A preliminary exploration into the inquiring teaching mode of de-velopmental biology

Abstract: Modern developmental biology is changing everyday at a fast pace, and it also possesses the most application potential. As an important selected course for undergraduates, its most up-to-date knowledge challenges the traditional teaching method. We discussed the current situation in developmental biology teaching, introduced in detail our method of using original papers in each research area, especially the classical papers that won the Nobel Prizes, and summarized the advantages of combining students’ presentation in the classroom and their discussion on the web. We proposed some practical strategies to improve developmental biology teaching.

Abstract: 本世纪70年代以来,随着生物细胞组织培养、DNA重组和转基因技术等一系列现代生物技术的不断改进和完善,生物技术已经成为当今世界发展最快、最活跃和最具潜力的高新技术领域之一。从我国农业生产的现状和发展趋势来看,仅仅利用传统的常规育种方法已经很难满足我国农业生产对作物新品种的要求,因而借助于农业生物技术与常规育种方法相结合的方式将会创造出更多的新种质,进而培育出更多高产、优质和多抗的新品种。作物生物技术育种所研究的主要内容涉及到在生物体内的细胞组织、染色体和基因等方面对其遗传基础进行改造和改良,以便获得具有更大增产潜力的作物新品种。

Abstract: We have developed a method for sorting specific cells in plant. Here, WER::GFP transgenic lines in Arabidopsis thaliana were as the experimental material; laser scanning confocal microscopy was used to identify the expression location of GFP; enzymatic digestion was applied for preparation of root tip protoplasts; fluorescence activated cell sorting (FACS) was applied to collect GFP positive cells; and then the total RNA was extracted from these cells. The results showed that only the early developmental non-root hair cells in the root epidermis express GFP in Wer::GFP transgenic lines; larger number of root tip protoplasts were obtained by using enzymatic digestion; and then GFP positive cells were collected successfully by FACS. The RNA extracted from these specific cells can be used for gene expression profiling. The method for sorting specific cells had been established successfully.

Abstract: 介绍由Lagrange实验、分析及拟合,得到未知材料的动态本构方程的方法.实验在轻气炮上进行,测量多点应力历程,经Lagrange分析得到数值本构关系,再拟合得到可供设计计算使用的动态本构方程.给出酚醛玻璃钢受冲击载荷时,一维应变状态下的一个本构方程.方程计算值与实测结果一致.

Abstract: ?建立了应用流式细胞仪分选植物特定类型细胞的方法。以拟南芥(Arabidopsisthaliana)Wer::GFP转基因株系为材料,用激光共聚焦显微镜鉴定GFP的表达位置,采用酶解法制备拟南芥根尖原生质体,应用流式细胞仪荧光激活细胞分选技术(FACS)分选收集GFP阳性细胞,并提取细胞的RNA。结果表明,Wer::GFP转基因株系仅在根表皮发育早期的非根毛细胞中表达GFP;利用酶解法制备的根尖原生质体数目较多;从FACS分选收集的细胞中提取的RNA质量较好,可用于研究特定类型细胞的基因表达谱。应用流式细胞仪分选拟南芥非根毛细胞的方法为研究植物特定类型细胞的基因表达谱及基因功能奠定了技术基础。
DELLA蛋白参与拟南芥幼苗对一氧化氮逆境的抵抗

Abstract: ?DELLA蛋白是赤霉素信号途径中的一类对植物生长起抑制作用的重要蛋白质,在拟南芥(Arabidopsisthaliana)基因组中已经鉴定出5个DELLA蛋白基因。目前研究发现,DELLA蛋白在抗逆中也起了重要的作用。近年来,一氧化氮(nitricoxide,NO)的研究工作取得重要进展,低浓度的NO能够促进植物的生长,但在高浓度下它对植物生长起抑制作用甚至导致细胞死亡。通过外施一氧化氮供体硝普钠(sodiumnitroprusside,SNP),研究高浓度NO对拟南芥幼苗生长的影响,发现植物体内H2O2积累,幼苗死亡。通过研究DELLA蛋白基因表达的变化及其相关突变体的表型,证明DELLA蛋白在抵抗NO逆境中起了重要作用。研究结果揭示了DELLA蛋白与NO逆境的关系,为今后科学指导农业生产提供了理论依据。
南疆近55年棉播期气温变化的小波分析

Abstract:
本文以南疆区域15个代表气象站1961~2015年4月份(即棉播期)的月平均气温观测数据为基础,利用线性趋势分析、Morlet小波分析等方法,分析近55年来南疆区域棉播期平均气温变化趋势、平均气温时间序列的小波周期和方差变化特征。结果表明:近55年,南疆区域棉播期平均气温呈现明显的上升趋势,升温速率为0.32℃/10a,但年际间波动明显;棉播期平均气温主要存在4个明显的周期变化,分别是6~8a、12~14a、16~20a、24~27a,其中6~8a的振荡周期最为显著;小波方差有3个峰值:7a周期的振动最强,其次是4a,再次为18a,7a为近55年时间段的主要周期。
Based on the data of April, i.e. cotton seeding period, average temperature of Southern Xinjiang from 1961 to 2015, linear analysis, Morlet Wavelet analysis are used to reveal the average tem-perature trend of cotton seeding period and the characteristics of Wavelet change in recent 55 years. The results show that the average temperature of cotton seeding period increases annually and the climate tendency rate is 0.32?C/10a. There are four characteristics on time scale: 6 - 8a, 12 - 14a, 16 - 20a and 24 - 27a. There are also three peaks of variance, 7a, 4a and 18a, and the charac-teristic time scale of 7a is dominant period in recent 55 years.
饲草专用谷子生长动态及最佳刈割期研究

Abstract: 谷子在我国农牧结合区和牧区正在发展成为专用饲草作物,但对谷子饲草的生长发育和营养变化动态缺乏研究,对最佳刈割期不了解,影响了谷子饲草生产和产业发展。本研究以饲草谷子1号为材料,研究了饲草谷子株高、粗蛋白、粗脂肪、粗纤维、中性洗涤纤维和酸性洗涤纤维等生长和变化动态,结果表明:从拔节至开花后一周的灌浆初期,是饲草谷子鲜质量和干质量的主要形成期,并在灌浆初期鲜质量和干质量达到最高,分别为33.0t/hm2和12.80t/hm2。饲草谷子粗蛋白含量在幼苗期含量最高为干草质量的22%,随发育进程的推进而逐渐减低,到乳熟期干饲草粗蛋白含量降为7.3%;饲草谷子粗脂肪的含量随生育期的进程表现了前期含量低,中期含量高,后期含量再降低的动态趋势。单位面积谷子饲草的粗蛋白和粗脂肪产量均在开花后1周的灌浆初期达到最高,以后开始迅速降低。粗纤维、中性洗涤纤维和酸性洗涤纤维也在开花后的灌浆初期达到最大产量。综合本研究的结果,开花后7d左右是饲草谷子的最佳刈割期,结合生产中的实际情况,从开始开花至群体开花结束的灌浆初期是饲草谷子的适宜刈割期。
