Estudios de genotoxicidad de antibióticos antitumorales en células eucariotas

Abstract: during the past 20 years, the laboratory of cytogenetics and mutagenesis at imbice has developed various studies to analyze the genotoxicity caused by different chemical mutagens to eukaryotic cells. research has mainly focused on the analysis of the genotoxic mechanisms involved and the effects caused by three antibiotics of known antitumor properties, bleomycin (blm), streptonigrin (en), and streptozotocin (ez). several cell lines and human peripheral blood lymphocytes were used as experimental models. currently, our studies are devoted to analyze the effects of those antibiotics on telomeres and interstitial telomeric sequences of vertebrate chromosomes. since blm and en exert direct damage on dna and chromosomes by free radicals release, these antibiotics are considered radiomimetic compounds. ez is frequently used to induce diabetes mellitus in laboratory rats, and though it exerts its genotoxic action partially by free radical release, in contrast to blm and en, it is an alkylating agent. the present work summarizes the most significant results obtained during the last 15 years by the research group at the laboratory of cytogenetics and mutagenesis (imbice) on the genotoxic effects of the antibiotics above mentioned.
Braquiterapia endobronquial de alta tasa de dosis en pacientes con obstrucción de la vía aérea central: Experiencia en el Instituto Nacional del Cáncer y revisión de la literatura High dose rate endobronchial brachytherapy in patients with central airway obstruction

Abstract: Introducción: La indicación principal de la braquiterapia endobronquial moderna (BTEB) de alta tasa de dosis (HDR), es la paliación de síntomas por crecimiento endobronquial de cánceres pulmonares. Métodos: Se realizó BTEB HDR a 27 pacientes sintomáticos de patología tumoral endobronquial o traqueal, debido a patologías primarias bronquiales o secundarias metastásicas. Los tumores se ubicaban en tráquea, carina o a nivel bronquialproximal. Para observar mejorías en la sintomatología clínica, a todos se les realizó una fibrobroncoscopía (FBC) para medir el grado de obstrucción bronquial e instalar catéteres endobronquiales de braquiterapia. Se administraron entre 1 y 4 fracciones de 7 a 7,5 Gy. Se registró subjetivamente la disnea, tos y hemoptisis antes y después del tratamiento, de acuerdo a una escala internacionalmente validada. Resultados: Tras el tratamiento todos los síntomas disminuyeron considerablemente, desapareciendo toda sintomatología severa. La hemoptisis y disnea desaparecieron en el 100% y 40% de los pacientes respectivamente, y la tos desapareció o disminuyó a grado leve en el 90% de los pacientes. Methods: EBBT was performed to 27 patients who suffered symptomatic tumoral endobronchial or tracheal pathology, due to primary bronchial disease or secondary metastasis cancer, evaluating the rate of clinical benefit. The tumors were located at tracheal, carinal or proximal bronchial level. Fibrobronchoscopy was performed to all the patients to measure the degree of airway obstruction and to install brachytherapy endobronchial catheters. Between 1 to 4 fractions of 7 to 7.5 Gy were administered. Dyspnea, cough and hemoptysis were subjectively registered before and after treatment, according to an international validated scale. Results: After treatment, all symptoms considerably decreased, disappearing all of the severe categories. Hemoptysis and dyspnea resolved in a 100% and 40% of patients, respectively; and cough disappeared or was reduced to a minimum grade in 90% of cases.
Semiclassical regime of Regge calculus and spin foams

Abstract: Recent attempts to recover the graviton propagator from spin foam models involve the use of a boundary quantum state peaked on a classical geometry. The question arises whether beyond the case of a single simplex this suffices for peaking the interior geometry in a semiclassical configuration. In this paper we explore this issue in the context of quantum Regge calculus with a general triangulation. Via a stationary phase approximation, we show that the boundary state succeeds in peaking the interior in the appropriate configuration, and that boundary correlations can be computed order by order in an asymptotic expansion. Further, we show that if we replace at each simplex the exponential of the Regge action by its cosine -- as expected from the semiclassical limit of spin foam models -- then the contribution from the sign-reversed terms is suppressed in the semiclassical regime and the results match those of conventional Regge calculus.
Mechanical laws of the Rindler horizon

Abstract: Gravitational perturbations of flat Minkowski space make the Rindler horizon dynamical: the horizon satisfies mechanical laws analogous to the ones followed by black holes. We describe the gravitational perturbation of Minkowski space using perturbative field-theoretical methods. The change in the area of the Rindler horizon is described in terms of the deflection of light rays by the gravitational field. The difference between the area of the perturbed and the unperturbed horizon is related to the energy of matter crossing the horizon. We derive consistency conditions for the validity of our approximations, and compare our results to similar ones present in the literature. Finally, we discuss how this setting can be used in perturbative quantum gravity to extend the classical mechanical laws to thermodynamic laws, with the entanglement of field modes across the Rindler horizon providing a notion of thermodynamic entropy.
Ionospheric Doppler measurements by means of HF-radar techniques

Abstract: Studies of the dynamics of the ionosphere and its related phenomena are mainly based on Doppler Drift measurements. The time variation (ionisation/recombination) of plasma density, thermospheric wind and others can be observed by means of HF-radars. The technique of Doppler Drift measurements is a quite complex technique that is now affordable by means of an advanced ionospheric sounder. The combination of vertical sounding and interferometric Doppler detection discloses the Doppler sources. The echo signal contains the Doppler shift in frequency imposed on the wave carrier by each point source where the signal is reflected. Other phenomena like environmental noise and the intrinsic error of the measurements that, together with the change in time of the refractive index, affect the measurements in various ways impeding to better quantify the results.

A Single Amino Acid Change Converts the Sugar Sensor SGLT3 into a Sugar Transporter

Abstract: Sodium-glucose cotransporter proteins (SGLT) belong to the SLC5A family, characterized by the cotransport of Na+ with solute. SGLT1 is responsible for intestinal glucose absorption. Until recently the only role described for SGLT proteins was to transport sugar with Na+. However, human SGLT3 (hSGLT3) does not transport sugar but causes depolarization of the plasma membrane when expressed in Xenopus oocytes. For this reason SGLT3 was suggested to be a sugar sensor rather than a transporter. Despite 70% amino acid identity between hSGLT3 and hSGLT1, their sugar transport, apparent sugar affinities, and sugar specificity differ greatly. Residue 457 is important for the function of SGLT1 and mutation at this position in hSGLT1 causes glucose-galactose malabsorption. Moreover, the crystal structure of vibrio SGLT reveals that the residue corresponding to 457 interacts directly with the sugar molecule. We thus wondered if this residue could account for some of the functional differences between SGLT1 and SGLT3.

Abstract: In this paper we present an analysis of the geological, meteorological and climatic data recorded in Acquapendente (VT) over 24 years. These data are compared to check local variations,long term trends, and correlation with maen annual temperature. The ultimate goal of this work is to understand logn term climatic changes in this geographic area. The analysis is performed using a statistical approach. From each long series of data calculate the hourly averages and that the monthly averages in order to reduce the fluctuations in the short time due to the day / night cycle. A particular care is used to minimize any effect due to prejudices in case of lack of data. Finally, we calculate the annual average from the monthly ones.

Abstract: This paper contains the result of the elaboration of informations about Solar Irradiation, Geological, Meteorological and Climatic from the point of view of the quantitative data and social interaction recorded in Taviano (LE) over 24 years. These data are compared to check local variations, long term trends, and correlation with mean annual temperature. The ultimate goal of this work is to understand long term climatic changes in this geographic area. The classes of event considerated are hydrogeological phenomena, sun irradiation, seismic, volcanic, meteorological and climatological event. Only event occurred between 1990 and 2014 are considerated. The analysis is performed using a statistical approach. A particular care is used to minimize any effect due to prejudices in case of lack of data. Finally, we calculate the annual average from the monthly ones. Data on this paper don't come from a complete census of phenomena; they are considered enough representative of the accepted vulnerability level at the beginning of this study.
