伏安型植物组织生物微电极的研究——茄子组织碳糊微电极的研究

Abstract: 茄子组织生物微电极在儿茶酚测定中表现出了较高的生物催化活性,而对抗坏血酸测定则为稳态响应,这表明该电极能有效地消除抗坏血酸的干扰.该电极的灵敏度高,重现性好,对多巴胺测定的线性范围为5.8×10-6~6.5×10-4mol/L,检出下限为:2.9×10-6mol/L.

Abstract: 由于香蕉组织中含有的氧化酶能够生物催化氧化多巴胺、去甲肾上腺素、L-多巴等,大大地提高了植物组织电极的灵敏度;又由于茄子组织中所含有的氧化酶在催化氧化儿茶酚的同时,能有效地抑制抗坏血酸在电极上的反应,将上述两种植物组织混合制成电极,使其更适于神经递质的测定,线性范围:2.4×10-6~9.8×10-4mol/L;检出下限3.2×10-7mol/L.
分子印迹纳米管膜的制备及对人体尿液中儿茶酚胺类药物的检测

Abstract: 摘要 以多巴胺(DA)为模板,多孔阳极氧化铝膜(AAO)为反应载体,合成了多巴胺分子印迹聚合物纳米管膜(AAO@MIP).利用扫描电子显微镜对分子印迹纳米管膜的形貌进行了表征,并用高效液相色谱(HPLC)研究了其对儿茶酚胺类(CLs)药物的吸附性能.实验结果表明,在最优萃取条件下,AAO@MIP纳米管膜对多巴胺、肾上腺素和去甲肾上腺素具有较高的选择性,3种儿茶酚胺类药物在0.50~300 μmol/L浓度范围内呈良好的线性关系(r2>0.9970);检出限(S/N=3)分别为15.5,12.6和22.5 ng/L.AAO@MIP纳米管膜对多巴胺的最大吸附容量可达82.1 μmol/g;6次吸附-解吸附重复利用后,吸附容量仅降低3.3%.将AAO@MIP纳米管膜应用于萃取人体尿液中3种儿茶酚胺,样品加标回收率为74.0%~100.4%,相对标准偏差(RSD)为3.6%~6.8%.该方法简便、快速、选择性高,适用于检测人体尿液中的儿茶酚胺类药物的含量

Abstract: ?制备甘薯多酚氧化酶丙酮粉,并以分光光度法研究了多酚氧化酶作用的最适条件、动力学性质以及抑制剂和激活剂对酶活性的影响.结果表明:[1]最适作用温度为15.5℃和50℃;[2]最适作用ph为4.5;[3]适宜底物为儿茶酚;[4]以cu2十为激活剂;[5]vc和so2为强烈抑制剂.

Abstract: This study is to explore the relationship between polymorphism of Catechol O-methyltransferase gene and schizophrenia.Search for a gene predisposing to schizophrenia in the Han nationality in China.Five pedigrees with high incidence of schizophrenia were studied by polymerize chain reaction(PCR) and restriction fragment length polymorphism(RFLP) technique.Statistics analysis of the transmission/disequilibrium test (TDT) showed that the COMT gene is associatted with schizophrenia in the five pedigree with high incidence schizophrenia( P =0.0455).The results suggest that there might have a schizophrenia liability gene on 22 chromosome (22q11.2) in our studied pedigrees.

Abstract: 目的:探讨儿茶酚胺氧位甲基转移酶(catechol O-methyltransferase,COMT)Val108/158 Met基因多态性与抗 精神分裂症药物疗效之间的关系,为抗精神分裂症药物的个体化用药提供一定的依据。方法:计算机检索PubMed, Embase,Cochrane Library,中国知网,万方,维普,中国生物医学文献数据库获取相关文献。在评价原始研究质 量,提取有效数据后,采用Stata12.0进行统计分析。结果:共筛选出9篇符合标准的文献,包含868例患者。Meta分 析结果显示各基因型合并后的RR值、95% CI及P值,以患者的总体症状改善情况来评价药物疗效时,Met与Val比较, RR=1.18,95% CI:1.04~1.35,P=0.013;Met/Met与Val/Val比较,RR=1.40,95% CI:1.08~1.82,P=0.010。以患者的阴 性症状改善情况来评价药物疗效时,Met与Val比较,RR=1.24,95% CI:1.05~1.46,P=0.013;Met/Met与Val/Val比较, RR=1.60,95% CI:1.04~2.46,P=0.031。结论:COMT Val108/158Met基因多态性与抗精神分裂症药物疗效显著相关, 其中Met基因为优势基因,对抗精神分裂症药物反应较好
萘降解菌的分离、鉴定及降解途径
Isolation and identification of naphthalene degrading bacteria and characterization of its degradation pathways

Abstract: 从武汉市金口张公堤处污泥富集分离出2株能以萘为唯一碳源生长的优势降解菌株XA1和XB1,经形态学观察、生理生化鉴定及16S rDNA测序分析,确定它们属于荧光假单胞菌 (Pseudomonas fluorescens)。其最适生长温度均为28℃,最适生长pH分别为7.5和7.0。在最适生长条件下,菌株XA1、XB1以3%的接种量对500 mg/L萘的降解率在第3天时分别达到了93.4%和74.7%。采用PCR和琼脂糖凝胶电泳技术检测到XA1、XB1中有儿茶酚2,3-双加氧酶基因(nahH)等萘降解过程中的关键酶基因,与NCBI中发布的基因序列相比较,相似度均达到99%。
Two strains,named as XA1 and XB1 utilizing naphthalene as the sole carbon source,were isolated from Zhanggong Dam of Jinkou in Wuhan City.XA1 and XB1 were both identified as Pseudomonas fluorescens through morphological characterizations,physiological and biochemical identification and alignment analysis of the 16S rDNA sequence.Their optimal growth temperature were both 28℃ and the optimal growth pH value were 7.5 and 7.0,respectively.When XA1 and XB1 were inoculated in the liquid inorganic salts medium with naphthalene as the sole carbon source,the degradation rates of naphthalene after three days were 93.4% and 74.7%,respectively.Catechol 2,3-dioxygenase gene and some other genes were characterized with the PCR method and agarose gel electrophoresis.Results showed that sequences of catechol 2,3-dioxygenase gene and the others all possessed 99% similarity with the sequences in the NCBI.
用高效液相色谱—电化学检测器测定人血浆中儿茶酚胺浓度

Abstract: 国内外文献已报道过很多测定儿茶酚胺的方法,作者在此基础上用反相离子对色谱电化学检测器,也达到了分离测定人血浆儿茶酚胺浓度的要求,结果与文献报道基本一致,可用于临床及科研中人血浆儿茶酚胺浓度的测定。
