Distribución y diversidad genética de Begomovirus que infectan tomate (Solanum lycopersicum L) en Colombia

Abstract: Título en ingles: Distribution and genetic diversity of tomato –infecting Begomovirus in Colombia Resumen: Las enfermedades causadas por los begomovirus, (Familia Geminiviridae) constituyen una serie limitante para la producción del tomate en Colombia. Sin embargo, la caracterización de estos virus no ha sido realizada al momento. Aquí presentamos los resultados de un muestreo a nivel nacional que buscaba conocer la distribución y diversidad genética de los geminivirus que están afectando el cultivo de tomate en Colombia. Los virus fueron detectados mediante PCR, empleando primer universales específicos para el género Begomovirus. Los fragmentos amplificados por PCR fueron sometidos a un análisis tipo RFLP cuyos resultados evidenciaron presencia de infecciones mixtas e individuales de geminivirus en la mayoría de las muestras recolectadas en campo. Los fragmentos amplificados por PCR fueron clonados y secuenciados. El análisis de secuencia y filogenético mostró que los aislados begomovirales colombianos eran gemininivirus bipartitas típicos del Hemisferio Occidental y que algunos eran variantes de PYMV y otros de ToTEV. Mediante el análisis de los elementos cis-regulatorios (iterones) presentes en el promotor del gen de la proteína asociada a replicación (Rep) de los begomovirus aislados es posible postular eventos de pseudorecombinación que podrían suceder entre ellos durante la ocurrencia de infecciones mixtas en tomate. Abstract: Among the problems affecting tomato production phytopatogens are the most important.Among these, nematodes of the genus Meloidogyne cause large annual economic yield losses in tomatocultivation around the world. The knowledge of the physiological indices of growth is extremely important in theinterpretation of pathogen-host-environment relationships. This way, the effect of growing populations of M.javanica (Treub) on the leaf area expansion rate (LAX) during the early development of cherry tomato accesswas evaluated. The negative effects of nematodes were expressed in early development of tomato plants. Therewas linear increase in population (FP) of M. javanica in all cherry tomato access as it was increased the initiallevel of nematode, resulting in reduction of linear TEAF. Thus any management practice that reduces thenematode population of soil during the growing season of tomatoes, will may favor the growth and developmentof plants and subsequently the production.Entre os problemas que mais afetam a produ o do tomateiro est o os fitopatógenos. Dentreesses, os nematoides do gênero Meloidogyne causam consideráveis perdas econ micas anuais no cultivo detomate em todo o mundo. O conhecimento dos índices fisiológicos de crescimento é extremamente importantena interpreta o das rela es patógeno-hospedeiro-ambiente. Neste contexto, quantificou-se a influência doparasitismo de popula es crescentes de M. javanica (Treub) sobre a taxa expans o da área foliar (TEAF) nafase de desenvolvimento inicial de acessos de tomateiro tipo cereja. Os efeitos negativos do parasitismo dosnematoides se manifestaram na fase inicial de desenvolvimento das plantas. Verificou-se crescimento linear dapopula o final (PF) de M. javanica em todos os acessos de tomateiro cereja avaliados à medida que seaumentou o nível do inóculo inicial do nematoide, ocasionando redu o linear da TEAF. Dessa forma, práticasde manejo que reduzam a popula o de nematoides do solo durante a fase vegetativa dos tomateiros, poder ofavorecer o crescimento e desenvolvimento das plantas e a consequente produ o de frutos.

Abstract: phosphorus, despite its relatively low content in most crops is the most applied nutrient as mineral fertilizer and is an important fraction of the cost of agricultural production. knowledge of the differences among plant varieties in relation to soil and fertilizer p uptake is fundamental for breeding improvement programs and management of phosphate fertilization. differences in p uptake from soil and fertilizer (triple superphosphate), soil p availability through l and a values, and the physiologic, ecophysiologic and agronomic efficiencies of two commercial tomato (lycopersicum esculentum will) varieties, santa clara and santa cruz, were evaluated by the 32p technique. the experiment was carried out in a greenhouse, using two soils, a dystrophic dark red latosol and an eutrophic red yellow latosol, and two p fertilizer application rates (0 and 31.1 mg kg-1). plant responses were markedly different in the two different soils. there was a clear increase in dry matter production, l value, and p uptake from the soil due to fertilizer application, but not in the a values. differences in dry matter production, p concentration and physiologic, ecophysiologic and agronomic efficiency between varieties differed more for the eutrophic red yellow latosol. there was no difference between the two varieties in relation to the uptake of soil and fertilizer p, l and a values.
Correlations between insect resistance and horticultural traits in potatoes

Abstract: The objective of this study was to determine relationships between insect resistance and some horticulturaltraits and the implications on selection. A potato population of 11 clonal families derived from crosses between insect resistantand genotypes adapted to the southern region of Brazil was used. The following traits were evaluated: plant size and vigor,incidence of insect attacks on leaves and tubers, tuber yield and appearance components. Phenotypic and genotypic correlationsbetween leaf and tuber resistance were not significant for the population, while phenotypic correlations between the tworesistance types were significant for individual families, with exception of the family originated from the cross involving ND263-32 . The incidence of insect attacks on tubers was significantly correlated with tuber yield and appearance in familiesderived from crosses of NYL235-4 . The manifold correlations of leaf resistance with horticultural traits for all families weresignificant and unfavorable.

SolDB: A Database of Solanum lycopersicum and Solanum tuberosum Primers

Abstract: SolDB is the Database of Solanaceae Family. It is an interactive, free online specialized database for Solanaceae family. Currently, it spans complete nucleotide sequences of expressed genes of Solanum lycopersicum and Solanum tuberosum along with their annotation. We have designed PCR oligonucleotide primer sequences for each gene, with their features and conditions given. This feature alone greatly facilitates researchers in PCR amplification of genes sequences, especially in cloning experiments. We also provided chloroplast genome section which gives access to fully sequenced plastid genomes and their annotations. Flexible database design, easy expandability, and easy retrieval of information are the main features of SolDB. The Database is publicly available at www.soldb.pakbiz.org.
Utiliza o de fósforo do solo e do fertilizante por tomateiro

Abstract: O fósforo, embora seja o macronutriente em menor concentra o na maioria das culturas, é o mais aplicado como fertilizante mineral, constituindo uma parcela significativa do custo de produ o agrícola. O conhecimento das diferen as entre variedades de plantas em rela o à absor o do fósforo do solo e do fertilizante torna-se de grande importancia para programas de melhoramento e manejo da aduba o fosfatada. Utilizando a técnica do tra ador radioativo 32P, foram avaliadas as diferen as na absor o de fósforo proveniente do solo e do fertilizante (superfosfato triplo), na disponibilidade de P do solo pelos valores L e A e nas eficiências fisiológica, ecofisiológica e agron mica do nutriente pelas variedades comerciais de tomate (Licopersicum esculentum Will) Santa Clara e Santa Cruz. Conduziu-se um experimento em condi es de casa de vegeta o com dois solos (Latossolo Vermelho distrófico e Latossolo Vermelho-Amarelo eutrófico) e dois níveis de P (0 e 31,1 mg kg-1). A resposta das plantas foi diferente nos dois solos e em concordancia com suas características. Houve aumentos significativos, em ambos os solos, na produ o de matéria seca, no valor L e na absor o de P do solo devido à aplica o do fertilizante mineral, mas n o no valor A. Diferen as entre variedades quanto à produ o de matéria seca, concentra o de fósforo, eficiência fisiológica, ecofisiológica e agron mica foram mais marcadas no solo Latossolo Vermelho-Amarelo eutrófico. N o houve diferen as entre as variedades na absor o de P do solo e do fertilizante, valores L e A.
Characterization of Fruits in Contrasting Environments and Germination of Tamarillo Seeds at Different Temperatures  [PDF]

Abstract: Tamarillo (Solanum betaceum Cav. Syn Cyphomandra betacea Cav.) Sendt is a small tree native to the subtropical Andean region of South America from Colombia to Chile, popularly known as arboreal tomato, belonging to the family Solanaceae. The objective of this study was to evaluate the physicochemical characteristics of fruits and the germination of tamarillo (Solanum betaceum) seeds produced in the Caparaó-ES and MG regions. The following procedures were performed: Fruit and peel biometrics; fruit weight; pulp and peel weight; thousand-seed weight, pulp and peel yield and fruit dry mass. In the physicochemical analysis, the following parameters were determined: pH; soluble solids; titratable acidity and ratio. For the analysis of the physiological quality of the seeds, different temperatures (20°C; 25°C; 30°C and 20°C - 30°C) were evaluated in the presence and absence of light and the following was determined: Seed moisture content; germination; germination speed index; germination mean time; shoot length; root and total seedling dry mass. Tamarillo in the region of Caparaó-ES presents greater chemical characteristics of the fruits, germination and vigor, and in the region of Caparaó-MG presents greater physical characteristics of the fruits. Tamarillo can germinate both in the presence and absence of light, although in the presence of light, it presents greater vigor.
Optimizing Extraction of Phenolics and Flavonoids from Solanum ferox Fruit  [PDF]

Abstract: Various phenolic and flavonoid compounds that are secondary plant metabolites are known to contribute to physiological wellbeing. Extraction efficiency of such compounds from plant sources is dependent on the extraction solvent type and composition, and its pH. In this study, different extraction variables were examined: heating time (20 to 180 min), temperature (60°C, 75°C and 90°C) and pH (2.5, 3.0, 4.0, 5.0, 6.0 and 7.0). Hot water was used in the extraction of dry samples. For phenolics, the most efficient extraction was by using water at 60°C for 180 min, whereby 5.95 mg GA equivalent/dry extract was achieved. The most efficient extraction of flavonoids was achieved with water at 60°C for 150 min, whereby 43 μg Quercetin equivalent/dry extract was obtained. Adjusting the solvent to pH 2.5 increased the yield to 45.3 μg Quercetin equivalent/dry extract.
