Novel Drug Delivery System Based on Docetaxel-Loaded Nanocapsules as a Therapeutic Strategy Against Breast Cancer Cells

Abstract: In the field of cancer therapy, lipid nanocapsules based on a core-shell structure are promising vehicles for the delivery of hydrophobic drugs such as docetaxel. The main aim of this work was to evaluate whether docetaxel-loaded lipid nanocapsules improved the anti-tumor effect of free docetaxel in breast cancer cells. Three docetaxel-loaded lipid nanocapsules were synthesized by solvent displacement method. Cytotoxic assays were evaluated in breast carcinoma (MCF-7) cells treated by the sulforhodamine B colorimetric method. Cell cycle was studied by flow cytometry and Annexin V-FITC, and apoptosis was evaluated by using propidium iodide assays. The anti-proliferative effect of docetaxel appeared much earlier when the drug was encapsulated in lipid nanoparticles than when it was free. Docetaxel-loaded lipid nanocapsules significantly enhanced the decrease in IC 50 rate, and the treated cells evidenced apoptosis and a premature progression of the cell cycle from G(1) to G(2)-M phase. The chemotherapeutic effect of free docetaxel on breast cancer cells is improved by its encapsulation in lipid nanocapsules. This approach has the potential to overcome some major limitations of conventional chemotherapy and may be a promising strategy for future applications in breast cancer therapy.

Abstract: El profesorado de primaria se encuentra inmerso en un marco social y en un mercado económico y laboral caracterizado por mutaciones constantes. El papel de la escuela, el rol del docente, la participación social de los diferentes agentes educativos, el acceso a la información, son factores que se han modificado en los últimos tiempos, creando nuevas y diferentes necesidades educativas. La evolución que se genera en estos ámbitos obliga a los docentes a actualizarse y formarse de manera permanente. Una formación específica que, además, enfatice el valor de esta etapa y redefina sus objetivos. La etapa de Primaria ha pasado a ser la parte intermedia de un proceso educativo más amplio, aunque no por ello pierde su especificidad. En este artículo se analizan las características propias de la etapa de Primaria y los nuevos retos que tiene que afrontar el profesorado. Pero qué formación hace falta organizar para abordar estos retos y dar respuesta a todas las necesidades y expectativas que se plantean? Desde la interpretación de estos cambios y del análisis de las necesidades de los docentes de esta etapa se estructuran las posibles líneas de trabajo en la formación de formadores de este profesorado. _______________________________________________________________________________________________________________________________________________________________________________________________________________ Les enseignants du primaire sont immergés dans un cadresocial ainsi que dans un marché économique et professionnel qui se caractérisent par de constantes mutations. The Chemotherapeutic Drug 5-Fluorouracil Promotes PKR-Mediated Apoptosis in a p53- Independent Manner in Colon and Breast Cancer Cells

Abstract: The chemotherapeutic drug 5-FU is widely used in the treatment of a range of cancers, but resistance to the drug remains a major clinical problem. Since defects in the mediators of apoptosis may account for chemo-resistance, the identification of new targets involved in 5-FU-induced apoptosis is of main clinical interest. We have identified the ds-RNA-dependent protein kinase (PKR) as a key molecular target of 5-FU involved in apoptosis induction in human colon and breast cancer cell lines. PKR distribution and activation, apoptosis induction and cytotoxic effects were analyzed during 5-FU and 5-FU/IFNα treatment in several colon and breast cancer cell lines with different p53 status. PKR protein was activated by 5-FU treatment in a p53-independent manner, inducing phosphorylation of the protein synthesis translation initiation factor eIF-2α and cell death by apoptosis. Furthermore, PKR interference promoted a decreased response to 5-FU treatment and those cells were not affected by the synergistic antitumor activity of 5-FU/IFNα combination. These results, taken together, provide evidence that PKR is a key molecular target of 5-FU with potential relevance in the clinical use of this drug.
gef Gene Expression in MCF-7 Breast Cancer Cells is Associated with a Better Prognosis and Induction of Apoptosis by p53-Mediated Signaling Pathway

Abstract: Breast cancer research has developed rapidly in the past few decades, leading to longer survival times for patients and opening up the possibility of developing curative treatments for advanced breast cancer. Our increasing knowledge of the biological pathways associated with the progression and development of breast cancer, alongside the failure of conventional treatments, has prompted us to explore gene therapy as an alternative therapeutic strategy. We previously reported that gef gene from E. coli has shown considerable cytotoxic effects in breast cancer cells. However, its action mechanism has not been elucidated. Indirect immunofluorescence technique using flow cytometry and immunocytochemical analysis were used to detect breast cancer markers: estrogen (ER) and progesterone (PR) hormonal receptors, human epidermal growth factor receptor-2 proto-oncogene (c-erbB-2), ki-67 antigen and p53 protein. gef gene induces an increase in ER and PR expressions and a decrease in ki-67 and c-erbB-2 gene expressions, indicating a better prognosis and response to treatment and a longer disease?free interval and survival. It also increased p53 expression, suggesting that gef?induced apoptosis is regulated by a p53-mediated signaling pathway. These findings support the hypothesis that the gef gene offers a new approach to gene therapy in breast cancer.

Executive Accountability in Parliamentary Democracies: A Comparative Overview: Britain, Germany, India and Ethiopia  [PDF]

Abstract: Democracies especially parliamentary democracies must ensure that public bodies and officials are performing to their full potential, providing value for money in the provision of public services, building confidence in the society, and being responsive to the community they are meant to be serving. The objective of the article is to explain and compare on the functioning parliamentary democracies, Britain, Germany, India and Ethiopia, to give an insight how the parliaments account the executive to bring better governance. Britain is the oldest parliamentary democracy; Germany with handy parliamentary democracy and functional federalism and India is the largest parliamentary democracy. These countries have much experience Ethiopia has to learn. Thus the article considered the institutional structure of accountability; parliamentary control strategies without jeopardizing the concept of separation of powers in a these four countries. The parliamentary system is portrayed under FDRE Constitution and other legislations. The article explored these laws and practical situations and tried to associate with other parliamentary democracies.
Duration of Crime Investigation Quandary under Ethiopian Criminal Justice System  [PDF]

Abstract: Justice is set in motion following the commission and report of an offence to the police targeting to initiation of crime investigation (CPC, 1961). Ethiopian laws do not locate when the investigation shall start. Though laws are silent, police officers may start inquiry at any time to set justice in motion in practice. After justice is set in motion through channels of crime reporting (CPC, 1961, Articles 11, 12 & 19), the question of frequency and duration of investigation is under quandary since it is treated by different legislations differently. The 1961 Ethiopian criminal procedure code was the first document which dealt with the frequency and duration of investigation for the first time in Ethiopian criminal justice history. The FDRE constitution is the other document which locates the concept of duration of investigation at higher level. The Ethiopian criminal policy document envisaged different frequency and duration of crime investigation to be provided by law depending on the complexity and nature of the offence (FDRE Criminal Justice Policy, 2011). After the coming forth of this policy document, different legislations are enacted. The vagrancy control proclamation, the anti-terrorism proclamation, the new draft criminal procedure and evidence law are among the laws. These laws have intricacy concerning constitutionality, consistency and merit. Thus, the writer investigated these issues and analyzed the legislations with regard to the objectives of criminal justice policy.

Abstract: summary: the cytoskeleton of muscle cell is a complex network of proteins which expression pattern is related with the differentiation process. the a-actinin, a protein which plays an important role in the muscular contraction, is essential to the sarcomeres conformation, serving as an anchoring point or center of organization for the preformation of actin filaments. the modulation of the a-actinin expression during the muscular cell maturation, has an important morphofuntional implications to the myocardiocytes. immunofluorescence studies on cryotome sectins of chick embryo hearts and myocardiocyte cultures showed that a-actinin expression was weak in early stages (hamburger hamilton stage 18), staining the cellular membrane of myocardiocytes. a diffuse label for citoplasmic compartment was observed in these stages. a-actinin expression increased during the chick cardiac development. we found in hamburger hamilton stage 29, an intense label for the ventricular myocardium coinciding with the presence of a-actinin along the myocardiocyte myofibrills. analysis of the hamburger hamilton stage 36 showed an increase in the a-actinin expression but without changes in their immunolocalization. the expression pattern of a-actinin during the cardiac development indicated that this protein is a good marker to the degree of myogenic differentiation, the importance of the 29 hh stage to the morphofuntional maturation of myocardiocytes and, the differential expression on the atrial and ventricular myocardium.
Detección mediante RT-PCR de células tumorales circulantes en pacientes con melanoma maligno en estadio III: Correlación con el grosor y la histología del tumor

Abstract: the objetive of our study was to evaluate the utility of the reverse transcriptase-polymerase chain reaction (rt-pcr) in the detection of the tyrosinase enzyme as a marker of circulating melanocites in melanoma patients (stage iii) and the correlation with prognostic factors such as thickness and histologic type tumors. melanoma cells were identified through the tyrosinase mrna by rt-pcr from 28 patients with stage iii of malignant melanoma (classified to the ajcc guidelines). our findings showed a high pcr positivity from patients in stage iii, which were more frequent in primary tumors measuring 4mm (75%). however, no correlation was observed between the positive pcr results and the histological appearance of the primary tumor. our results suggest the possible application of the pcr assay for tyrosinase mrna as a new marker in judging the prognosis of malignant melanoma.
A Closer Look at Distance Learning From Students' Perspective: A Qualitative Analysis of Web Based Online Courses

Abstract: This paper attempts to analyze responses of 40 respondents from Marshall University. The on-line survey that was conducted in 2003 asks: "What are the advantages/disadvantages of taking college web-based courses?" on behalf of the university's English As a Second Language program. Nud*ist 6 and Sphinx Survey, were used to analyze the qualitative data gathered from the online students at Marshall University. After reviewing and analyzing the survey, several themes have emerged. Online courses have also been perceived beneficial, because they are more convenient and are more accessible. Many students see Web CT as a great way to continue their education. Absence of long travel time was one of the major positive aspects listed by many of the students. A detailed examination of the qualitative data reveals that most disadvantages relate in some way to lack of face to face interaction.
