case of aggregation processes developing through well-defined energy barriers, our results can open the way to new strategies to induce more stable in vitro on-pathway intermediate species through a minute change in the initial conformational flexibility of the protein. This will allow isolating and experimentally studying such transient species, often indicated as relevant in neurodegenerative diseases, both in terms of structural and cytotoxic properties.
Economic and Financial Comparison between Organic and Conventional Farming in Sicilian Lemon Orchards

Abstract: Sicily has a long tradition in citrus fruit cultivations that with vineyard and olive tree represent the main Mediterranean tree crops. In this paper we have evaluated the economic and financial sustainability of lemon production, both in organic farming and in conventional farming; the two systems differing just for inputs utilized in production process. Economic analysis has been carried out in a representative case study located in the Sicilian northwestern coast, considering an orchard economic life equal to 50 years. Results, which referred to one hectare area, showed both a higher economic and financial sustainability of organic farming respect to conventional farming. The higher profitability of organic farming was due to minor labor requirement and to greater market appreciation for organic products that granted a premium price respect to conventional prices. Moreover, greater profitability of organic farming and use of environmentally friendly inputs in production process make farms competitive and eco-friendly.
The Mélange of Innovation and Tradition in Maltese Law: The Essence of the Maltese Mix?

Abstract: Aim of this paper is to provide valuable insights into the Maltese legal system with a special focus on private law. The assumption is that this legal system is the byproduct of the "mixing" of innovation and tradition, resulting from the interaction of English law and continental law. A major role in the development of the system is played by courts. Some examples (moral damages and pre-contractual liability) are considered which highlight the importance of the function displayed by Maltese judges. Abstract: At present, the problem who can and who should solve the question concerning the preparationand recurrent training of pilots of Ukrainian civil aviation is not solved. The necessity of updatingof aircraft’s park in aviation branch and aircraft operation, which was ripened 10 years ago,demands a corresponding infrastructure. It is necessary to understand, that the pilots ready toperform the flights on modern aircrafts, will not appear by themselves, therefore the real actions onmodernization of existing system of aircrew and the aviation personnel preparation as a whole arerequired. Main objective of this work is the determination of the basic components concerning thesolving of the problematic questions on preparation of aircrew on modern types of aircrafts. Duringthe problem analysis it was specified that the present development of system of aircrew preparationin Ukraine is not perfect, and does not correspond the ICAO and EU requirements, therefore needsan immediate intervention at the highest State level. Trainings are not complex, as do not containthe elements of selection of aircrew members cooperation. Abstract: Substantially nonlinear objects function at limitations of arbitrary kind. These limitations are not necessarily convex areas. For such objects the construction of area of the fully the fully-controlled state area is a serious problem. In the article it has been suggested to replace this task the task of the fully-controlled state area estimation from within. The secure estimation method of the fully-controlled state area has been developed for this purpose. Other dynamic system has been used as a tester (measure) of the fully-controlled state area. Construction and estimation of controllability and accessibility area of this dynamic system well methodically validate and deserved researchers confession. The descriptions concordance requirements of this new object with properties of initial object must be executed and provide property of immersion of new object in an initial object. Abstract: The system method of immersion and virtualization of measured systems in real process is offered. The method works out problem of virtual systems formation and sets conditions at which virtual object to become measuring system for of completely controlled condition field. Here the system of interactively oriented rectangular n-dimensional parallelepipeds is used as measuring system. Imbeddedness of measuring system in real process is understood as process of approximation of a real system controlled condition cone by system of rectangular n-dimensional parallelepipeds.
