A PRELIMINARY STUDY ON FERMENTATION OF CANAVALIA ENSIFORMIS SEEDS

Abstract: Pada umumnya di Indonesia kacang kedele difermentasi menjadi tempe kedele. Hasil fermentasi ini merupakan sumber protein dan vitamin yang penting dalam makanan penduduk sehari-hari. Di daerah-daerah tertentu di pulau Jawa beberapa jenis kacang-kacangan seperti Koro benguk (Mucuna pruriens), koro pedang (Canavalia ensiformis) dan kacang hiris (Cajanus cajan) juga difermentasi menjadi semacam tempe kedele. Dalam penelitian ini telah dilakukan fermentasi biji C. ensiformis menjadi tempe. Sejumlah kapang Rhizopus telah diperiksa kemampuannya untuk memfermentasikan biji-biji tersebut. R.oryzae R128 mampu menghasilkan tempe koro pedang dalam waktu 26 jam inkubasi pada suhu 30°C. Telah diteliti beberapa perubahan biokimia yang terjadi dalam substrat. Selama proses fermentasi berlangsung terjadi peningkatan dari kadar senyawa asam total, nitrogen amino dan karbohidrat terlarut. Analisa zat gizi dalam biji C. ensiformis mentah dan sesudah difermentasi menjadi tempe telah dilaku-kan.
Characteristics Of Phytases From Soybeans And Microorganisms Involved In The Tempe Production

Abstract: Tempe was prepared from Forest variety of soybeans inoculated with pure culture of the mould Rhizopus oligosporus strain CT11K2. Partially purified phytases from soybeans, mould of the R. oligosporus strain CT11K2, yeasts (Endomycopsis burtonii, Candida diddensiae and Candida tropicalis) and bacteria (Streptococcus faecium and Streptococcus dysgalactiae) involved during tempe production were prepared according to procedure developed by Sutardi (1988). The crude phytases were characterized and phytic acid, inorganic and total phosphorous content of soybeans, intermediate products and tempe, were also determined.Results show that phytases, produced by microorganisms involved in tempe production, and by endogenous soybean phytase, had significantly different characteristics especially on optimum pH, temperature and substrate, Km and Vmax values, inactivation and activation energy and also temperature coefficient. All phytases contribute to the reduction in phytic acid content; and mould phytase, especially the extra-cellular enzyme, and the endogenous soybean phytase, showed the dominant effect on phytic acid degradation compared with other microbial phytases and physical treatments such as soaking, boiling and steaming.Phytic acid content of soybeans was reduced by more than 50% from about 10.4 to about 4.9 - 5.0 mg/g (DWB) in fresh tempe (after 40 h fermentation at 30oC)
Role of Bacteria in Tempe Bitter Taste Formation: Microbiological and Molecular Biological Analysis Based on 16S rRNA Gene

Abstract: Tempe is traditional Indonesian food. It has a variety of tastes, sometimes with a hint of bitterness, which may differ in intensity. The cause of bitterness in tempe has never been reported previously. In this study, the aim is to identify whether bacteria play a role in the formation of bitter tastes in tempe. Sensory tests were carried out in order to determine the scoresof bitter-taste-intensity in tempe. The sensory test on EMP, WJB, CLR, DRG, and MLB tempe shows that EMP tempe has the highest score (2.3) and WJB has the lowest (1.3). It is revealed that the processing method has no impact on the formation of the bitter taste in tempe. Plating analysis, showed that EMP soaking water contained a higher number of Enterobacteria group bacteria, approximately 103-104 CFU ml-1 and spore-forming bacteria groups, 102 CFU ml-1, compared to WJB. Similarly, other bacteria groups in fresh EMP tempe was 102 CFU g-1 higher than those in fresh WJB tempe. Based on sequencing the16S rRNA gene, the dominant bacteria on PCA media in EMP tempe are Acetobacter indonesiensis, Klebsiella pneumoniae, Bacillus subtilis, and Flavobacterium sp. On the other hand those in WJB tempe were Klebsiella sp., Brevundimonas sp., Bacillus sp., Pseudomonas putida, and Acinetobacter sp. Bacillus, a group of proteolytic bacteria was found 105 CFU m-1 higher in the soaking water of EMP compared to WJB. Nevertheless, the types and numbers of fungi were not significantly different between tempe types. Accordingly, it is concluded that the difference in the number and the types of bacteria involved in the tempe production process leads to the difference in the bitter taste intensity in both EMP and WJB tempe.
Comparison of DNA Extraction Methods for Microbial Community Analysis in Indonesian Tempe Employing Amplified Ribosomal Intergenic Spacer Analysis

Abstract: Tempe fermentation involved complex microbial communities which are only revealed partially through culture dependent methods. Culture-independent methods would be potential to unravel this complex microbial fermentation. Appropriate DNA extraction is an essential tool to obtain reliable data from culture independent method. In this study, we employed two commercial DNA extraction methods to find the best one for microbial community characterization employing amplified ribosomal intergenic spacer analysis (ARISA). Our result showed that PowerFood Microbial DNA Isolation Kit-MOBIO (PFMDIK) is an excellent method for microbial DNA extraction from tempe. It gave high quantity and quality of DNA suitable for PCR amplification of 16S-23S rRNA intergenic spacer to yield a diverse and reproducible ARISA profile.
Influence of Using Coconut, Palm, and Corn Oils as Frying Medium on Concentration of Acrylamide in Fried Tempe

Abstract: The aim of this research is to study the formation of acrylamide with different kind of vegetable oils as the cooking media. The samples were prepared by cooking and baking above 120℃ then extracted with dichloromethane-ethanol and separated by SPE (C-18) with methanol 60% as eluent. The extracts were analysed by HPLC, with condition as followed: C-18 column; acetonitrile-water (5:95) pH 2.52 mobile-phase; 0.5 ml/minute flow rate; and 210 nm wavelength. It was figured out that a fried tempe using corn oil contained 0.5778 μg/g acrylamide (8.202.10-3 standard deviation and 1.4195% coefficient variation), using coconut oil 0.192 μg/g acrylamide (5.656.10-3 standard deviation 2.946% coefficient variation), using palm oil 0.1455 μg/g acrylamide (6.081.10-3 standard deviation and 4.1794% coefficient variation).

Abstract: ?测量了某重塑粉质粘土三轴击实试样在脱水条件下的土-水特征曲线,并对其特点进行了分析比较。三轴击实试样下部的土-水特征曲线(swcc)和上部的swcc都具有一般swcc的典型特点,但前者较后者明显右移;结合层的swcc有一定幅度下移;受三轴击实试样制作过程影响,较密实的下部区段具有较高的进气值和较高的残余含水量,而且,沿试样高度不同,持有不同残余含水量的各区段对应的基质吸力基本相同。

Abstract: Using the data of monthly mean temperature of a year, the annual temperature curve was simulated by means of the spline interpolation. On the basis of the simulated curve, the beginning/last date of different boundary temperature and the cumulative temperature can be calculated. Spline-function is the smooth connection of continuous segments of a polynomial. The quadratic spline-function can be expressed as following: Based on the interpolation condition, connecting condition and boundary condition, the undetermind coefficient a, b,c can be calculated. The recurrence formulas are: On this basis, the annual temperature curve can be simulated, then the date of boundary temperature can be calculated.
THE PRICE OF FANATICISM: THE CASUALTIES OF THE BULHOEK MASSACRE

Abstract: The Bulhoek massacre is a well-known event in popular memory of many black South Africans and a standard feature in accounts of South African history. Despite this fact historians have not yet established the exact number of Israelites who were killed as a result of their attack on the Police on 24 May 1921. It is the purpose of this article to establish a reliable number of Israelites who were killed during the Bulhoek massacre. The Union Defence medical personnel that accompanied the Police force to Bulhoek consisted of Major M. Welsh (in charge), Sergeant Major W. Richardson, Sergeant S. Allan, Sergeant Wallace, Private Kriel and Driver Valentine with one motor ambulance and field equipment. En route from Pretoria to Queenstown, the team was joined by Private Sutton from Tempe hospital in Bloemfontein. On their arrival at Queenstown the medical personnel accompanied Colonel Truter to an interview with the senior Magistrate of Queenstown E.C.A. Welsh. The latter stressed the seriousness of the situation and warned that the Police would not be able to carry out the Government's instructions without bloodshed. The medical personnel became convinced that bloodshed was unavoidable. They decided to ascertain the exact number of available accommodation at the Frontier Hospital in Queenstown. They found that only about 25 Europeans and up to 40 Africans could be accommodated. After checking available accommodation at Frontier Hospital, they erected a tent hospital at the show grounds of Queenstown.
