Effect of multi-generation culture on virulence of Lecanicilliurn lecanii in different media
不同基质多代培养对蚧虫病原真菌蜡蚧霉致病力的影响

Abstract: Abstract: Objective] The strain No.V3.4504 of Lecanicilliurn lecanii (Zimmermann), an entomopathogenic fungus, was studied on the effect of successive multi-generation culture in seven different media on its colony growth characteristics, extracellular enzyme activities and the virulence against scale insects. Methods] The strain No.V3.4504 of L. lecanii was original isolated from a natural infected scale insect. The two species of scale insects used were Rhodococcus sariuoni Borchsenius and Ceroplastes japonicus Green. Seven media were used and fungus colony characteristics, growth rate and sporulation, extracellular protease and chitinase activity, and infective effect against the two species of scale insects were conducted. Results] The fungus cultured on PDA medium for successive nine generations showed the most fast in colony growth, the minimal in sporulation, straight decline of extracellular protease and chitinase activity with generation increasing, and the minimal mortality of the scale insects. There was no significant effect to promote virulence of the fungus by increasing peptone into medium. On the media D, E and F, that with the body materials of the two scale insects, although the fungus appeared lower in the colony growth rate, its sporulation was higher upward 8.83×106-9.13×106 spores/cm2, extracellular protease and chitinase activities averagely reached 2.16-2.13 U/g and 1.01-1.03 U/g respectively, and the mortalities of the two scale insects were 55%-58% and 39%-42% respectively. Cultured three generations in vitro of the two scale insects, the fungus exhibited the highest activities in its protease and chitinase that were 3.08 -2.92 U/g and 1.45 -1.42 U/g respectively and the best infection effect against the two scale insects with mortalities of 71.30% and 58.89% respectively. A linear correlation was found between extracellular protease and chitinase activities of the fungus and the mortalities of the scale insects. Conclusion] Cultured on PDA medium successive multiple generations made retrogradation of the strain No.V3.4504 of of L. lecanii. It was significant effect on keeping the vigor and higher virulence of the fungus adding the body materials of the scale insects into the medium. The vitro by using live scale insects as medium materials was the best way for the rejuvenation of the entomopathogenic fungus and promoting its virulence.
Isolation and identification of toxins inhibiting Dentrolimus tabulaeformis from an antagonistic strain of Beauveria

Abstract: Objective] We used submerged fermentation to cultivate a strain of the entomopathogenic fungus Beauveria tenella isolated from the infected larvae of Dentrolimus tabulaeformis in Pinus tabulaeformis forest in Chengde of Hebei Province in China Methods] We used ethyl acetate to extract antagonistic components from the fermentation broth and used silica gel column chromatography and GC/MS to separate and identify the components. Results] Six compounds were obtained by silica gel column chromatography. The sixth compound had higher activity to kill the larvae of Den-trolimus tabulaeformis with a corrected mortality rate of 80%. Seventeen compounds were separated and identified by GC/MS in the 6th group, of which 3compounds were more than 10%, 2-Piperidinone(14.02%), 2-coumaranone(47.10%), and Pyrrolo1,2-a]Pyrazine-1,4-dione, hexahydro (21.05%). Conclusion] 2-Piperidinone and 2-coumaranone had insec-ticidal activity (corrected mortality rate reached 83.32% and 91.61% respectively) and were the most important toxic sub-stances to control pests.
Comparison of Two Methods for Extracting Total RNA from Citrus Leaves

Abstract: 为了简便、快速提取高质量的柑橘(Citrus reticulata Banco)叶片总RNA,采用TRlzol法,对北京天根公司RNAplant Reagent和日本TaKaRa公司RNAiso Reagent两种RNA提取试剂进行比较并适当改进.结果表明:通过琼脂糖凝胶电泳,使用TaKaRa公司试剂提取的总RNA28S和18S条带清晰,紫外分光光度计检测分析A260/A280为1,820,A260,A230为2.088,RNA的浓度为2.840 μg μl-1,用于RT-PCR反应可成功克隆柑橘β-actin看家基因228 bp片段;采用天根公司试剂,琼脂糖凝胶电泳显示RNA带型较模糊,紫外分光光度计检测分析A260/A280为1.464,A260/A230为1.603,RNA的浓度为2.020 μg μl-1,达不到RNA的标准.TaKaRa公司RNAiso Reagent试剂提取的柑橘叶RNA纯度和完整性较好,能用于Northern杂交、cDNA文库的建立及基因克隆等分子生物学实验,为柑橘的进一步分子生物学研究奠定了基础.

Abstract: 采用扫描电镜和气相色谱/质谱联用技术研究了角蜡Ceroplastesceriferus(Fabricius)和日本龟蜡蚧C.japonicusGreen蜡泌物的超微结构及化学成分。结果表明,这2种蜡蚧分泌蜡质和形成蜡壳的过程基本相似。在1、2龄期分泌的蜡质为“干蜡”,蜡壳为星芒状,虫体周缘的蜡芒均为2大节,与其2个龄期发育相对应,每一个大节又分为若干小节。同时,虫体背面中央蜡质堆积成帽状,也分为均匀的多层。由此说明泌蜡过程具有节律性。虫体周缘与蜡芒对应的突起区上分布着密集的刻点状腺孔,每一个腺孔分泌1根蜡丝,这在以往玻片标本中是观察不到的。雌性第3龄幼虫和成虫期,虫体分泌“湿蜡”,形成龟背状蜡壳,泌蜡腺孔主要为三格腺和四格腺。在肛突区发现了密集的泌蜡腺孔,排列为纵条纹状。从角蜡蚧蜡泌物甲酯化处理样品中检测到14个组分,从直接测试(未经甲酯化处理)样品中检测到14个组分;而从日本龟蜡蚧则分别检测到10个组分和25个组分。它们的主要成分是一系列高级的长链饱和与不饱和的烃、脂肪酸、脂肪醇、酯类、醛类以及杂环、多环或大环状化合物。对它们可能的生物生态学功能进行了讨论。
瘤坚大球蚧蜡泌物的超微形态与红外光谱特征

Abstract: 采用扫描电镜和红外光谱技术研究了瘤坚大球蚧Eulecaniumgigantea在不同发育阶段蜡泌物的超微形态和化学成分。结果发现:该虫在低龄期分泌湿蜡,最初在虫体背面凝结成晶体状的不同大小颗粒,接着连接成片状和块状构造,最后蜡质堆积形成龟背状蜡壳。蜡壳的各个板块与体壁上突起的腺孔分布区位置相对应。雄虫蜡壳与低龄期若虫蜡壳质地相同。雌成虫分泌的蜡质为长而卷曲的空心蜡丝。雌成虫背面和腹面蜡泌物的红外光谱特征说明二者所含的主要化学基团和成分是相同的。尽管雄虫蜡壳与雌虫的化学成分基本相似,但存在的官能团有明显差别,主要表现在雄虫蜡泌物光谱图上1736.3cm-1处出现两个吸收峰,而雌虫只有一个;在振动频率1242.0cm-1~1106.6cm-1出现3个C-O键振动峰,而雌虫的是一系列锯齿状长碳链-C-C-C-骨架振动峰。根据红外光谱特征分析,推测该虫蜡泌物的化学成分可能主要是由长链烃、长链脂肪酸、脂肪醇和某些带芳香环的化合物组成。
朝鲜毛球蚧蜡泌物的超微形态与红外光谱特征

Abstract: 采用扫描电镜和红外光谱技术研究了朝鲜毛球蚧Didesmococcuskoreanus在不同发育阶段蜡泌物的超微形态和化学成分。结果发现:该虫在低龄期的蜡泌物的形态是长丝状,到3龄和成虫期是由湿蜡凝结成不同大小的颗粒,块状和片状。雄虫蜡壳是薄蜡片构成,表面呈鱼鳞状凹凸。低龄期若虫蜡泌物的红外光谱特征与雌成虫的十分相似,说明二者所含的主要化学成分是相同的,雄虫和低龄若虫及雌成虫蜡壳萃取物的红外光谱的吸收峰型基本一致,但在1240.7cm-1处出现一个中等强度的特征峰,这充分说明3种蜡泌物的有机化合物种类和结构一致,只是雄虫分泌物与另二者比较具有特征官能团,从而,利用该吸收峰可以区别出雄虫。此外,根据红外光谱特征分析,可以知道该虫蜡泌物的化学成分主要为长链脂肪酸、脂肪醇和一些带芳香环的化合物。
植物挥发物对天敌昆虫异色瓢虫的引诱效应

Abstract: 采用嗅觉仪测定和比较了国槐、柳树、杨树、臭椿、榆树、白丁香、红花刺槐、蕃茄、枣树、云杉10种植物枝叶挥发物对蚧虫和蚜虫的重要天敌――异色瓢虫的引诱效应.结果表明,在所试验的10种植物中,国槐枝叶对异色瓢虫的引诱作用最强,其平均趋向率达60%.通过用热脱附气相色谱/质谱联机分析(tct-gc/ms)方法,从国槐枝叶的挥发物中检测出38种组分,烯类化合物含量最多,占63.3%(其中萜类化合物6种);烷烃类化合物占16.2%;其它为萘及其衍生物、苯及其衍生物、酯类、醛类和醇类等.从检出的国槐挥发物组分中选取了6种物质引诱瓢虫,发现α-蒎烯在10-4~10-6g/ml、沉香醇10-4g/ml和已醛10-5g/ml的浓度下对瓢虫有显著的引诱作用.图1表3参14

Abstract: Scale insects (Hemiptera: Coccoidea) compose a group of important plant pests of agricultural crops, forest plants, ornamental plants and fruit trees. The history of the study of entomopathogenic fungi as a biological insecticide is reviewed according to the three developmental stages: the pioneering stage, the slow development stage, and the prosperity stage. Additionally, the status of this field in China was discussed. A list including approximately 140 species within 55 genera of the recorded fungal pathogens of scale insects in the world was provided. Finally, we provide four suggestions for the development of entomopathogenic fungi in the future.
优化流程规范连台手术预防性抗菌药物的用药时机

Abstract: 目的优化抗菌药物使用流程,保证连台手术术前预防性抗菌药物的合理使用。 方法2015年12月优化某院抗菌药物使用流程,选择2015 年 5—11月6 072例连台手术作为对照组,2015年12月—2016年5月5 832例连台手术作为实验组,比较两组抗菌药物的规范使用合格率,分析延迟/提前使用原因。结果抗菌药物使用流程优化前后抗菌药物使用率分别为77.16%、78.80%,两者比较,差异有统计学意义(χ2=8.305,P=0.004)。抗菌药物使用流程优化后,抗菌药物0.5~1 h内使用率(82.36%)较优化前(41.11%)明显上升;距切皮时间<0.5 h 抗菌药物使用率由优化前的57.11%下降至4.32%;但距切皮时间>1 h的抗菌药物使用率由1.78%增加至13.32%。连台手术抗菌药物使用延误/提前原因主要包括医护缺乏有效沟通导致巡回护士对连台手术间隔时间的评估欠准确(62.13%),麻醉医生插管或穿刺时间过长(13.57%)。结论优化流程,可以提高连台手术预防性抗菌药物0.5~1 h 内使用率,有助于保障预防性应用抗菌药物的效果
