Abstract:
巨噬细胞是重要的天然免疫细胞,在抗细菌感染等免疫应答中发挥重要调控作用。当细菌等病原微生物感染机体时,准确的定量评价巨噬细胞的非调理素吞噬能力,常常被作为评价机体非特异免疫应答能力的重要指标。本文采用流式细胞仪结合细菌计数的方法,定量的检测了定居巨噬细胞和炎症浸润的巨噬细胞对细菌的非调理素吞噬能力变化。该研究方法将有助于定量评价在免疫应答中巨噬细胞的吞噬能力变化,以更准确反映在各种生理或病理条件下巨噬细胞的免疫应答状态。
Macrophage is an important component of innate immunity, which plays a great role in anti-infection immunity. The accurate evaluation of non-opsonic phagocytosis of macrophages is often used as an important indicator of the non-specific immune responses when bacterial infection occurs. In the present study, the non-opsonic phagocytosis of macrophages in resident or inflammatory infiltrating condition was quantificationally detected by using flow cytometry method combined with bacterial counts. This method will contribute to the quantitative evaluation for the ability of macrophage phagocytosis and the accurate response to macrophage immunity in a variety of physiological and pathological conditions.

Abstract: We have developed a method for sorting specific cells in plant. Here, WER::GFP transgenic lines in Arabidopsis thaliana were as the experimental material; laser scanning confocal microscopy was used to identify the expression location of GFP; enzymatic digestion was applied for preparation of root tip protoplasts; fluorescence activated cell sorting (FACS) was applied to collect GFP positive cells; and then the total RNA was extracted from these cells. The results showed that only the early developmental non-root hair cells in the root epidermis express GFP in Wer::GFP transgenic lines; larger number of root tip protoplasts were obtained by using enzymatic digestion; and then GFP positive cells were collected successfully by FACS. The RNA extracted from these specific cells can be used for gene expression profiling. The method for sorting specific cells had been established successfully.

Abstract:
对葡萄染色体倍性的间接、直接鉴定方法进行了综述,并对各种方法的优越性和不足之处进行了评价。指出葡萄染色体倍性鉴定应因陋就简,早期采用简单鉴定和破坏性较小的鉴定方法,其后用准确的鉴定方法进行综合鉴定。对不同来源的葡萄倍性采用不同的鉴定办法,准确确定倍性为葡萄育种实践服务。
It summarized indirect and direct identification methods of grape chromosome ploidy, and eva-luated that the ploidy identification should be rough. In the early stage, we have to use the simple appraisal method with less destruction. Then, we should use the accurate method to do compre-hensive appraisal, adopting different means for grape ploidy from different sources. Determining the grape ploidy accurately can serve for grape breeding practice.

Abstract: 细胞凋亡是当今环境毒理学领域的研究热点之一.为了探讨甲醛能否诱导细胞凋亡,以Hela细胞为实验材料,通过不同浓度的甲醛处理24h后,应用形态学观察、四甲基偶氮唑蓝(MTT)实验和流式细胞仪检测了Hela细胞凋亡情况.实验结果表明低浓度甲醛(≤0.2mmol·L^-1)处理Hela细胞24h后,对细胞生长有显著的抑制作用(p<0.05),但对细胞形态改变不大,且对细胞核DNA没有损伤;而高浓度甲醛(≥0.4mmol·L^-1)处理Hela细胞24h后,对细胞生长有极显著的抑制作用(p<0.01),细胞形态逐渐变的模糊不清,并有少量细胞萎缩成圆形,且会造成细胞核DNA有规律的断裂,细胞凋亡率随甲醛浓度的增加而增大.以上实验结果显示,高浓度甲醛处理Hela细胞24h后,会诱导细胞凋亡;并且随甲醛浓度的升高凋亡率逐渐增大,呈一定的剂量-效应关系.

Abstract: The chromosome abnormalities of blastocysts (6 d ~ 8 d ) produced by IVF(in vitro fertilization) with flow cytometrically-sorted, stained and unsorted sperm from 3 bulls were investigate in this study, which determine if the sorting procedure and staining cause the chromosomal abnormality of embryos. The results revealed that there were no significant differences in chromosomal abnormality (mosaic) rates of blastocysts derived from sorted, stained unsorted and unsorted sperm (40.7%, 59/145; 35.8%, 40/106 and37.0%,37/100, respectively); Chromosomal abnormality rates were variant among bulls (30.0% vs 44.6%; p<0.05); These results indicated that: 1) neither sorting procedure nor staining affect the chromosomal composition of embryos; The chromosomal abnormality rates of embryos were variable among bulls.

Abstract: 葡萄风信子是一种著名春季观赏球根花卉,具有重要的经济价值和生态价值。但由于其常见品种的倍性存在争议,严重限制了该植物科研和遗传育种工作的进展。以6种典型的葡萄风信子品种为材料,通过染色体常规压片法鉴定葡萄风信子蔚蓝为二倍体(2n=18),后以葡萄风信子蔚蓝作外标,进一步利用流式细胞仪鉴定其他5种品种葡萄风信子的倍性。结果表明,‘黑眼睛’、‘日出’为二倍体,‘白色丽人’、‘海洋的魔法’为三倍体,亚美尼亚为四倍体。其中葡萄风信子蔚蓝、‘黑眼睛’、‘日出’、‘白色丽人’和‘海洋的魔法’均为首次报道,希望为后期葡萄风信子的品种选育工作提供参考。
Grape hyacinth is a popular ornamental bulbous in spring,with great economic and ecological valueS.However,the dispute of ploidy of its common cultivars have seriously restricted the progress of its research and genetic breeding.Six typical grape hyacinth varieties were used in this study.First,Muscari azureum was identified diploid (2n=18) through conventional pressing plate method.M.azureum were then used as external standard,other five grape hyacinths were identified with flow cytometry.The results showed that M.armeniacum ‘Dark Eyes’ and M. ‘Pink Surprise’ were diploid,M.aucheri ‘White beauty’ and M.aucheri ‘Ocean Magic’ were triploid,M.armeniacum was tetraploid.Five of them were first reported

Abstract: 流动室堵塞是流式细胞仪使用中常见的问题,可造成检测误差或检测失败。本文根据流动聚焦原理,分析了流动室堵塞时的压力变化情况,据此提出了通过检测样本流压力升高判断流动室堵塞,并利用液体反冲疏通被堵塞流动室的方法,设计并搭建了实验系统。实验表明,该方法可以有效解决流动室堵塞问题。
In order to solve the problem of the micro flow cell clogging, and to improve the reliability of the flow cytometry system, a new method was proposed for hydrodynamic self-cleaning system. By analyzing the flow cell focus principle, we considered that to obtain stable single cell flow, the stable pressure in the flow chamber must be ensured. Therefore, we established a diagnosis method of clogging by the pressure detecting, and designed a self-cleaning system. Then we built up corresponding experimental systems. Experiments and testing showed that the self-cleaning system could improve the flow and resolve the clogging problem.

Abstract: We observed the heterogeneity of consensus interferon (cIFN) expressed by Pichia Pastoris, such as aggregation and degradation. The effect of induction pH on degradation of cIFN was studied in a 5-L bioreactor. The heterogeneity of cIFN was less serious when induced at pH 4.0~5.0. The bioactivity of the fermentation supernatant reached 2.5×108 IU/mL when pH was controlled between 4.0 and 5.0. To seek the reason of cIFN degradation, we analyzed the activity of protease and cell mortality by flow cytometry. At lower pH, cell mortality was higher and more protease was released. Protease activity increased at higher pH. The proteolytic degradation of cIFN was enhanced at either high or low pH. The cIFN was degraded entirely when pH was 7.0, where the protease had its optimal pH.

Abstract: Regeneration plants were initiated from bamboo (Dedrocalamus latiflorus) anthers, and chromosome number and nuclear DNA content were detected by chromosome analysis and flow cytometry. Among 100 regeneration plants, 4 were hexaploid, and the chromosome number of the other plants was twice that of the parents. These results demonstrate the anther culture of bamboo and have importance for functional genomics research and genetic improvement..
