普伐他汀对大鼠急性心肌梗死后热休克蛋白b7表达的影响

Abstract: ?目的观察急性心肌梗死(ami)大鼠梗死区心肌组织中热休克蛋白b7(heatshockproteinb7,hspb7)mrna和蛋白的表达及普伐他汀的干预作用。方法80只ami大鼠随机分为心肌梗死组(ami)和普伐他汀组(p),每组40只,同时设假手术组(sh)大鼠40只,再将3组大鼠分为1h组、3h组、6h组和12h组,每个时间组10只。sh组开胸后不结扎冠脉,ami组开胸后结扎冠状左前降支,p组开胸后结扎冠状左前降支,同时给予普伐他汀0.5mg/(kg?d)灌胃(其余组给予等量蒸馏水灌胃)。分别于各时间点处死大鼠,取左心室梗死区心肌组织,假手术组取对应部位的心肌组织,分别采用rt-pcr和免疫组化法检测hspb7mrna和蛋白的表达。结果:各ami组和p组大鼠梗死区心肌组织hspb7mrna和蛋白与sh组的表达相比,差异均有统计学意义(p<0.001);梗死后1h,梗死区心肌hspb7mrna和蛋白的表达增加(p<0.05),在3h时达到高峰,6h组、12h组mrna和蛋白的表达仍高于假手术组,且p组中各时间点hspb7的表达比ami组高。结论:hspb7能够在ami早期表达,普伐他汀可促进ami后梗死区心肌组织hspb7表达,可能是其在心梗早期起到保护心肌作用的机制之一

Abstract: 目的:探讨芦丁对大鼠心肌缺血再灌注损伤的保护作用及机制。方法:将雄性SD大鼠随机分假手术组,缺血再灌注损伤模型组,芦丁高(80 mg/kg)、中(40 mg/kg)、低剂量组(20 mg/kg),共5组,每组10只。结扎冠状动脉左前降支建立心肌缺血再灌注损伤模型,心肌缺血再灌注后超声心动图评估心功能,测定各组大鼠心肌梗死面积,酶标仪检测血清及组织心肌酶生成量和氧化指标活性的变化,HE染色法观察组织细胞形态,免疫印迹法检测血红素加氧酶-1(HO-1)的表达。结果:与模型组比较,芦丁治疗组大鼠左心室射血分数及左心室短轴缩短分数升高,心肌梗死面积减小,血清肌酸激酶同功酶、乳酸脱氢酶、谷草转氨酶水平和丙二醛含量降低,超氧化物歧化酶活性和谷胱甘肽含量升高,心肌内HO-1表达增多(P均<0.05)。结论:芦丁可提高HO-1的表达、减少和清除氧自由基,起到保护心肌缺血再灌注损伤的作用。
Aim: To investigate the cardioprotective effect of rutin on myocardial ischemia-reperfusion injury in rats.Methods: Male SD rats were randomly allocated into Sham operation group, myocardial ischemia-reperfusion group(MI/R group),rutin high dose group(RH group,80 mg/kg), rutin middle dose group(RM group,40 mg/kg), and rutin low dose group(RL group,20 mg/kg)(n=10). Left anterior descending coronary artery was ligated to establish the myocardial ischemia-reperfusion injury model. Echocardiography evaluation of cardiac function after myocardial ischemia-reperfusion injury was performed. The area of myocardial infarction was determined. The myocardial enzymes(CK-MB, LDH, and AST)production and oxidation indexes(SOD, GSH and MDA)activity in serum and tissue were determined with enzyme-labeled instrument. The cell morphology was observed by HE staining. The expression of heme oxygenase-1 enzyme(HO-1)was detected by Western blot.Results: Compared with the MI/R group, EF and FS in the rutin groups were significantly increased, the area of myocardial infaction and myocardial enzymes were significantly decreased, the production MDA was significantly decreased, the activity of SOD and the production of GSH significantly increased, and the expression of HO-1 in the rutin groups was markedly increased(P<0.05).Conclusion: Rutin could increase HO-1 expression, therefore promote the endogenous antioxidant system against the myocardial ischemia-reperfusion injury in rats

Abstract: 目的:探讨左西孟旦对LPS诱导的乳鼠心肌细胞损伤的保护作用。方法:将原代培养的SD乳鼠的心肌细胞分为3组:对照组、模型组和左西孟旦干预组。模型组和左西孟旦干预组用10 mg/L的LPS处理6 h建模,其中左西孟旦干预组于造模前用终浓度为0.30 μmol/L的左西孟旦预处理24 h。全自动生化仪检测3组细胞培养液LDH、CK含量; ELISA检测培养液TNF-α、IL-6含量; 流式细胞仪检测细胞凋亡; Western blot检测凋亡相关蛋白Cleaved-Caspase-3及抗凋亡蛋白Bcl-2的表达。结果:与对照组比较,模型组细胞培养液中LDH、CK、TNF-α和IL-6含量增加,细胞凋亡率升高,凋亡蛋白Cleaved-Caspase-3表达增高,抗凋亡蛋白Bcl-2表达降低(P<0.05); 与模型组比较,左西孟旦干预组细胞培养液中LDH、CK、TNF-α、IL-6含量减少,细胞凋亡率降低,Cleaved-Caspase-3表达量降低,Bcl-2表达增高(P<0.05)。结论:左西孟旦对LPS诱导的乳鼠心肌细胞损伤有保护作用,其机制可能与减少炎症因子释放,抑制Caspase-3蛋白的表达,促进Bcl-2蛋白的表达有关。
Aim: To investigate the protective effect of simdax on lipopolysaccharide(LPS)-induced neonatal rat cardiomyocyte injury.Methods: The neonatal rat cardiomyocytes were allocated into 3 groups: control group, LPS group and LPS+simdax group. The cells in LPS+simdax group were pretreated with simdax(0.30 μmol/L)for 24 h. The cells in LPS group and LPS+simdax group were incubated with LPS(10 mg/L)for 6 h. The contents of LDH and CK in the culture medium were tested by automatic biochemistry analyzer,and those of TNF-α and IL-6 were evaluated by ELISA; the apoptosis was determined by flow cytometry; the expressions of Cleaved-Caspase-3 and Bcl-2 were measured by Western blot.Results: Compared with control group,the contents of LDH, CK, TNF-α and IL-6 as well as the apoptosis rate in LPS group were elevated significantly; the expression of Cleaved-Caspase-3 was increased apparently while that of Bcl-2 was reduced significantly(P<0.05). However, compared with LPS group, the contents of LDK, CK, TNF-α and IL-6 and the apoptosis rate in LPS+simdax group were significantly decreased, the expression of Cleaved-Caspase-3 was reduced, and that of Bcl-2 was increased(P<0.05).Conclusion: Simdax may inhibit the LPS-induced neonatal rat cardiomyocyte injury via decreasing the production of proinflammatory cytokines, down-regulating the expression of Caspase-3 and up-regulating the expression of Bcl-2

Abstract: 摘要 运用纳米金复合探针结合蛋白芯片, 建立了一种检测心肌损伤标志物的新方法. 构建了2种纳米金探针:标记有检测抗体和DNA探针1的检测探针和标记有DNA探针2(与DNA探针1的碱基互补配对)的信号探针. 当目的抗原存在时, 检测探针经检测抗体和抗原, 与芯片上捕获抗体结合固定在芯片上, 信号探针通过碱基互补配对与检测探针结合使信号放大, 最后利用纳米金成核原理染色, 通过显微镜观察结果并定量分析. 该体系在40 min内可检测多种标志物, 其中肌钙蛋白Ⅰ(cTnⅠ)的检出限为10 pg/mL, 与临床电化学发光法(ECLIA)灵敏度相当; 肌红蛋白(MYO)与新型脂肪酸结合蛋白(HFABP)的检出限分别为640和10 pg/mL, 与ECLIA及酶联免疫吸附法(ELISA)相比, 灵敏度显著提高
“关系圈”稀释“受害者圈”:企业环境污染与村民大多数沉默的乡村逻辑—山东N村个案研究

Abstract: 基于山东N村手套加工厂及其环境影响的调查发现,村落内生企业的关系嵌入与乡村社会分化的叠加效应,是大多数村民面对环境危害而保持沉默的乡村逻辑。乡村作为兼具乡土底色与经济理性的生活共同体,被企业污染建构为具有统合性的整体受害者圈。“同住一个村”的共同体意识形塑大部分村民的环境沉默行为与少数积极分子环境抗争的自我克制,而企业生产建构的获利者群体进一步分化了受害者圈层,村落“双重圈层”互嵌式关系格局阻隔村民通过集体行动制止企业污染的底层路径。农村内生污染企业的嵌入属性加深了村落社会分化、侵蚀了农村环境治理的社会基础,更提醒关注农村污染企业的社区嵌入与村民环境行为选择的复杂性关联。
Based on the investigation of N village in Shandong Province, it is found that the relationship between the village"s endogenous enterprise and the rural social differentiation is the rural logic that shaping the silence of the majority of the villagers in the face of environmental hazards. Rural as the community,with both local and economic rationality, but also the construction of corporate pollution as a whole with the overall community of victims. The community sense of "Living in a village" shaping silence Behavior of plastic most of the villagers and minority activists environment resistance self denial, but the enterprise production construction profit groups further differentiate the victim circle, the interlocking relationship pattern of village "double layers" barrier the bottom path of villagers" collective stop polluting enterprises.
