急性酒精中毒的鼠脑内乙醛及其单胺神经递质缩合物的测定

Abstract: 应用高效液相色谱法测定了急性酒精中毒后新生鼠脑中乙醛的含量,以及乙醛分别与多巴胺、5-羟色胺的缩合产物1-甲基-6,7-二羟基-1,2,3,4-四氢异喹啉(Salsolinol,Sal)和6-羟基-1-甲基-1,2,3,4-四氢-β-咔啉(6-OH-MTHβC)的含量。采用2,4-二硝基苯肼(DNPH)的乙腈溶液在60℃水浴中衍生15min提取脑匀浆中的乙醛,离心后进行色谱分离,检测波长为360nm,本方法简单准确,灵敏度高。应用本方法测定鼠脑组织中的乙醛含量时发现,急性酒精中毒后新生鼠脑内乙醛的含量在24h内显著高于盐水对照组(P<0.05)。同时,采用HSF5柱分离和库仑阵列电化学检测的方法对Sal和6-OH-MTHβC进行分析表明,乙醛浓度升高后,急性酒精中毒鼠脑内Sal和6-OH-MTHβC的含量显著升高(P<0.05)。乙醛的非正常代谢及内源性神经毒素的生成可能在酒精的神经毒性机理中发挥了重要作用。
傣百解对模拟微重力下大鼠脾淋巴细胞功能的影响

Abstract: 探讨模拟航天飞行失重状态下大鼠免疫功能的变化和傣百解的免疫调节作用.将Wistar大鼠分为空白对照组、吊尾组、吊尾加傣百解低、中、高剂量组.连续吊尾21d后比较各组大鼠脾淋巴细胞的增殖能力、免疫相关细胞因子分泌水平,以及脾淋巴细胞中NFκB和IκBα蛋白表达水平.结果显示与正常组比较,吊尾组大鼠脾淋巴细胞刀豆蛋白刺激后的增殖能力减弱,细胞因子IL-2、IL-6、IFN-γ释放减少,而且NFκB信号通路激活水平低于正常组.傣百解能够提高失重大鼠脾淋巴细胞的NFκB信号通路激活水平,诱导淋巴细胞增殖和释放细胞因子,具有改善免疫功能的作用.

Abstract: The CP4-EPSPS gene is widely used in herbicide-tolerant plants/crops all over the world. In this study, a method was developed by coupling liquid chromatography with high sensitivity to tandem mass spectrometry to quantify the amount of CP4-EPSPS expression in Nicotiana tabacum leaves. The quantification of protein was converted to measure the unique peptide of CP4-EPSPS protein. One peptide unique to CP4-EPSPS was synthesized and labeled with H2 18O to get 18O stable isotope labeled peptide. The peptide served as the internal standard. The validated method had good specificity and linearity. The intra-and inter-day precisions and accuracy for all samples were satisfactory. The results demonstrated that the novel method was sensitive and selective to quantify CP4-EPSPS in the crude extract without time-consuming pre-separation or the purification procedures.

Abstract: Aging refers to a multidimensional process that all changes were accumulated in a person over time. These aging changes are associated with progressive increases in the chance of disease and death. Thus, it is necessary to establish a model to screen biomarkers to characterize and evaluate aging degree. In this study, an in vitro aging model was set up by formaldehdye and human serum albumin (HSA), the most abundant protein in human plasma, based on Maillard Reaction. The liquid chromatography tandem mass spectrometry (LC-MS/MS) method with 18O-labeling technique was employed to quantify modification degree of peptides cleaved from HSA. This model was established and optimized for further quantitative biomarker study.

海糖平改善小鼠脂质代谢紊乱作用研究

Abstract: 研究射干茎叶提取物-海糖平(HTP)对高脂饲料诱导的小鼠脂代谢紊乱的改善作用.ICR小鼠连续12周高脂饲料喂养诱发脂质代谢紊乱.造模成功后分别灌胃给予海糖平50,100,200mg/kgBW,治疗4周,并以吉非罗齐胶囊(100mg/kgBW)作为阳性对照药.治疗结束后检测肝脏和血清中甘油三酯(TG)、总胆固醇(TC)含量,附睾脂肪组织重量等指标,同时做肝脏组织病理检查.结果显示海糖平能够降低肝脏中TG、TC水平及血清中TG水平,减轻附睾脂肪组织重量,减少肝细胞中脂滴的蓄积.证实海糖平具有改善高脂饲料喂养小鼠的血清和肝脏中脂质代谢紊乱的作用.
保肝I号对D-氨基半乳糖和脂多糖致小鼠急性肝损伤的保护作用研究

Abstract: 采用D-氨基半乳糖(D-GalN)联合脂多糖(LPS)诱导的小鼠急性肝损伤模型对保肝I号(BGYH)的保肝降酶及其可能机制进行研究.将小鼠随机分为正常对照组,模型组,BGYH低、中、高剂量组,水飞蓟素组和联苯双酯组.每天给药1次,连续9d,末次给药后除正常组均腹腔注射D-GalN/LPS建立急性肝损伤模型.结果显示高剂量BGYH可以显著降低小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,同时,肝组织切片观察结果表明高剂量BGYH使小鼠肝细胞坏死程度有所减轻.在细胞因子方面,中或高剂量BGYH均可明显降低肿瘤坏死因子(TNF-α)及白细胞介素6(IL-6)等炎性细胞因子的表达,并同时升高抗炎因子白细胞介素(IL-10)的含量.此外,小鼠肝脏中一氧化氮(NO)、诱导性一氧化氮合酶(iNOS)水平在给予BGYH后也有下降的趋势.研究结果表明,BGYH对D-GalN和LPS联合诱导的小鼠急性肝损伤具有保护作用,可能是通过抑制免疫因子的释放,调节免疫反应而实现.
龙血竭对Lewis肺癌小鼠放疗模型的辅助治疗效果

Abstract: 龙血竭具有一定的防辐射作用。通过在C57/BL小鼠右前腋窝皮下注射Lewis肺癌细胞,对小鼠进行4Gy全身一次性γ辐射建立Lewis肺癌小鼠放疗模型,并给予小鼠龙血竭,进行肿瘤生长、肿瘤转移和血象变化检测,研究了龙血竭对Lewis肺癌小鼠放疗模型的血象及肿瘤生长的辅助治疗效果。实验发现,龙血竭不会促进放疗Lewis肺癌小鼠的肿瘤生长和转移,并能够有效保护放疗Lewis肺癌小鼠的血小板。研究表明,龙血竭应用于肿瘤的放射治疗可以对辐射引起机体的血象损伤起到保护作用。
基于双剪强度理论的混凝土板极限冲切承载力计算方法

Abstract: 将混凝土板的冲切破坏简化为平面应变问题,采用双剪应力三参数强度准则对冲切破坏时的混凝土板剪压区的复合应力进行分析,得到了剪压面上的极限竖向剪应力,利用静力平衡条件求得混凝土板的极限冲切承载力计算公式.与国内外246个试件的试验数据进行比较表明,该公式的计算结果与试验结果吻合良好.计算公式的计算模式概念清晰,可供结构设计时参考.
自激振荡线性化舵机延迟补偿时间处理方法研究

Abstract: ?针对某单通道控制炮射导弹所采用的自激振荡线性化舵机,给出了一种有效的舵机延迟补偿时间处理方法。自激振荡线性化舵机的高频振荡特性使得对其延迟时间的测量和处理相当困难,为解决此问题提出采用弹体滤波去除舵机高频振荡所造成的不确定性,并由导弹法向加速度延迟确定舵机延迟补偿时间的方法。数据处理结果表明,采用所提出的方法能够有效地确定自激振荡线性化舵机的延迟补偿时间。还从舵机延迟补偿难易的角度对比了调幅方波和调幅正弦波两种舵机指令形式的优劣,得到了自激振荡线性化舵机对正弦波指令的响应特性优于方波指令的结论。
