Global Regulator SATB1 Recruits β-Catenin and Regulates TH2 Differentiation in Wnt-Dependent Manner

Abstract: In vertebrates, the conserved Wnt signalling cascade promotes the stabilization and nuclear accumulation of β-catenin, which then associates with the lymphoid enhancer factor/T cell factor proteins (LEF/TCFs) to activate target genes. Wnt/β -catenin signalling is essential for T cell development and differentiation. Here we show that special AT-rich binding protein 1 (SATB1), the T lineage-enriched chromatin organizer and global regulator, interacts with β-catenin and recruits it to SATB1's genomic binding sites. Gene expression profiling revealed that the genes repressed by SATB1 are upregulated upon Wnt signalling. Competition between SATB1 and TCF affects the transcription of TCF-regulated genes upon β-catenin signalling. GATA-3 is a T helper type 2 (TH2) specific transcription factor that regulates production of TH2 cytokines and functions as TH2 lineage determinant. SATB1 positively regulated GATA-3 and siRNA-mediated knockdown of SATB1 downregulated GATA-3 expression in differentiating human CD4+ T cells, suggesting that SATB1 influences TH2 lineage commitment by reprogramming gene expression. In the presence of Dickkopf 1 (Dkk1), an inhibitor of Wnt signalling, GATA-3 is downregulated and the expression of signature TH2 cytokines such as IL-4, IL-10, and IL-13 is reduced, indicating that Wnt signalling is essential for TH2 differentiation. Knockdown of β-catenin also produced similar results, confirming the role of Wnt/β-catenin signalling in TH2 differentiation. Furthermore, chromatin immunoprecipitation analysis revealed that SATB1 recruits β-catenin and p300 acetyltransferase on GATA-3 promoter in differentiating TH2 cells in a Wnt-dependent manner. SATB1 coordinates TH2 lineage commitment by reprogramming gene expression. The SATB1:β-catenin complex activates a number of SATB1 regulated genes, and hence this study has potential to find novel Wnt responsive genes. These results demonstrate that SATB1 orchestrates TH2 lineage commitment by mediating Wnt/β-catenin signalling. This report identifies a new global transcription factor involved in β-catenin signalling that may play a major role in dictating the functional outcomes of this signalling pathway during development, differentiation, and tumorigenesis.

Sustainable Ground Water Development in Hard Rock Aquifers in Low-Income Countries and the Role of UNESCO _ IUGS - IGCP projec -GROWNET-

Abstract: Hard rock aquifers for the purpose of this Paper mean the non-carbonate, fractured rock aquifers in the terrain covered by crystalline basement complex, metamorphic rocks and also by extensive effusive volcanic rocks like the basalts of western India (Deccan traps. Ground water development in hard rock aquifer areas has always played a secondary role compared to that in the areas having high-yielding unconsolidated or semi-consolidated sediments and carbonate rocks. This has been due to the relatively poor ground water resources in hard rocks, low specific capacity of wells, erratic variations and discontinuities in the aquifer properties, and difficulties in exploration and assessment of the resource. It should, however, be remembered that for the millions of farmers in developing countries, having their small farms in the barren landscape of fractured hard rock terrain, whatever small supply available from these poor aquifers is the only hope for upgrading their standard of living by growing irrigated crops or by protecting their rain-fed crops from the vagaries of rainfall. It is also their only source for drinking water for the family and cattle. In many developing countries, hard rock hydrogeologists have therefore, an important role to play. In the developed countries, the interest in hard rock hydrogeology, apart from drinking water supplies to small communities, is recently promoted by the prospects of using these low permeability rocks for the storage of hazardous nuclear and chemical wastes. The study of ground water flow through faults, fissures and fractures is also of interest to scientists studying the migration of contaminants. Hard rock hydrogeologists, the world over, are therefore divided into two main groups: Those interested in obtaining ground water for domestic, irrigational or industrial use by exploring fractured and permeable zones in a relatively less permeable matrix of hard rock and those interested in locating impermeable or the least permeable zones for storage of hazardous nuclear waste. Ironically, for the first group even the most permeable zones are often not good enough to yield adequate water supply, while for the second group even the least permeable zones are often not good enough for safe storage of hazardous nuclear waste over a prolonged period of a few hundred years. This Paper discusses the occurrence of ground water and the precautions for sustainable development of ground water in arid and semi-arid regions, in view of the forthcoming climatic changes. Abstract: We first establish a simple procedure to obtain with 11-figure accuracy the values of Chandrasekhar's H-function for isotropic scattering using a closed-form integral representation and the Gauss-Legendre quadrature. Based on the numerical values of the function produced by this method for various values of the single scattering albedo and the cosine of the azimuth angle of the direction of radiation emergent from or incident upon a semi-infinite scattering-absorbing medium, we propose a rational approximation formula, which allows us to reproduce the correct values of the H-function within a relative error of 2.1/100000 without recourse to any iterative procedure or root-finding process.
POTENTIAL ANTIBACTERIAL AND ANTINFUNGAL ACTIVITY OF AQUEOUS EXTRACT OF CYNODON DACTYLON

Abstract: The use of medicinal plants in the world and especially in India, contributes significantly to primary health care. Whole plant of the Cynodon dactylon is traditionally used to treat painful and inflammatory condition. We have undertaken this study since no detailed scientific study was available regarding the antifungal and antibacterial activity of the aqueous extract of the whole plant. In this study the aqueous extract of Cynodon dactylon was used to determine the antimicrobial activity against Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Proteus mirabilis and Candida albicans. The antimicrobial activity was determined using the agar well diffusion method. The diameter of the clear zone of inhibition surrounding the well was measured. The aqueous extract of Cynodon dactylon had antimicrobial activity against all the test organisms except Candida albicans. Phytotoconstituents present included Saponins, Tannins, steroids and Flavonoids. It can be concluded that aqueous extract of whole plant of Cynodon dactylon may be considered as an antibacterial agent and can be used to source antibiotic substances for possible treatment of bacterial infections.

Abstract: Synchronizer used for data synchronization posses non-zero probability of failure in Globally Asynchronous Locally synchronous System on Chip. It plays very crucial role in determining the reliability of system. Synchronizer is susceptible to metastability if asynchronous data changes in setup and hold time window. Theoutput of synchronizer is indeterminate .This risk must be mitigated. In this paper structure of synchronizer is proposed for improved reliability. It determines change of data in the metastability window with signal setup start (SS) and hold end(HE) . To receive synchronised data, it samples synchronizer which is placed in parallelwith operating synchronizer .This parallel placed synchronizer receives delayed data. This proposed architecture is modelled with VHDL and simulated with ModelSim SE6.2C for functional correctness. Asynchronous data rate is generated at varying frequency .MTBF is improved substantially with the increase penalty of one cycle.
AN APPROACH TO DESIGN ADVANCED STANDARD ENCRYPTION ALGORITHM USING HARDWARE / SOFTWARE CO-DESIGN METHODOLOGY

Abstract: An Advanced Standard Encryption Algorithm (AES) is widely used in modern consumer electronicproducts for security. The IEEE 802.15.4 Low-Rate wireless sensor networks also use AES algorithm wherelow power consumption is the priority. To reduce the time taken for encryption of huge data, the algorithm hasto be implemented in hardware. To meet the requirement for low area the algorithm has to be implemented insoftware. Hence, a balance has to be achieved between hardware and software implementations in terms of areaand speed, so as to improve the overall performance of the system. Also with the co-design methodology totalthermal power dissipation is reduced. In this paper, 128 bit AES algorithm is implemented with hardware incombination with software using Altera NIOS II Processor platform. Altera’s Quartus II environment is used fordesign of the system. Cyclone II FPGA is used as a development platform. Software program is written in C language. NIOS II ntegrated Development Environment (IDE) is used to integrate hardware and software together. By adopting hardware / software co-design methodology for implementation of AES, results show that a onsiderable improvement in speed can be achieved as compared to software only approach. Further, the significant reduction in area is achieved as compared to hardware only approach. By the approach of co-design an optimized design in terms of speed and area is achieved and also the thermal power dissipation is reduced
