Oxygen, nitric oxide and articular cartilage

Abstract: Molecular oxygen is required for the production of nitric oxide (NO), a pro-inflammatory mediator that is associated with osteoarthritis and rheumatoid arthritis. To date there has been little consideration of the role of oxygen tension in the regulation of nitric oxide production associated with arthritis. Oxygen tension may be particularly relevant to articular cartilage since it is avascular and therefore exists at a reduced oxygen tension. The superficial zone exists at approximately 6% O2, while the deep zone exists at less than 1% O2. Furthermore, oxygen tension can alter matrix synthesis, and the material properties of articular cartilage in vitro.The increase in nitric oxide associated with arthritis can be caused by pro-inflammatory cytokines and mechanical stress. Oxygen tension significantly alters endogenous NO production in articular cartilage, as well as the stimulation of NO in response to both mechanical loading and pro-inflammatory cytokines. Mechanical loading and pro-inflammatory cytokines also increase the production of prostaglandin E2 (PGE2). There is a complex interaction between NO and PGE2, and oxygen tension can alter this interaction. These findings suggest that the relatively low levels of oxygen within the joint may have significant influences on the metabolic activity, and inflammatory response of cartilage as compared to ambient levels. A better understanding of the role of oxygen in the production of inflammatory mediators in response to mechanical loading, or pro-inflammatory cytokines, may aid in the development of strategies for therapeutic intervention in arthritis.
Enhancing ordering dynamics in solvent-annealed block-copolymer films by lithographic hard masks supports

Abstract: We studied solvent-driven ordering dynamics of block copolymer films supported by a densely cross-linked polymer network designed as organic hard mask (HM) for lithographic fabrications. The ordering of microphase separated domains at low degrees of swelling corresponding to intermediate/strong segregation regimes was found to proceed significantly faster in films on a HM layer as compared to similar block copolymer films on silicon wafers. The ten-fold enhancement of the chain mobility was evident in the dynamics of morphological phase transitions and of related process of terrace-formation on a macroscale, as well as in the degree of long-range lateral order of nanostructures. The effect is independent of the chemical structure and on the volume composition (cylinder-/ lamella-forming) of the block copolymers. In-situ ellipsometric measurements of the swelling behavior revealed a cumulative increase in 1-3 vol. % in solvent up-take by HM-block copolymer bilayer films, so that we suggest other than dilution effect reasons for the observed significant enhancement of the chain mobility in concentrated block copolymer solutions. Another beneficial effect of the HM-support is the suppression of the film dewetting which holds true even for low molecular weight homopolymer polystyrene films at high degrees of swelling. Apart from immediate technological impact in block copolymer-assisted nanolithography, our findings convey novel insight into effects of molecular architecture on polymer-solvent interactions.
Diet-induced obesity differentially regulates behavioral, biomechanical, and molecular risk factors for osteoarthritis in mice

Abstract: Female C57BL/6J mice were fed either a 10% kcal fat or a 45% kcal fat diet from 9 to 54 weeks of age. Longitudinal changes in musculoskeletal function and inflammation were compared with endpoint neurobehavioral and osteoarthritic disease states. Bivariate and multivariate analyses were conducted to determine independent associations with diet, percentage body fat, and knee osteoarthritis severity. We also examined healthy porcine cartilage explants treated with physiologic doses of leptin, alone or in combination with IL-1α and palmitic and oleic fatty acids, to determine the effects of leptin on cartilage extracellular matrix homeostasis.High susceptibility to dietary obesity was associated with increased osteoarthritic changes in the knee and impaired musculoskeletal force generation and motor function compared with controls. A high-fat diet also induced symptomatic characteristics of osteoarthritis, including hyperalgesia and anxiety-like behaviors. Controlling for the effects of diet and percentage body fat with a multivariate model revealed a significant association between knee osteoarthritis severity and serum levels of leptin, adiponectin, and IL-1α. Physiologic doses of leptin, in the presence or absence of IL-1α and fatty acids, did not substantially alter extracellular matrix homeostasis in healthy cartilage explants.These results indicate that diet-induced obesity increases the risk of symptomatic features of osteoarthritis through changes in musculoskeletal function and pain-related behaviors. Furthermore, the independent association of systemic adipokine levels with knee osteoarthritis severity supports a role for adipose-associated inflammation in the molecular pathogenesis of obesity-induced osteoarthritis. Abstract: Aim of this paper is to provide valuable insights into the Maltese legal system with a special focus on private law. The assumption is that this legal system is the byproduct of the "mixing" of innovation and tradition, resulting from the interaction of English law and continental law. A major role in the development of the system is played by courts. Some examples (moral damages and pre-contractual liability) are considered which highlight the importance of the function displayed by Maltese judges. Abstract: At present, the problem who can and who should solve the question concerning the preparationand recurrent training of pilots of Ukrainian civil aviation is not solved. The necessity of updatingof aircraft’s park in aviation branch and aircraft operation, which was ripened 10 years ago,demands a corresponding infrastructure. It is necessary to understand, that the pilots ready toperform the flights on modern aircrafts, will not appear by themselves, therefore the real actions onmodernization of existing system of aircrew and the aviation personnel preparation as a whole arerequired. Main objective of this work is the determination of the basic components concerning thesolving of the problematic questions on preparation of aircrew on modern types of aircrafts. Duringthe problem analysis it was specified that the present development of system of aircrew preparationin Ukraine is not perfect, and does not correspond the ICAO and EU requirements, therefore needsan immediate intervention at the highest State level. Trainings are not complex, as do not containthe elements of selection of aircrew members cooperation. Abstract: Substantially nonlinear objects function at limitations of arbitrary kind. These limitations are not necessarily convex areas. For such objects the construction of area of the fully the fully-controlled state area is a serious problem. In the article it has been suggested to replace this task the task of the fully-controlled state area estimation from within. The secure estimation method of the fully-controlled state area has been developed for this purpose. Other dynamic system has been used as a tester (measure) of the fully-controlled state area. Construction and estimation of controllability and accessibility area of this dynamic system well methodically validate and deserved researchers confession. The descriptions concordance requirements of this new object with properties of initial object must be executed and provide property of immersion of new object in an initial object. Abstract: The system method of immersion and virtualization of measured systems in real process is offered. The method works out problem of virtual systems formation and sets conditions at which virtual object to become measuring system for of completely controlled condition field. Here the system of interactively oriented rectangular n-dimensional parallelepipeds is used as measuring system. Imbeddedness of measuring system in real process is understood as process of approximation of a real system controlled condition cone by system of rectangular n-dimensional parallelepipeds. Рассмотрена современная концепция управления безопасностью полетов, показывающая, что только соблюдения установленных правил недостаточно. Abstract: Determination of small forms is considered in architecture and in a sculpture. Certainly difference between national and international style in a design. Their use is considered in the design of interiors of passenger spaces of air terminals on existent examples. Connection is analysed between architectural small forms and by the sculpture of small forms, which consists in the transmission of the general semantic and aesthetically beautiful loading on a man, namely to create feeling of comfort, national self-esteem, national pride, and but to consciousness Рассмотрено определение малых форм в архитектуре и скульптуре. Определена разница между национальным и интернациональным стилями в дизайне. Показано их использование в дизайне интерьеров пассажирских пространств аэровокзалов. Проанализирована связь между архитектурой и скульптурой малых форм, заключающаяся в передаче смысловой и эстетической нагрузки на человека, создании ощущения комфорта, национального самоуважения, национальной гордости и самосознания. Розглянуто використання в дизайн нтер’ р в аеровокзальних простор в малих арх тектурних форм та скульптур малих форм у нац ональному та нтернац ональному стилях.
