Abstract:
在传统BP算法的基础上,将Levenbery-Marquardt优化法与神经网络模型相结合的L-M优化BP算法进行了深入应用和分析。此方法与传统算法相比提高了系统的学习速度,加快了网络的收敛。针对某风电场58台机组额定功率为850 kw的风电机组20(15分钟一个预测点)的历史数据使用L-M算法优化下的前馈神经网络模型——BP神经网络模型进行了该风电场的实时预测,结果表明该方法在一定程度上更好的逼近了真实的曲线。
Based on the traditional BP algorithm, combining Levenhery-Marquardt optimized algorithm and a neural network forecasting methodthis paper put forward a L-M optimized BP algorithm. The algorithm quickens the train, improves stability. For the real power data of 58 wind turbines of some wind farm in somewhere, a real-time prediction has been made based on L-M optimized BP algorithm, and the result shows that the algorithm produces better results than traditional method.

Abstract: 研究富硒螺旋藻(SeSP)培养生物转化含硒蛋白质(SeP)中硒的含量及分布,观察SeP在体外对氧自由基的清除效应。藻体总蛋白(TP)用SDS-PAGE电泳分离后,用电感偶合等离子体质谱技术(ICP-MS)对不同分子量范围蛋白质中的硒含量进行精确定量;通过激光烧蚀(LA)-ICP-MS对纯化含硒藻蓝蛋白(SePC)硒含量在电泳胶上进行原位检测;用制备SDS-PAGE凝胶电泳分离纯化不同分子量的SeP组份,化学发光方法检测SeP在体外对超氧自由基和羟自由基的清除作用。结果发现,与未富硒培养的螺旋藻(SP)相比,SeSP总蛋白中硒含量提高了32倍,可达805.48 μg/g,其中75.81%分布于25 ku以下小分子量含硒蛋白质(LMWSeP)中,LA-ICP-MS证明纯化SePC亚基含有稳定共价结合活性硒元素,LMWSeP对自由基的直接最大清除率在70%以上。结果提示,利用SeSP培养可转化生产活性SeP,SeSP中低分子量SeP组份是一种具有较高抗氧化活性的天然活性硒资源,其中硒的存在形态、合成转化机制和体内生物活性有待深入研究。
Selenium (Se) content and distribution of Se-containing biotransformation proteins (SeP) in Se-enriched Streptomyces platensis (SeSP) were explored. Additionally, the in vitro scavenging activity of oxygen free radicals by SeP was studied. Total proteins (TP) in S. platensis were separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Se content in different molecular-weight proteins was measured using inductively-coupled plasma mass spectrometry (ICP-MS). Laser ablation (LA)-ICP-MS was used for in situ detection of Se content in purified Se phycocyanin (SePC) within the electrophoresis gel. The in vitro clearance of superoxide and hydroxyl free radicals by SeP was investigated by chemiluminescence. The results showed that Se content in SeSP-TP was 32-fold higher than that in non-enriched S. platensis (SP), reaching 805.48 μg/g, while 75.81% oftotal Se was distributed in low-molecular weight SePs (LMWSeP), with molecular weights ≤ 25 ku. LA-ICP-MS results confirmed that purified SePC subunits contained stable, covalently-bound, active Se. The maximum clearance rate of free radicals by LMWSeP was over 70%. The findings suggest that SeSP cultures could be used to produce active SeP by biotransformation. The LMWSeP components in SeSPs are a source of natural Se with high anti-oxidation activity. Further studies are required to determine the form of Se, transformation mechanism, and in vivo bioactivity.

Abstract:

随着光动力学疗法(photodynamic therapy,PDT)基础研究的不断深入和临床应用的广泛开展,光敏剂的光漂白现象日渐成为PDT研究的热点问题。实验在ALA的终浓度为10 mM/ml,辐射光波长为410 nm,辐照光功率为5 mW/cm2等条件下采用荧光探针标记技术在自行设计的光动力疗法反应室进行了PDT实验研究。Cell Counting Kit-8 (CCK-8)法用来检测细胞的活性,通过获得的荧光光谱研究了亚细胞的光漂白情况。结果表明:以每种细胞器光照前的平均荧光强度为基准,则光照60 min后线粒体、溶酶体、内质网的荧光强度下分别降低了95%、91.5%和51.2%。线粒体区域光漂白速率较快,其荧光强度的变化基本表征了HL60细胞活性的变化,推断线粒体是光动力效应的主要作用靶点之一。
With the development of the fundamental research of photodynamic therapy (PDT) and its clinical applica-tion, the photobleaching properties of photosensitizers have become a hotspot of research. The experimental research of ALA mediated PDT was carried out in the own designed reaction chamber when the final concentration of ALA is 10 mM/ml, the wavelength and the output power of the light source are 410 nm and 5 mW/cm2 separately. Cell Counting Kit-8 (CCK-8) was used to evaluate the cell viability while the photobleaching of PpIX in the subcellular structure of HL60 was detected by fluorescence spectroscopy. The results showed that after 60 min irradiation the fluorescence in-tensity of mitochondria, lysosomes and endoplasmic reticulum had decreased by 95%, 91.5% and 51.2% respectively compared to that before irradiation. The rate of photobleaching in mitochondria which can characterize the activity of HL60 was significantly higher than others, this suggest that mitochondria might be one of main therapeutic targets of photodynamic therapy.

大鼠肝癌细胞系5-羟色胺及其受体的免疫细胞化学定位

Abstract: 近年已发现除神经内分泌细胞肿瘤外,其它肿瘤也能分泌神经肽和经典的神经递质,某些结肠癌的癌细胞已被检出-羟色胺(5-HT).而且有人证明,5-HT能刺激结肠癌细胞的分裂增殖,5-HT1受体介导了这种作用.然而,5-HT及其受体在肝癌细胞的定位尚未见报道.我们以大鼠肝癌细胞系FSK 7902作为研究对象,采用免疫细胞化学方法观察了5-HT及其受体的分布,为进一步研究5-HT在肝癌细胞中的意义提供形态学依据.
漂浮基柔性空间机械臂关节运动的分块神经网络控制及柔性振动模糊控制

Abstract: 讨论了载体位置不受控、姿态受控情况下,漂浮基柔性空间机械臂关节运动及柔性振动主动抑制的控制问题。由系统动量守恒关系及假设模态法,利用拉格朗日方法建立了漂浮基柔性空间机械臂的系统动力学方程。之后采用奇异摄动理论,将其分解为表示刚性运动的慢变子系统和柔性振动的快变子系统。据此,设计了柔性空间机械臂系统载体姿态及机械臂关节铰协调运动的组合控制器。针对慢变子系统——柔性空间机械臂的刚性运动,设计了分块神经网络控制器,以完成系统参数未知情况下关节空间协调运动的轨迹跟踪;而对于快变子系统——柔性臂的振动,则设计了模糊控制器来主动抑制柔性杆的振动。数值仿真证实了提出方法的有效性。

Abstract: 该文研究了在本体位置不受控、姿态受控的情况下,漂浮基双臂空间机器人系统惯性空间协调运动的轨迹跟踪问题。针对存在外部扰动和未知系统惯性参数的情况,设计了一种基于模糊基函数网络的自适应调节控制方案。用模糊基函数网络对双臂空间机器人系统建模,利用鲁棒技术处理建模误差和外部扰动在内的不确定性。所提控制方案的特点是:不需要动力学方程中的惯性参数符合线性规律,也无需预知系统的惯性参数;同时可以利用学习规则在线自适应调节模糊基函数网络的所有权值和参数,因此控制方案是灵活的。该文稳定性分析证明了控制方案的全局稳定性和良好的轨迹跟踪性能。仿真算例表明了该方案能有效地控制双臂空间机器人的本体姿态和末端爪手协调地跟踪期望的运动轨迹。
微波化学反应器与实验结果的重复性

Abstract: 对基于家用微波炉改装的微波化学反应器建立模型,用有限元方法联合求解麦克斯韦方程和热传导方程并考虑了介电常数随温度和时间的变化,得到烧杯中溶液的温度分布。计算发现烧杯在反应器腔体中摆放的位置对加热效果有着显著影响。而化学反应是一个非平衡体系,这种位置差异带来的影响会更加明显。通过实验进一步验证了以上结论,并且发现:由于反应器磁控管的不稳定性,即使烧杯位于相同位置,多次实验的结果也不相同。因此,基于家用微波炉改装的微波化学反应器很难保证微波化学实验的重复性。
