Abstract: 综述了icp-ms法应用于蛋白质定量技术方面的研究进展.蛋白质定量研究已成为蛋白质组学研究领域的热点,它是解析生物体蛋白质功能的重要途径.基于同位素标记和生物质谱分析技术是蛋白质定量最常用的方法之一,近年来,随着质谱技术的发展,电感耦合等离子体质谱(icp-ms)技术成为元素测量的重要手段,这使其在蛋白质定量中具一定的应用前景.

Abstract: Automated multidimensional liquid chromatography-tandem mass spectrometry (LC-MS/MS) is routinely applied in large scale proteome profiling. However global proteome analysis remains a technical challenge due to the issues associated with sample complexity by tryptic digestion. The application of tag containing peptide enrichment approach for sample pre-separation could reduce the complexity of protein digest. Here, we demonstrated a simple and highly efficient cysteine-containing peptide enrichment method using a thiol specific covalent resin. The cysteine-containing peptides from the tryptic digests of the complex protein mixtures were selected by covalent chromatography based on thiol-disulfide exchange, identified by mass spectrometry. The strategy was firstly optimized and evaluated by using the tryptic peptides of bovine serum albumin (BSA). Then the method was applied with a relatively complicated sample from a five standard protein mixture. The results of these studies show that the enrichment method of cysteine-containing peptides is highly specific, efficient and reproducible. The effectiveness of this method in reducing the sample complexity and improving the identification of peptides by mass spectrometry has enabled high-throughput, automatic and large-scale qualitative and quantitative proteome analyses.
螯合稀土金属标记蛋白质技术的建立与优化

Abstract: 建立了一种基于螯合稀土金属标记蛋白质的技术,优化了影响标记效率的各种条件。在50mmol/LTEAB缓冲体系(pH7.5),DTPA与肽段摩尔比为100:1,常温反应30min的条件下,DTPA标记完全;在100mmol/L乙酸铵缓冲液(pH6.0),稀土金属与DTPA摩尔比为4:1,37℃反应1h的条件下,稀土金属螯合反应完全。考察了不同稀土金属的螯合效率,发现离子半径最小的Lu螯合效率最高。本方法被成功应用于肽混合物标记。
The Performance Analysis of Chaotic Frequency-Hopping Sequences

Abstract: The performance analysis of chaotic Frequency-Hopping(FH)sequences,which are generated by quantization function and reshaping operation based on chaotic map,is presented in this paper.Theory analysis and performance experimental at results show that this sequence is Bernoulli sequence and its Hamming correlation is shown to be Poisson distributed.It is comparable to other FH sequences on the properties of uniform distribution,Hamming correlation and linear complexity when they have the same number of frequency slots and the same period,but much less requirements of iterative operation.It can be concluded that more FH sequences can be generated by this method and they are suitable for FH code-division multiple-access systems.

Abstract: A new encryption technique for color image information is proposed based on melting encryption in light of color science.Linear and nonlinear transformation of quantity are applied to the two color images in the RGB and XYZ color space,and these two color images are individually the origin color image used for transporting or storing and the color image used for encryption or decryption.Then color image information is melted to encryption map,which can't be identified and decryption easily.So the texts and images can be transported secretly and securely.

Abstract: 氮肥投入是保证世界粮食总产量不断增加的重要因素.如何在高投入集约化生产条件下,提高氮肥利用效率、减少氮肥损失及其带来的环境问题,是当前作物生产中面临的重要课题.高产高投入玉米生产体系中,硝酸盐淋失是氮肥损失的重要途径之一.本文论述了土壤硝态氮运移特点、玉米吸氮规律及土壤氮素有效性对根系生长的调节作用,提出了玉米氮高效理想根系构型.通过改良根系构型、增加深层土壤中根系分布,有可能减少氮素向深层的淋失,从而提高氮肥利用率,同步实现玉米高产与氮高效利用.
应用超高分辨双向凝胶电泳技术进行正常人类肝脏蛋白质组研究

Abstract: 肝脏是人体最大的实质器官,承担着人体许多关键的生理功能,在生命活动中占有重要地位.根据人类肝脏蛋白质组计划,本实验旨在构建人肝脏蛋白质双向凝胶电泳表达谱,尽可能分离和鉴定更多的蛋白.在双向凝胶电泳第一向等电聚焦水平上,从上样方式、水化液配方、聚焦时间等方面优化了碱性蛋白的分离条件,利用超放大胶分离技术搭建了高分辨的双向凝胶电泳分离平台,构建了人类肝脏蛋白质2-DE参考谱,检测到5481个蛋白质点.这是目前国际上最为全面的人体器官蛋白质组2-DE参考谱,为其他肝病的研究提供了较好的参照系.成功鉴定了429个非冗余蛋白,对pH4.0~7.0,5.0~6.0,5.5~6.7,6.0~9.0部分蛋白质点在胶上进行了注释,由此构建了人肝2-DE蛋白质表达谱数据库.对蛋白质的理化性质、功能及亚细胞定位进行了全面分析.研究中构建的人类肝脏蛋白质2-DE图谱将为肝脏比较蛋白质组学研究提供参考.

Abstract: 氮素吸收代谢是所有生物生命活动的核心部分,因此,各种生物对氮的吸收与同化过程都有精细的调节。这种调节依赖于生物对体内氮素状况信号及体外(生长介质中)氮素信号的感受。过去几十年中,在细菌、真菌中已经对氮素营养信号有了较好的研究,而植物中则相对缓慢,但也有了一定的认识。本文重点比较细菌、真菌及植物中氮信号系统的组成,以期为进一步认识植物中的氮信号接受与转导系统提供启示。
