Systematic Dissection of Coding Exons at Single Nucleotide Resolution Supports an Additional Role in Cell-Specific Transcriptional Regulation

Abstract: In addition to their protein coding function, exons can also serve as transcriptional enhancers. Mutations in these exonic-enhancers (eExons) could alter both protein function and transcription. However, the functional consequence of eExon mutations is not well known. Here, using massively parallel reporter assays, we dissect the enhancer activity of three liver eExons (SORL1 exon 17, TRAF3IP2 exon 2, PPARG exon 6) at single nucleotide resolution in the mouse liver. We find that both synonymous and non-synonymous mutations have similar effects on enhancer activity and many of the deleterious mutation clusters overlap known liver-associated transcription factor binding sites. Carrying a similar massively parallel reporter assay in HeLa cells with these three eExons found differences in their mutation profiles compared to the liver, suggesting that enhancers could have distinct operating profiles in different tissues. Our results demonstrate that eExon mutations could lead to multiple phenotypes by disrupting both the protein sequence and enhancer activity and that enhancers can have distinct mutation profiles in different cell types.
The Band-Gap Problem in Semiconductors Revisited: Effects of Core States and Many-Body Self-Consistency

Abstract: A novel picture of the quasiparticle (QP) gap in prototype semiconductors Si and Ge emerges from an analysis based on all-electron, self-consistent, GW calculations. The deep-core electrons are shown to play a key role via the exchange diagram --if this effect is neglected, Si becomes a semimetal. Contrary to current lore, the Ge 3d semicore states (e.g., their polarization) have no impact on the GW gap. Self-consistency improves the calculated gaps --a first clear-cut success story for the Baym-Kadanoff method in the study of real-materials spectroscopy; it also has a significant impact on the QP lifetimes. Our results embody a new paradigm for ab initio QP theory.
Optical and radio properties of giant radio quasars: Central black hole characteristics

Abstract: We analysed the optical and radio properties of lobe-dominated giant-sized (> 0.72 Mpc) radio quasars and compared the results with those derived for a sample of smaller radio sources to determine whether the large size of some extragalactic radio sources is related to the properties of their nuclei. We compiled the largest (to date) sample of giant radio quasars, including 24 new and 21 previously-known objects, and calculated a number of important parameters of their nuclei such as the black hole mass and the accretion rate. We conclude that giant radio quasars have properties similar to those of smaller size and that giant quasars do not have more powerful central engines than other radio quasars. The results obtained are consistent with evolutionary models of extragalactic radio sources which predict that giant radio quasars could be more evolved (aged) sources compared to smaller radio quasars. In addition we found out that the environment may play only a minor role in formation of large-scale radio structures.

Abstract: stellate ganglion blockage (sgb) is a frequently procedure used for alleviating chronic pain of head, neck and the upper extremity. the paratracheal approach has been reported as safe and the most commonly used technique. according to the literature, sgb involve several risks, most of them uncommon and of short duration. it is important to establish the distinction between sgb secondary effects such as those produced by horner's syndrome and complications such as hematomas, subarachnoid injection and convulsions produced by an intravascular injection. sgb is anatomically localized near important structures such as the carotid sinus and the phrenic and vagus nerves. therefore it is important that the health professional that executes this procedure has an excellent knowledge of the neck anatomy and enough experience to guarantee an effective blockage with few complications or none at all. this article is an analytic review of the stellate ganglion and its blockage for the nowadays management of chronic pain.

Partial Dollarization: A currency-matching rule and its implications for monetary policy and welfare

Abstract: Este papel contribuye a los estudios previos de objetivos económicos de infla-ción en economías parcialmente dolarizadas incorporando el efecto de una regla de calce monetario. Específicamente, dicha regla implica la imposición de una restricción para la dolarización crediticia para garantizar que cualquier forma de deuda nominada en divisa (o crédito bancario) se asigne solamente a la expor-tación del sector comercial de la economía. Los resultados son evidentes. Cuan-do la economía no está expuesta financieramente al riesgo del tipo de cambio real: (i) la volatilidad de las principales variables macroeconómicas se reduce, reflejando ganancias en términos de bienestar, y (ii) la función de reacción de política óptima es menos sensible a los cambios en la prima de riesgo y la tasa de interés foránea, y más reactiva a los movimientos en la diferencia del produc-to y la inflación esperada. Las consecuencias de (i) y (el ii) sugieren que el con-sejo que llama a la desdolarización de los pasivos en las economías peque as y abiertas, debe aplicarse solamente al sector no-exportador.

Abstract: El bloqueo del ganglio estrellado es un procedimiento frecuentemente utilizado para aliviar los dolores crónicos de miembros superiores, cabeza y cuello. La técnica paratraqueal ha sido reportada como la más segura y la más utilizada. El bloqueo del ganglio estrellado conlleva riesgos que según la literatura son poco frecuentes y suelen ser de corta duración. Hay que hacer la distinción entre los efectos secundarios, como los producidos por el síndrome de Horner, y las complicaciones, como los hematomas, la inyección intramedular y las convulsiones debidas a la inyección intravascular. El ganglio estrellado se encuentra anatómicamente cerca de estructuras importantes, como el nervio frénico, el laríngeo recurrente, el vago y las grandes arterias. Para su bloqueo, es importante que el profesional tenga un buen conocimiento, tanto de la anatomía como de la técnica utilizada. Este artículo es una revisión analítica sobre el ganglio estrellado y su bloqueo para el manejo del dolor en la actualidad. Interval Estimation for the Two-Parameter Pareto Distribution Based on Record Values

Abstract: This paper considers the interval estimation problem for the Pareto distribution based on record statistics. We present some exact confidence intervals and exact joint confidence regions for the parameters of Pareto distribution based on record statistics. One of the applications of the joint confidence regions of the parameters is to find confidence bounds for the functions of the parameters. In addition, the predictive interval of the future record based on the observed record values is also proposed. A Monte Carlo simulation is conducted to compare the proposed joint confidence regions. Finally, two numerical examples are given also to illustrate the methods proposed in this paper.

The evolutionary optimization of selected welded structures

Abstract: Purpose: of this paper is to present the recent possibility of evolutionary optimization method application to predicting the proper welding parameters in the weld process. The objective of the welding simulation is to study the temperature generated during the welding process and to investigate residual stresses in the component after the welding. Such results give the possibility to determine stress and strain state of welded parts and properties of materials in welding zones. From other side it gives the possibility to perform optimization process looking for welding parameters (welding speed, welding power source etc.) or initial shape of welded sheets according to displacement state (welding of thin metal sheets with stiffeners – T joints). Those results are the base for fatigue analysis too.Design/methodology/approach: In the paper the foundations of FEM simulation of welding process are presented. Also a grid based evolutionary optimization of welding parameters influences on strength parameters in the heat affected zone (HAZ) is shown. Numerical simulation of a welding process using the finite element method is applied.Findings: Results for coupled thermo-mechanical problem are prescribed. Two examples, the grid based evolutionary optimization of HAZ parameters and grid based evolutionary optimization of T-joint component deformation, illustrate the possibility of computational simulation and optimization of welding are presented.Practical implications: Computational simulation and evolutionary optimization give a lot of information very important for engineers. An undesirable side-effect of welding is the generation of residual stresses and deformations in the component and the quality of the weld has a substantial impact on the fatigue life of the structure. These resultant deformations may render the component unsuitable for further use.Originality/value: The presented the grid based evolutionary optimization procedure is a new tool for better understanding and predicting the welding behaviour from the thermo-mechanical process point of view. It gives the possibilities to optimize main welding parameters in order to achieve better structures, taking into account nearly full set of welding parameters, temperature dependent material parameters and simulating the coupled thermo-mechanical problem.
