Abstract: The influence of the difficulty of the target problem namely the related isomorphic problems which have some surfase similarity, and the awareness of the related problems of solvers had been examined by experimental research. Results suggested that during the process of geometry problem solving, the level of awareness of related problems is one of the major factors that influence the effect of problem-solving transfer. Either more difficult or easier the transfer problem will hinder the influence of awareness of related problems during problem-solving transfer, even the achievements of the problem solvers will show floor effect or ceiling effect, and the degree of difficulty of the transfer problems have interctions with the level of awareness of related problems in affecting problem-solving transfer.

Abstract: 利用1960—2004年新疆北部36个气象站月降水资料和美国国家气候数据中心重建的1960—2004年全球2°×2°月平均海洋表面温度资料,研究了新疆北部夏季降水与海温异常的关系。研究表明,新疆北部夏季降水变化与ENSO事件关系不密切,而与前期冬末—春季的5个海温敏感区密切联系,这5个海温敏感区分别为北印度洋、西太平洋暖池及黑潮区、热带中东太平洋、北大西洋和热带大西洋。前期春季海温异常与新疆北部夏季降水变化联系最显著,表现为显著正相关关系。研究为预测新疆北部夏季降水提供了有益的因子。

Abstract: To investigate the effect of Kozak sequence (+4A or +4G) on expression of green fluorescent protein (GFP) gene in HEK293 cells. The eukaryotic expression vectors containing GFP gene with different Kozak sequence (+4A or +4G) were constructed by classic DNA recombination methods, including PCR, enzyme digestion, ligation, transformation, identification, et al. Two different Kozak sequences (+4A or +4G) were obtained through PCR with different mutagenic primers. The right recombinant plasmids pHGFP-A and pHGFP-G were transfected into HEK293 cells by liposome-mediated gene transfer method. The expression level of GFP was observed by fluorescent microscope, flow cytometry and Western blot. The flow cytometry revealed that the expression levels of GFP fluorescence in pHGFP-A and pHGFP-G transfected cells were about 15% and 45%, respectively. Western blot showed the specific bands of about 27 kD (GFP) both in pHGFP-G and pHGFP-A sample lanes; and the GFP expression density of pHGFP-G was about 3.87-fold as that of pHGFP-A by ImageJ software analysis. These results indicated that the +4G in Kozak sequence (when ?3 site is purine base pair) plays an important role in GFP protein translation, which enhances the GFP expression up to 4-fold in HEK293 cells.

Abstract: The collection efficiency and the emission rate of methane in a municipal solid waste landfill in Guangzhou were studied for improving the collection efficiency and reducing the emission of methane. The theoretical methane emission of 7.22×107 m3 were estimated by the US-EPA first order kinetics model, which corresponded to (2.17~2.89)×107 m3 of actual methane emission in 2010. In practice, 1.75×107 m3 of methane was collected, with the collection efficiency ranging from 60.6% to 80.6%. The operating area was the main emission source of methane, and the emission rate of methane was 0.175×107 m3·a-1 which accounted for 10.0 percent of total methane collected. The emission rates of methane in matured and fresh refuses were 4.17 mol·m-2·h-1 and 0.29 mol·m-2·h-1, respectively.
