酿酒酵母全基因组范围内筛选mtm1基因的抑制基因

Abstract: mtm1基因对于维持锰超氧化物歧化酶的活性和线粒体正常功能十分重要,mtm1基因的缺失会严重影响酵母锰超氧化物歧化酶活性,并损伤线粒体功能,因此在非发酵培养基上不能生长.利用mtm1基因缺失的突变体在非发酵培养基上的生长缺陷,转入酵母基因组文库筛选mtm1抑制基因,发现mtm1基因缺失造成的损伤一旦形成不可逆转,重新引入mtm1基因也无法挽救,直接筛选无法得到抑制基因.为了避免mtm1缺失造成的不可逆损伤,在野生型酵母中先转入带有mtm1基因的质粒,再敲除染色体上的mtm1基因,随后转入基因组文库,再利用药物5-氟乳清酸(5-foa)迫使细胞丢失表达mtm1基因的外源质粒,再筛选能在非发酵培养基上生长的转化子,通过这种方法筛选发现,por2等5个基因的过表达可以挽救mtm1基因缺失造成的非发酵培养基上的生长缺陷,为深入了解mtm1基因的功能提供了线索,对筛选其他造成不可逆损伤的突变基因的抑制基因提供了一条可行的研究思路.

Abstract: ?强流脉冲离子束在靶上的能量密度分布是其研究和应用的一个重要参数。为了解决现有束流能量密度诊断方法的不足,利用红外成像仪测量靶背面的温度分布,从而建立了强流脉冲离子束能量密度分布的红外诊断方法。对能量密度1j/cm2量级、脉冲宽度为102ns量级的束流,该方法测量误差好于5%,空间分辨率可达到1~2mm,具有操作简便、成本低的优点,是强脉冲能量沉积分布的一种高效迅速的诊断方法。

Abstract: Objective] MTM1 gene is essential for superoxide dismutase 2 activity and normal mitochondrial functions. MTM1 deletion results in decreased superoxide dismutase 2 activity,impaired mitochondrial functions and growth defect on nonfermentable carbon source. Abstract:
随着高校数字化校园网络的广泛应用,校内机器增多,无线网络的覆盖,相应的IP地址分配也在不断增多,IP地址冲突现象与日俱增,在一定程度上影响了网络的正常运行。本文依据实践经验,从基础网络管理和网络技术入手,提出解决方案,为增强校园网网络管理和安全管理、提高维护效率提供参考。
With increasing applications of network digitalization and coverage of wireless network in the campus, relevant facilities and IP address distribution also increase. The IP address interference issue is affecting regular network operation. This paper is based on the practical experience to work out the solution for enhancing campus network management and security management, facilitating maintenance efficiency from basic network management and technology.

Abstract: 北宋是中国文学艺术创新发展的高峰期,从宋代开始中国古典美学审美意向出现了重大转折,文艺创作者的主体精神自此走向一个逐步解放的路途,这在某种程度上意味着“尚意”时代的到来。集文学家、书画家等多重身份于一身的苏轼是这一转型中不可回避的重要人物。禅宗是宋代文化中的重要组成部分,禅宗思想为苏轼提供丰富的人生智慧和精神源泉,禅宗作为一种哲学思想从创作思想、审美情趣等深层文化心理结构上深刻影响着苏轼的文艺创作与文艺理论。本文主要讨论禅宗关照下的苏轼“尚意”书法美学,通过揭示苏轼与禅宗的渊源,分析禅宗思想在苏轼书法“尚意”美学形成发展中的作用以及“尚意”美学对后世的影响。
In ancient Chinese history, the Northern Song Dynasty was the peak time of creative literature and arts development. Due to the changes of Chinese classical aesthetic intention in Song Dynasty, the authors’ subject spirits started to be emancipated and this signaled the birth of a new age called Shangyi which advocates significance and intention. It was Sushi, who was known as a famous litterateur and famous artist, that had made a huge contribution in many areas of art. As the result of imperial examination system growing mature and literati and officialdom awakening, Zen became the most important part of Song Dynasty culture. Zen brought Sushi various life wisdom and became one of the sources of his great personality. In the same time, as a philosophical thought and aesthetic experience, Zen had a profound impact on Sushi, especially in creating and criticizing literature from the angles of creation thoughts and aesthetic taste or other aspect in deep cultural psychological structure. This paper mainly discusses about Sushi’s Shangyi calligraphy aesthetics under the impression of Zen through the analysis of relation between Sushi and Zen. It is a brief analysis of the impact of Zen in the progress of Sushi’s Shangyi aesthetics and it ends the article with a simple introduc- tion in how Sushi’s Shangyi calligraphy influences the later generations.

Abstract:
本文对“阳光体育运动”背景下长三角地区主要大型城市中学课外体育活动开展的影响因素进行了调查分析,结果显示:应试教育、错误的教育价值观和现代体育理念的缺失;运动场地器材不足和体育经费投入不足;体育教师工作量过大、待遇低以及对学生缺乏应有的指导;家长对孩子过分的溺爱;运动能力差、找不到喜爱的运动项目、怕苦怕伤以及缺乏同伴,分别是影响中学课外体育活动开展的社会、学校、教师、家长和学生因素。
Under the background of “sunshine sport”, this study is to analyze the influencing factors about the development of extracurricular sports activities in secondary schools located in the main giant cities in Yangtze River Delta. The result shows there are various influencing factors which prevent extracurricular sports activities developing in communities, schools, teachers, parents and students. They are examination-oriented education, incorrect educational values and the lack of modern sports concepts; the shortage of sports field and equipment and the inadequate investment on sports; the overload and low-treatment of PE teachers and lack of guidance for students, parents’ excessive fondness; and the disadvantages among students such as poor exercise capacity, less favorite sports, fear of pain and hurt, and lack of companion.

Abstract: 将传统的k-?模型应用到RTI引起混合的数值模拟中,模型中起驱动作用的速度梯度由压力和密度梯度取代,给出了合理描述RTI诱导湍流混合特征的湍流产生项的表达式。采用该模型对Youngs和Kucherenko等的常加速度和变加速度条件下的RTI实验进行了数值模拟。通过与实验结果的比较分析,验证了所采用的模型封闭、模型常数、数值算法和程序实现是合适的。在此基础上,进一步探讨了轻重介质密度比、加速度大小以及稳定阶段持续时间对混合的影响。

Abstract: 采用电化学交流阻抗技术并结合物相分析技术研究了溅射CoCrAlY涂层对Ni3Al、Ni3Al-Fe金属间化合物在700℃熔融(Na,Li,K)2SO4中腐蚀行为的影响.结果表明,Ni3Al和Ni3Al-Fe的耐熔融盐腐蚀性能较差,其交流阻抗Nyquist图高频端表现为一曲率半径很小的圆弧,低频端为一直线,腐蚀反应受氧化剂的扩散所控制;施加CoCrAlY涂层后,Nyquist图则呈双容抗特征,发生了沿涂层缺陷的腐蚀;提出了交流阻抗模型并解析了其中的参数.

Abstract: 国际情绪图片系统的发展是为了提供大量标准化,能激发情绪且可以跨国使用的包含广泛语意类别的照片。国际情绪图片系统的研发者使用自我评定模型“一种非语文的图形评估技术”来测量参与者在图片的愉悦度、唤醒度和控制度上的反应。本研究的目的是检验自我评定模型与九点量表间的差异与相关。研究对象:26位受试者平均分为两组,每组针对72IAPS照片在间隔4周以九点量表与SAM量表分别进行评估,但两组施测顺序采对抗平衡设计。研究结果:对于年轻成人两种评估方式在愉悦度、唤醒度和控制度皆有良好的内部一致性,且两种评估方式的结果没有明显差异。结论与建议:两种方式在年轻成人评估IAPS时都适用,但本研究也存在受试者控制度评估变异较大,未能对年轻成人以外的群体进行两种方式比较的限制,建议欲应用本研究结果时应注意前述限制。
The International Affective Picture System (IAPS) was developed to provide a large set of standardized, emotionally evocative, internationally accessible photographs from a wide range of semantic categories. The researcher used the Self-Assessment Manikin (SAM), a non-verbal pictorial assessment technique, to measure the pleasure, arousal and dominance associated with the effects of IAPS on the participants.
