Abstract: In this review of methods for purity assessment of isolated chromosome X- and Y-bearing sperm, we compared the principles, operating procedures, as well as pros and cons for various methods. We conclude that nested PCR of single sperm will become a conventional and popular method with lower costs, and the method will play a very important role in optimizing the X, Y sorting method, if the sensitivity and accuracy of the method can be increased and the testing time decreased, and promote the new progress in other genetic testing techniques on single sperm.

Abstract: ?采用精子计数,精子活动率,精子形态学指标,观察了槟榔水提取液对雄性小鼠精子的影响。结果发现在3.75g/kg.bw剂量时,能使小鼠精子数量显著减少(t=1.8116,p<0.05);在7.50g/kg.bw、15100g/kg.bw时,精子活动率明显降低(u值分别为2.8142、3.8966,p<0.01);在剂量7.50g/kg.bw、3.750g/kg.bw时,精子畸形率明显升高(χ2值分别为22.8837、59.50,p<0.01);且以无钩、不定形与胖头增加为主。提示槟榔对雄性小鼠生殖功能可能造成一定影响。

Abstract: Abstract: Our early study found that goat spermatozoa could spontaneously take up foreign DNA and vary in capabilities of spermatozoa from different donors to bind and internalize exogenous DNA. In this study, three goats with considerable differences of capability were used to investigate the effect of exogenous DNA on goat spermatozoa, and feasibility and efficiency of transgenic embryo production by sperm-mediated gene transfer method. The viability, acrosomal reaction fre-quencies and cleavages were decreased in the groups co-cultured with exogenous DNA, compared with the control groups, and the range of decrease was correlated with the capability of sperm cells up-take foreign DNA. After fertilizing with co-cultured spermatozoa, GFP gene was introduced into oocytes and expressed in early embryos. However, different effi-ciencies of transgenic embryos appeared in sperm donors (P<0.05). GFP gene was detected in 16.2% (25/154), 5.3% (4/76), and 0% (0/36) embryos, respectively, when high, middle and low capability of sperm donors were used. But only 6.5% (10/154) embryos from high capability sperm donor expressed GFP. Our results demonstrate that selecting high capability of sperm donor is a key step for improving efficiency of sperm mediated-gene transfer method. However, the adverse influ-ence of foreign DNA on spermatozoa needs to be further studied.

精子载体法制备转基因动物的研究进展

Abstract: 精子载体法是一种低成本、简单快速制备转基因动物的方法。外源基因主要通过两种方式实现基因整合,一种是直接整合到精子的基因组中;另一种是通过精子携带进入卵细胞,然后整合到受精卵的基因组中。尽管现在已经有很多利用精子载体法获得了转基因动物的研究,但是基于精子在受精过程中,结合、内化、以及转运外源基因等方面能力的差异,转染效率仍有待提高。就利用精子载体法制备转基因动物过程中,精子载体法制备转基因动物的分子机制和方法方面进行系统的阐述和分析。
冷冻人精子的染色体畸变分析

Abstract: ?背景与目的:人精液冷冻是辅助生殖技术的重要环节。自国家批准设立人类精子库,冷冻精子在辅助生殖中大量应用,对冷冻保存后人精子染色体的畸变研究就更加重视。本文就冷冻保护剂(cryoprotectivemediem,cpm)对人精子染色体畸变进行了分析。材料与方法:分为4个冷冻保护剂组。ⅰ组:甘油-卵黄柠檬酸型(g-y-c);ⅱ组:甘油-蜂蜜型(g-h);ⅲ组:甘油-hepes-hty型(g-h-h);ⅳ组:甘油-tyrede型(g-t)。将加入cpm的人精子采用缓冻法处理,于冻后半年取出,以37℃水浴复温后作人精子染色体畸变分析。结果:4组cpm的精子染色体断裂率和畸变率均没有显著提高(p>0.05)。结论:这4种cpm在临床上应用是可靠的。
