抗A型肉毒毒素人源单链抗体融合蛋白的重组设计*

Abstract: 以获得的抗A型肉毒毒素单链抗体为模板,进行融合改构,将人IgG1的Fc片段连接到ScFv的C端,在大肠杆菌中实现抗体融合蛋白ScFv-Fc的表达,表达量30%以上,蛋白以包含体形式存在,经过体外变复性的抗体融合蛋白ScFv-Fc,进行Prote in G Sepharose柱亲和层析纯化,纯度达90%~95%。体外活性检测结果表明,重组抗体融合蛋白ScFv-Fc可以特异结合A型肉毒类毒素抗原,其相对亲和力近似于母本单链抗体,其稳定性高于母本单链抗体。
山羊生殖细胞特异报告载体pvasa-egfp的构建及表达

Abstract: 为监测成体干细胞向生殖细胞分化的过程,分析生殖细胞特异性dead-box家族atp依赖rna解旋酶vasa在不同日龄山羊睾丸组织中的表达情况并构建山羊生殖细胞特异性报告载体pvasa-egfp。通过免疫荧光及rt-pcr法监测vasa的表达情况,利用分子技术构建报告载体pvasa-egfp,脂质体法转染山羊骨髓间充质干细胞(bonemesenchymalstemcells,bmscs),经过视黄酸(retinoicacid,ra)诱导后,观察绿色荧光蛋白表达情况以鉴定该报告载体的有效性。免疫荧光结果显示,vasa在性成熟不同阶段的山羊睾丸组织中均有表达,rt-pcr结果表明vasa基因在3月龄、10月龄山羊睾丸组织中显著高于10日龄组。测序及酶切鉴定结果表明,扩增的vasa基因启动子片段成功连至n1载体,转染bmscs后经4d的ra诱导,发现有绿色荧光蛋白表达,表明成功构建了山羊vasa基因启动子调控的报告载体pvasa-egfp。以上结果表明,vasa基因在不同日龄山羊睾丸组织中均有表达,所构建的山羊生殖细胞特异性报告载体pvasa-egfp具有示踪山羊成体干细胞向生殖细胞分化过程的能力,为下一步监测山羊bmscs向生殖细胞分化的过程提供了鉴定和筛选方法。
瓦斯对冲击性煤样能量耗散的影响

Abstract: 基于煤的冲击倾向性分类及指数测定方法,进行了不同瓦斯压力下煤样冲击倾向性测定,应用能量积聚与耗散的方法,分析了不同瓦斯压力环境中煤样单轴压缩与循环加卸载过程中能量积聚与耗散关系。研究表明:随着瓦斯压力的增大,煤样的冲击倾向性指数减小,冲击倾向性降低;煤样破坏前储存的弹性能降低,破坏时耗散能增率呈非线性快速降低趋势;在循环载荷作用下,煤样在相同循环中耗散能随着瓦斯压力的增加而增加。在不考虑瓦斯膨胀能的情况下,瓦斯压力的增加降低了煤层冲击地压的破坏性。因此,高瓦斯矿井在深部开采中进行冲击倾向性评价时应考虑瓦斯因素。

Abstract: 以海河流域水土保持动态监测数据为基础,采用面向服务架构(SOA)的设计思想,综合应用3S技术、元数据、海量数据存储等技术和二次开发方法,构建流域水土保持监测数据管理系统。该系统实现了基础数据管理、动态监测、海河流域ET(蒸发蒸腾)、普查数据管理和监测数据对比等功能。其成功运行为水土保持工作提供信息服务和分析手段,实现了水土保持信息规范化、现代化的管理和共享,全面提高了水土保持机构管理和服务水平,同时为水土保持监测数据管理提供良好的信息平台和科学依据。
The data management system for soil and water conservation monitoring is based on the dynamic monitoring data of soil and water conservation in Haihe river basin, which adopts the design idea of the service oriented architectures (SOA), applies 3S technology, metadata, mass data storage technology and method of secondary development to construct basin soil and water conservation monitoring data management system. This system has realized the basic data management, dy-namic monitoring, ET (evaporation and transpiration), census data management and monitoring data comparison and other functions. The operation of the system provides information service and analysis means for soil and water conservation work, realizes the information standardization of soil and water conservation, improve the soil and water conservation organization management and service level comprehensively. At the same time, it provides a good information platform and scientific basis for the management of soil and water conservation monitoring data.

Abstract: VH and Vkappa genes were amplified from human ScFv B17 specific against botulinum neurotoxin serotype A (BoNTa). Gene sequence encoding 5'-terminal 12 amino acid of heavy chain constant region CH1, as a linker, linked VH and Vkappa to construct a new three-domain antibody molecule VH/Vkappa. VH/Vkappa was expressed at high level over 34% of total host cell proteins in E. coli. Recombinant protein were purified up to 95% by affinity column. As a result, recombinant VH/Vkappa could recognize and bind specific to BoNTa in ELISA. However, comparing with its parent ScFv, VH/Vkappa has higher relative affinity and stability.
一种氧化型染发剂的致突变性研究

Abstract: ?目的:检测一种染发剂的致突变性作为毒理学评价的一部分。方法:用ames试验和体外哺乳动物细胞(chl)染色体畸变试验来检测氧化型染发剂的致突变性。结果:在加s9mix条件下,每皿2500μg至5000μg对测试菌株ta98具有明显的诱变性,其回变菌落数是自发回变数的2~3倍,ames试验检测结果为阳性。体外哺乳动物细胞(chl)染色体畸变试验显示在加或不加s9mix条件下,当受试物终浓度为300μg/ml,600μg/ml和1000μg/ml,除了300μg/ml在加s9mix时染色体畸变细胞率未显著增加外(p>0105),其余浓度无论加或不加s9mix,染色体畸变细胞率与阴性对照组相比均有显著性差异(p<0105)。结论:受试的氧化型染发剂具有一定的遗传毒性。

治疗用药豁免(tue)规则初探―――首例tue案分析

Abstract: 治疗用药豁免(therapeuticuseexemption,tue)规则属于世界反兴奋剂机制下的5项国际标准之一。tue的申请涉及当运动员因服用治疗性药物导致兴奋剂检测结果呈阳性时,是否能够免除处罚的问题。该规则的产生源于实践案例:人们意识到,只要存在违禁物质就处罚运动员是苛刻的,某些疾病的用药必然导致兴奋剂物质存于运动员体内,但并无益于提高运动员赛事能力。通过对cas(体育仲裁院)公开的首次涉及治疗用药豁免规则案例的叙述,分析其产生的理论逻辑,在肯定其提出的意义之后,对其实质性内容进行批判,以期得到进一步完善。
大麦全粉对面团特性及面包焙烤品质的影响

Abstract: 本文以不同比例的大麦全粉(以下简称大麦粉)替代高筋小麦粉,研究添加10%~60%(m/m)大麦粉对面团特性及面包焙烤品质的影响。结果表明,随着大麦添加量的增加,大麦-小麦混合粉的湿面筋含量降低,从36.43%(对照组)降至11.43%(添加60%大麦粉);面团吸水率增加,形成时间、稳定时间及C2(弱化值)值显著降低,延伸性与最大抗延伸阻力也显著下降;面团微观结构也随之发生变化,添加大麦粉的面团组织变得粗糙,在20%~60%添加范围内,气孔壁出现断裂,且几乎观察不到完整的气孔结构;对于面包品质而言,添加大麦粉的面包比容降低、硬度增加、弹性减小、感官品质下降,同时面包内部气孔所占面积减少、孔洞所占面积增加、面包片亮度变暗。大麦粉添加量为10%~20%时,但其整体感官品质仍可被接受;而添加过多的大麦粉(30%~60%)时,面包的焙烤品质显著降低,面包特有海绵状的纹理结构逐渐消失。
High gluten-wheat flour (10%~60%, w/w) was substituted with whole barley flour (hereinafter referred to as barley flour), and, subsequently, the effect of the substitution on dough properties and bread quality was investigated. The results revealed that the addition of barley flour effected a corresponding decrease in the content of wet gluten in barley-wheat mixed flour from 36.43% (control) to 11.43% (with 60% barley flour). In addition, the water absorption capacity increased, while the dough development time, time for stability, C2 value (weakening value), dough extensibility, and maximum resistance significantly decreased with increase in the concentration of barley flour. Meanwhile, dough microstructures were observed to change with the addition of barley flour, displaying coarser structures. The addition of barley flour in the range of 20%~60% resulted in the breakdown of the gas chamber walls; in fact, intact gas chamber structures could hardly be found. The specific volume, springiness, and sensory quality of the bread decreased, while the hardness increased with addition of barley flour. Moreover, a decrease in the area of gas chambers, an increase in the hole area, and a decrease in slice brightness were observed. The addition of 10%~20% barley flour imparted an acceptable overall sensory quality to the bread. However, the quality of bread baking decreased significantly with addition of excess barley flour (30%~60%), with the complete disappearance of the sponge-like texture of bread.
