Notch家族成员遗传学变异预示乳头状甲状腺癌高复发风险
Genetic alteration of Notch family members as a predicting factor for poor prognosis in papillary thyroid cancer

Abstract: 摘要:目的 分析Notch家族成员在乳头状甲状腺癌(PTC)中的表达及预后的相关性,探讨其参与肿瘤复发的可能机制。方法 下载TCGA数据库中PTC患者的临床资料及mRNA Seq芯片数据,采用cBioPortal在线分析Notch家族成员在正常甲状腺组织及不同TNM分期PTC中的表达情况,其遗传变异与PTC无复发生存期的相关性,同时采用KEGG进行差异基因信号通路聚类分析。结果 在PTC组织标本中NOTCH1、NOTCH3、NOTCH4 mRNA较非甲状癌甲状腺组织表达上调,而NOTCH2表达无显著变化。将PTC标本组织根据TNM分期分组后,NOTCH1、NOTCH3、NOTCH4 mRNA表达呈现先升后降趋势,NOTCH2则相反。携带Notch家族成员遗传变异(包括基因扩增、删失、错义突变及mRNA表达异常)的PTC患者无病生存期较短。通过对差异基因进行信号通路聚类分析,发现Notch成员遗传学异常与包括PI3K/Akt及MAPK信号通路在内的多条肿瘤信号通路均存在交互作用。结论 Notch家族成员异常预示PTC高复发风险,相关机制可能与细胞代谢及PI3K/Akt、MAPK等信号通路活性异常有关。
ABSTRACT: Objective To verify the alterations of Notch family members (NOTCH1, NOTCH2, NOTCH3 and NOTCH4) in papillary thyroid cancer (PTC) samples to investigate the association between their mRNA expressions and clinicopathologic features of PTC and explore the potential of these alterations as a prognostic predictor. Methods The mRNA Seq and clinical data of the PTC patients were downloaded from TCGA database. The mRNA expression of Notch family members in PTC and normal samples was analyzed with cBioPortal. Meanwhile, we analyzed the association between Notch genetic alterations and disease-free survival of PTC. Cluster analysis of differential genes’ signaling pathways was conducted using KEGG. Results NOTCH1, NOTCH3 and NOTCH4 were upregulated at the mRNA level in PTC samples compared with normal thyroid tissues. However, no significant difference was observed in NOTCH2 mRNA expression. With the progression from Stage Ⅰ to Ⅳ, NOTCH1, NOTCH3 and NOTCH4 underwent first a rise and then a decline while NOTCH2 exhibited an opposite tendency. PTC patients with Notch alterations including gene amplification, deletion, missense mutation and aberrant mRNA expression (z-scorethreshold±2) had shorter disease-free survival. Notch family members’ genetic alternations interacted with multiple tumor signaling pathways including PI3K/Akt and MAPK. Conclusion Alteration of Notch family members can be a potential factor for predicting poor prognosis of PTC. The underlying mechanism might be related to alterations in cellular metabolism, as well as in PI3K/Akt and MAPK signaling pathways

Abstract: 摘要:目的 探讨载脂蛋白A5(apoA5)基因-1131T>C(rs662799)及1259T>C(rs2266788)单核苷酸多态性与代谢综合征的关系。方法 采用聚合酶联反应及限制性片段长度多态性分析方法测定110例代谢综合征(MS)患者及110例健康对照组的apoA5基因-1131T>C(rs662799)及1259T>C(rs2266788)二位点的基因型,并检测血糖、血脂等指标,比较不同基因型与MS患病风险的关系。结果 -1131CC基因型携带者体重指数(BMI)、甘油三酯(TG)较TC、CC基因型升高,存在统计学差异(P<0.05)。1259CC基因型携带者TG、糖化血红蛋白(HBA??1C)较TC、CC基因型升高,存在统计学差异(P<0.05)。-1131CC基因型携带者发生MS的风险是TT基因型携带者的4.5倍(OR=4.504, 95% CI:1.766~11.490, P=0.002),且都得到统计学支持。结论 apoA5-1131T>C(rs662799)基因多态性与MS的发病可能相关;而apoA5 1259T>C(rs2266788)基因多态性与MS的发病无明显相关性。
ABSTRACT: Objective To investigate the relationship between apolipoprotein A5 (apoA5) gene and metabolic syndrome (MS). Methods Polymerase chain reaction and technique of restriction fragment length polymorphism were used to determine the genotypes of apoA5 -1131T>C (rs662799) and 1259T>C (rs2266788) in 110 patients with MS and 110 healthy control subjects. Results The levels of BMI and TG of subjects with apoA5-1131CC genotype were higher than those of subjects with TC and CC genotype (P<0.05). The levels of TG and HBA1C of subjects with apoA5 1259CC genotype were higher than those of subjects with TC and CC genotype (P<0.05). The risk of MS in subjects with apoA5-1131CC genotype was 4.5 times higher than that in subjects with TT genotype (OR=4.504, 95% CI: 1.766-11.490, P=0.002). Conclusion The genetic polymorphism of apoA5 -1131T>C (rs662799) may contribute to an increased risk of MS while that of apoA5 1259T>C(rs2266788) has no obvious relationship with the occurrence of MS
复方丹参滴丸对大鼠糖尿病肾病早期的肾脏保护及作用机制

Abstract: 摘要:目的 探讨复方丹参滴丸对早期糖尿病肾病的保护作用及可能机制。方法 腹腔注射链脲佐菌素建立高血糖大鼠动物模型。造模成功的大鼠随机分为模型组、复方丹参组以及厄贝沙坦组,分别给予蒸馏水、复方丹参滴丸及厄贝沙坦灌胃。另设正常对照组以同体积蒸馏水灌胃。测定尿白蛋白、血肌酐及血糖;肾脏组织行PAS、Masson染色,电镜观察超微结构,免疫组化检测肾组织血管内皮生长因子(VEGF)、结缔组织生长因子(CTGF)、巨噬细胞趋化蛋白-1(MCP-1)的表达。结果 复方丹参滴丸大剂量时可减少尿白蛋白排泄率、减轻肾脏的病理变化。降低肾小球VEGF、CTGF、MCP-1蛋白表达水平。结论 复方丹参滴丸能有效改善糖尿病所致的肾脏损害,其保护作用可能与降低生长因子和炎症因子有关。
ABSTRACT: Objective To explore the effects of Compound Danshen Dripping Pills (CDDP) on early diabetic kidney disease in streptozocin-induced rats of hyperglycemia, and its underlying mechanism. Methods Hyperglycemic rat animal model was established by intraperitoneal injection of streptozotocin. The model rats were then randomly divided into model group, Compound Danshen Dripping group and irbesartan group. They were then given distilled water, compound Danshen Dripping Pills and irbesartan by intragastric administration, respectively. An additional control group received the same volume of distilled water instead. Urinary albumin, serum creatinine and blood glucose were determined; kidney tissues were observed by PAS and Masson staining; the renal ultrastructures were assessed by electron microscopy. The VEGF, CTGF, and MCP-1 expressions were immunohistochemically detected. Results Compound Danshen Dripping Pills in large doses reduced blood sugar and urinary albumin excretion rate. The renal pathological changes were ameliorated by Compound Danshen Dripping pills. VEGF, CTGF, MCP-1 protein expression levels were decreased. Conclusion Compound Danshen Dripping Pills can effectively reduce renal damage caused by diabetes, which may be related to the decreased expressions of VEGF and inflammatory factors

Abstract: 摘要:目的 研究1-磷酸鞘氨醇(S1P)对2型糖尿病(T2DM)小鼠胰岛β细胞的促进增殖、抑制凋亡情况和对血糖的影响,以及S1P受体S1PR1-3在T2DM小鼠胰腺中的表达定位,探讨S1P对糖尿病胰岛β细胞损伤的保护作用。方法 雄性C57BL/6J小鼠,高脂饮食联合小剂量链脲佐菌素(STZ)制备T2DM小鼠模型,随机分为S1P高剂量组(S1PH)、S1P低剂量组(S1PL)、糖尿病对照组(DC)及正常对照组(NC),每日予S1PH组和S1PL组S1P溶液100μg/kg及20μg/kg腹腔注射,DC组及NC组予同体积溶剂替代。给药3周后行葡萄糖耐量试验(IPGTT),胰腺组织HE及TUNEL染色,免疫组化检测胰岛中胰岛素、Ki67及S1PR1-3蛋白的表达。结果 S1P给药3周后,S1PL组及S1PH组空腹血糖及IPGTT 2h血糖较DC组轻度下降。胰岛组织HE染色及胰岛素免疫组化染色示S1PL组及S1PH组较DC组胰岛病变减轻。Ki67免疫组化染色示S1PL组及S1PH组胰岛β细胞增殖率较DC组明显升高(P<0.05);TUNEL染色示S1PL组及S1PH组胰岛β细胞凋亡率较DC组明显降低(P<0.05)。胰岛组织免疫组化示S1P受体S1PR1-3均表达于小鼠胰岛β细胞中,在胰腺组织外分泌部表达甚微或不表达。其中S1PL组、S1PH组和DC组S1PR1及S1PR2的表达均较NC组明显增多,各组间S1PR3表达差异无统计学意义。结论 S1P与其受体S1PR1及S1PR2结合能够显著改善T2DM小鼠胰岛β细胞形态,促进小鼠胰岛β细胞增殖、抑制凋亡,提示S1P对T2DM胰岛β细胞损伤有保护作用。S1P/S1PR信号通路在T2DM中发挥着重要作用,有可能成为未来药物治疗的新靶点。
ABSTRACT: Objective To explore the potential protective effects of sphingosine-1-phosphate (S1P) on pancreatic islet β-cell proliferation and less cell apoptosis in type 2 diabetic mice and to detect the localization and expression of S1P receptors in the islets of type 2 diabetic mice. Methods Mice with type 2 diabetes induced by high-fat diet with low-dose streptozotocin were randomly divided into 4 groups: high-dose S1P administration group (S1PH), low-dose S1P administration group (S1PL), diabetic model control group (DC), and normal control group (NC). Exogenous S1P was intraperitoneally injected daily at a dose of 100μg/kg for S1PH group and 20μg/kg for S1PL group for 3 weeks. The potential protective effects of S1P on β-cell proliferation and apoptosis were determined. Intraperitoneal glucose tolerance test (IPGTT) and homeostasis model assessment (HOMA) index were assessed to test the effects of S1P on islet function. Expressions of insulin, Ki67, S1P receptor isoforms (S1PR1, S1PR2 and S1PR3) proteins in pancreatic islets were tested by immunohistochemical staining. Results Three weeks after S1P administration, fasting blood glucose and 2-hour glucose after injection during IPGTT in the two S1P administration groups were decreased slightly compared to DC group. β-cell proliferation rate was higher and the apoptosis rate was lower in S1PH and S1PL groups than in DC group (P<0.05). S1P receptors S1PR1, S1PR2 and S1PR3 were positively stained in the islet cell cytoplasm and membrane. S1PR1 and S1PR2 proteins were significantly expressed in S1PH and S1PL groups and DC diabetic groups compared with NC group (P<0.01), while S1PR3 expression did not significantly differ among these groups. 大鼠急性心肌梗死模型建立的影响因素

Abstract: 摘要:目的 探讨在非人工通气条件下体质量、胸廓前后径与左右径的比例对大鼠急性心肌梗死(AMI)模型建立的影响以及梗死后左室功能的变化。 方法 按体质量将SD大鼠随机分为4组:A组30只(200~250g),B组90只(250~300g),C组60只(>300g)及D组20只(假手术组)。在非人工通气条件下,测量胸廓径线后,通过开胸结扎左冠状动脉前降支(LAD)建立心肌梗死模型;同时记录心电图变化,观察并测量AMI术后2周、4周的心室壁厚度、室壁运动情况及心脏功能的变化。术后2周开胸,观察心脏大体形态的改变;同时取心脏标本行HE染色并观察心肌组织超微结构。以上数据通过SPSS13.0统计软件进行分析。结果 ①经过30次实验后成功建立第1只大鼠AMI模型,100次实验后成模率逐渐稳定在83%左右;②与A组比较,B组、C组大鼠AMI成模率较高(P<0.05),B组、C组成模率无差异(P>0.05);③未发现大鼠胸廓的前后径与左右径的比值与成模率有相关性(P>0.05);④术后2周开胸,见缺血区心肌颜色发白,室壁运动减弱。HE染色及电镜下可见心肌细胞丢失溶解,心肌结构不完整,肌丝断裂,大量纤维及胶原组织增生,线粒体染色变深,嵴模糊、水肿;⑤与D组相比,心梗组术后2周、4周的左室舒张末期内径(LVEDd)和左心室收缩末期内径(LVESd)增加(P<0.05),左心室射血分数(EF值)和心率下降(P<0.05)。结论 本方法可以成功建立大鼠AMI模型并出现心功能的改变;大鼠体质量是在非人工通气条件下建立AMI模型的重要因素, 而胸廓的径线对模型成功率的影响还需进一步证实。
ABSTRACT: Objective To study the effects of weight and anteroposterior diameter-to-transverse diameter ratio on establishing a model of acute myocardial infarction (AMI) in rats without artificial ventilation and changes in left ventricular function after infarction. Methods Healthy SD rats were randomly divided into group A (200-250g), group B (250-300g), group C (>300g), and group D (control group). The left anterior descending (LAD) coronary artery was ligated to establish a model of myocardial infarction under spontaneous breathing condition immediately after thoracic lines were measured. And changes of electrocardiography were recorded after model establishment. At 2 and 4 weeks after AMI, we observed ventricular wall thickness and ventricular wall motion and measured the changes of cardiac function. Histomorphological changes and myocardial ultrastructure of the heart were observed under thoracotomy 2 weeks after operation. The above data were analyzed by SPSS13.0 statistics software. Results ① The first AMI rat model was established successfully after 30 times of experiments, and after 100 times the model??s success rate gradually stabilized at about 83%. ② Group B and group C had a higher model success rate than group A (P<0.05), but group B and group C did not differ in modeling success rate (P>0.05). ③ There was no association between the rate of rat thoracic line and modeling success rate (P>0.05). ④ Two weeks after thoracotomy, ischemic myocardial color was white, and ventricular wall motion decreased. HE staining revealed that cardiomyocytes disappeared and were replaced by fibrous tissues and collagen. Remnant cardiomyocytes were arranged disorderly and myofibers were fractured, with interstitial damage and hyperplasia of fibrous tissue. Visible muscle cells were sparse and dissolved, the mitochondria had darker staining,

Abstract: 矿渣微粉具有一定的水化活性,掺入水泥浆体后能够明显提高水泥抗化学侵蚀性能,抗干缩,抗碳化,抗冻性等耐久性能,提高幅度基本可以达到10%左右;利用SEM,TG-DTA,XRD,压汞孔分析等方法对水泥水化物进行分析,结果表明:矿渣微粉能够吸收水泥水化产物Ca(OH)2生成C-S-H凝胶,导致降低孔径,优化孔结构,从而提高水泥石的密实度,从而改善水泥的各项性能。
Experiment of cement with ground slag shows that ground slag can improve the properties of ce-ment such as coefficient of resistance to chemical corrosion, dry shrinkage, and carbonation coef-ficient, freezing and thawing resistance by 10%. The microstructure-analysis by using SEM, TG- DTA, XRD and MIP shows that the ground slag can absorb the productions of hydrating cement: Ca(OH)2, and C-S-H gel is formed, which can refine the distribution of pore structure and increase the density of cement-stone, therefore improving the durability.

Abstract:
针对风力辅助提水机,在风速随机变化的情况下,依据柴油机控制水泵转速恒定问题,建立了系统仿射非线性T-S模型。基于模糊滑模变结构方法,本文设计了系统水泵速度跟踪控制器,在保证水泵额定转速运行的同时,实现了风能的最大功率跟踪,充分利用了风能。计算机仿真结果验证了该控制策略可行性,这为以后进一步研究奠定了基础。
Under the condition of random variation in wind speed, for controlling the pump speed constant problem of wind power aided pumping water machine by diesel engine, T-S affine nonlinear system model is established. Based on fuzzy sliding mode variable structure method, the water pump speed tracking controller is designed. By guaranteeing pump rated speed running at the same time, the maximum power tracking of wind energy is completed in order to make full use of wind power. Finally, the validity of the proposed control strategy is revealed via computer simulations, which will be a foundation for more study.

动结式的核心问题一直是汉语研究的一大热点。近二三十年来的研究主要是从语义核心和形态句法特征方面展开的。本文认为,所有核心都应具有决定整体结构范畴归属的特征,决定动结式范畴归属的核心则是其形态句法核心。本文认同将动结式分为同指和异指两种类型的观点,并在Chomsky最简方案(MP)框架下通过轻动词v这一功能语类,在确定动结式功能核心和词汇核心的同时,还从句法结构上对这两类动结式进行了表征
What is the head of a Chinese V-R compound is a longstanding hot topic in Chinese study. In the last 20 or 30 years, the research on this topic has been done either from the perspective of semantic head or from morphosyntactic features. This paper holds that a head should have the feature of determining the whole construction in which it is in and that a head should be morphosyntactic. Following the view that Chinese V-R compounds can be divided into two types, namely homo-referential and hetero-referential, this paper aims to fix the head of a Chinese V-R compound through the functional head v within the framework of Minimalist Program and to provide the syntactic representations for these two types of Chinese V-R compounds.
