FORCA, a promoter element that responds to crosstalk between defense and light signaling

Abstract: FORCA is a hexameric promoter motif that is conserved in clusters of Arabidopsis genes co-expressed in response to fungal or oomycete pathogens as well as defined light treatments. FORCA is generally more frequently present in Arabidopsis promoter regions than statistically expected. It constitutively interacts in a DNA-sequence specific manner with nuclear Arabidopsis proteins. These interactions are suppressed by defense-related stimuli and enhanced by prolonged exposure to constant light. Furthermore FORCA mediates constitutive reporter gene expression in transiently transformed Nicotiana benthamiana leaves as well as in stably transformed Arabidopsis plants. Its responsiveness to defense-stimuli is modulated by the duration of light exposure. In plants grown under normal light conditions or constant darkness defense-related stimuli result in suppression of FORCA-mediated reporter gene expression, while in plants grown under constant light exposure, defense-induction results in enhanced FORCA-mediated expression. In addition, we found plants subjected to constant light exposure to exhibit reduced susceptibility to virulent H. parasitica.We propose that FORCA is a regulatory cis-element that is present in a wide variety of Arabidopsis promoters. It integrates light- and defense-related signals and participates in adjusting the transcriptome to changes in environmental conditions.Molecular recognition of pathogenic microorganisms triggers in plants comprehensive transcriptional reprogramming. Regulation of the heat stress response in Arabidopsis by MPK6-targeted phosphorylation of the heat stress factor HsfA2

Abstract: So far little is known on the functional role of phosphorylation in the heat stress response of plants. Here we present evidence that heat stress activates the Arabidopsis mitogen-activated protein kinase MPK6. In vitro and in vivo evidence is provided that MPK6 specifically targets the major heat stress transcription factor HsfA2. Activation of MPK6 results in complex formation with HsfA2. MPK6 phosphorylates HsfA2 on T249 and changes its intracellular localisation. Protein kinase and phosphatase inhibitor studies indicate that HsfA2 protein stability is regulated in a phosphorylation-dependent manner, but this mechanism is independent of MPK6. Overall, our data show that heat stress-induced targeting of HsfA2 by MPK6 participates in the complex regulatory mechanism how plants respond to heat stress.

Abstract: Les deux dernières décennies ont vu na tre une discipline nouvelle: l’accueil et le soutien psychologique des personnes vivant des expériences exceptionnelles. Avoir l’impression de vivre dans une maison hantée, témoigner de l’apparition d’un OVNI, se questionner sur les possibilités d’influences télépathiques ou encore garder pour soi la sensation déréalisante d’une sortie hors-du-corps, tels sont quelques exemples de ce que rapportent ces personnes. L’hétérogénéité de ces plaintes fait qu’on les classe souvent dans la catégorie des expériences vécues comme paranormales . Si de tels récits sont toujours parvenus aux oreilles des cliniciens, on doit à des travaux récents une restructuration de l’écoute de ces personnes.

Avere l'impressione di vivere in una casa abitata, testimoniare l’apparizione di OVNI, interrogarsi sulle possibilità di influenze telepatiche o anche conservare per sé la sensazione irreale di uscire “fuori dal corpo”, sono alcuni esempi di ciò che provano queste persone. L'eterogeneità di queste testimonianze le classifica spesso nella categoria delle "esperienze vissute come paranormali". Se tali resoconti sono sempre pervenuti agli esperti, si deve ai recenti lavori di ripristino dell'ascolto di queste persone.

Abstract: à partir d’un corpus traitant de l’élégie classique, de Catulle à Ovide, et de l’élégie chrétienne représentée par les poèmes majoritairement élégiaques de Venance Fortunat, nous nous proposons de réfléchir sur les figures du sujet énonciateur par l’intermédiaire des motifs du vin et de l’eau et de nous interroger sur la possibilité d’une esthétique a-subjective de l’élégie. Le motif du vin permet aux élégiaques latins de représenter Dionysos comme obstacle ou adjuvant dans leur histoire d’amour ; il est même facteur de dépassement des tensions passionnelles ; le motif de l’eau est apollinien et caractérise en partie l’inspiration élégiaque. Venance Fortunat christianise ses deux motifs : le vin est le sang du Christ, l’eau est Vérité. Nous accédons alors au registre de la foi qui transcende toute forme de subjectivité singulière.

Abstract: Jaynes' transformation group principle is used to derive the objective prior for the velocity of a non-zero rest-mass particle. In the case of classical mechanics, invariance under the classical law of addition of velocities, leads to an improper constant prior over the unbounded velocity space of classical mechanics. The application of the relativistic law of addition of velocities leads to a less simple prior. It can however be rewritten as a uniform volumetric distribution if the relativistic velocity space is given a non-trivial metric.
A common polymorphism in NR1H2 (LXRbeta) is associated with preeclampsia

Abstract: We assessed associations between single nucleotide polymorphisms of NR1H3 (rs2279238 and rs7120118) and NR1H2 (rs35463555 and rs2695121) and the disease in 155 individuals with preeclampsia and 305 controls. Genotypes were determined by high-resolution melting analysis. We then used a logistic regression model to analyze the different alleles and genotypes for those polymorphisms as a function of case/control status.We found no association between NR1H3 SNPs and the disease, but the NR1H2 polymorphism rs2695121 was found to be strongly associated with preeclampsia (genotype C/C: adjusted odds ratio, 2.05; 95% CI, 1.04-4.05; p = 0.039 and genotype T/C: adjusted odds ratio, 1.85; 95% CI, 1.01-3.42; p = 0.049).This study provides the first evidence of an association between the NR1H2 gene and preeclampsia, adding to our understanding of the links between cholesterol metabolism and this disease.Preeclampsia (PE) is a frequent complication of the second half of pregnancy and is one of the leading causes of maternal perinatal mortality and morbidity [1]. This condition, affecting 2.5 to 3% of pregnant women is defined by gestational hypertension accompanied by proteinuria [2,3]. Many risk factors have been described, including, in particular, cardiovascular risks, such as diabetes mellitus and high body mass index [4,5]. Cholesterol metabolism may also contribute to the pathogenesis of preeclampsia. Indeed, high total cholesterol and LDL (low density lipoprotein)-cholesterol levels are significantly associated with this disease [6]. Moreover, it has been shown that a familial history of PE almost triples the risk of a woman developing this disease, consistent with the involvement of genetic factors [4]. Many risk factors have been described, but the molecular mechanisms underlying this disease remain unclear. Abstract: prenatal ethanol exposure, in our professional practice, is an almost neglected condition as an important etiological factor for the induction of a wide spectrum of neuropsychiatric diseases that may appear during childhood, adolescence or adulthood. children born to alcoholic mothers may show a profound mental retardation ranging to an apparent normality, and extending through epilepsy, attention deficit disorders with or without hyperactivity, autism and pervasive developmental disorders, and different types of learning disorders. when adolescents, they may develop different kinds of personality disorders and substance abuse disorders. finally, in adulthood, they may suffer from different types of affective and psychotic disorders, among others. a great number of those children may not develop their full mental and social potentiality as free individuals. they usually have diverse types of cognitive, attentional, mnemonic and affective impairments. not infrequently, they engage in antisocial behaviors or have school or work troubles. in this work, the present clinical classifications and diagnostic criteria for the disorders emerging from a prenatal ethanol exposure are reviewed in order to call attention to the medical pediatric and neuropsychiatric community about the increasingly, although underdiagnosed, frequency of these disorders in our country.
